Development of a Tc-Labeled Single-Domain Antibody for SPECT/CT Assessment of HER2 Expression in Breast Cancer.

Mol Pharm 2021 Jul 30. Epub 2021 Jul 30.

Department of Nuclear Medicine, Shanghai General Hospital, Shanghai Jiao Tong University School of Medicine, No. 100 Haining Road, Shanghai 200080, China.

Accurate determination of human epidermal growth factor receptor 2 (HER2) expression is essential for HER2-targeted therapy in patients with cancer. HER2 expression in a complex environment, such as in a heterogeneous tumor, makes the precise assessment of the HER2 status difficult using current methods. In this study, we developed a novel Tc-labeled anti-HER2 single-domain antibody (Tc-NM-02) as a molecular imaging tracer for the noninvasive detection of HER2 expression and investigated its safety, radiation dosimetry, biodistribution, and tumor-targeting potential in 10 patients with breast cancer. Subcutaneous adipose tissue metabolic function and insulin sensitivity in people with obesity.

Diabetes 2021 Jul 15. Epub 2021 Jul 15.

Division of Geriatrics and Nutritional Science, University School of Medicine, St. Louis, MO

We used stable isotope-labeled glucose and palmitate tracer infusions, a hyperinsulinemic-euglycemic clamp, positron-emission tomography of muscles and adipose tissue after [F]fluorodeoxyglucose and [O]water injections, and subcutaneous adipose tissue (SAT) biopsy to test the hypotheses that: i) increased glucose uptake in SAT is responsible for high insulin-stimulated whole-body glucose uptake in people with obesity who are insulin-sensitive, and ii) putative SAT factors thought to cause insulin resistance are present in people with obesity who are insulin-resistant but not in those who are insulin-sensitive. We found high insulin-stimulated whole-body glucose uptake in insulin-sensitive participants with obesity was not due to channeling of glucose into SAT, but was due to high insulin-stimulated muscle glucose uptake. Furthermore, insulin-stimulated muscle glucose uptake was not different between insulin-sensitive obese and lean participants even though adipocytes were larger, SAT perfusion and oxygenation were lower, and markers of SAT inflammation, fatty acid appearance in plasma in relation to fat-free mass, and plasma fatty acid concentration were higher in the insulin-sensitive obese than lean participants. Limitations and Pitfalls of FDG-PET/CT in Infection and Inflammation.

Semin Nucl Med 2021 Jul 7. Epub 2021 Jul 7.

Department of Nuclear Medicine & PET Center, Aarhus University Hospital, Aarhus N. Electronic address:

White blood cells activated by either a pathogen or as part of a systemic inflammatory disease are characterized by high energy consumption and are therefore taking up the glucose analogue PET tracer FDG avidly. It is therefore not surprising that a steadily growing body of research and clinical reports now supports the use of FDG PET/CT to diagnose a wide range of patients with non-oncological diseases. However, using FDG PET/CT in patients with infectious or inflammatory diseases has some limitations and potential pitfalls that are not necessarily as pronounced in oncology FDG PET/CT. Read More

Assessing the potential and kinetics of coupled nutrients uptake in mesotrophic streams in Chaohu Lake Basin, China.

Environ Sci Pollut Res Int 2021 Jul 3. Epub 2021 Jul 3.

College of Water Conservancy and Hydropower Engineering, Hohai University, Nanjing, China.

Interactions among multiple nutrients uptake certainly have a great effect on their retention in headwater streams, yet little research has been made to explore the quantitative characteristics of their interactions, especially in mesotrophic streams. In response, we conducted an identical series of instantaneous nutrient addition experiments, using ammonium nitrogen (NH-N) and phosphate phosphorus (PO-P) alone or together, in two mesotrophic agricultural headwater streams in Chaohu Lake Basin, China, to quantify the relationships between nutrient concentrations and uptake rates, and examine how NH-N and PO-P interact to affect their individual uptake. Both the Michaelis-Menten (M-M) equation and response surface model were utilized to analyze coupled NH-N and PO-P uptake patterns across a range of nutrient concentrations, by fitting the kinetic processes of NH-N and PO-P uptake in single- and dual-nutrient additions.
