Optimal Method for Reporting Prostate Cancer Grade in MRI-targeted Biopsies.

Am J Surg Pathol 2021 Jun 10. Epub 2021 Jun 10.

Departments of Pathology Urology Population Health, New York University Langone School of Medicine, New York, NY.

When multiple cores are biopsied from a single magnetic resonance imaging (MRI)-targeted lesion, Gleason grade may be assigned for each core separately or for all cores of the lesion in aggregate. Because of the potential for disparate grades, an optimal method for pathology reporting MRI lesion grade awaits validation. We examined our institutional experience on the concordance of biopsy grade with subsequent radical prostatectomy (RP) grade of targeted lesions when grade is determined on individual versus aggregate core basis. Variation in Methyl Jasmonate-Induced Defense Among Norway Spruce Clones and Trade-Offs in Resistance Against a Fungal and an Insect Pest.

Front Plant Sci 2021 24;12:678959. Epub 2021 May 24.

Department of Chemistry, Organic Chemistry, KTH, Royal Institute of Technology, School of Chemical Science and Engineering, Stockholm, Sweden.

An essential component of plant defense is the change that occurs from a constitutive to an induced state following damage or infection. Exogenous application of the plant hormone methyl jasmonate (MeJA) has shown great potential to be used as a defense inducer prior to pest exposure, and could be used as a plant protection measure. Here, we examined (1) the importance of MeJA-mediated induction for Norway spruce () resistance against damage by the pine weevil , which poses a threat to seedling survival, and infection by the spruce bark beetle-associated blue-stain fungus , (2) genotypic variation in MeJA-induced defense (terpene chemistry), and (3) correlations among resistance to each pest. Rupture of a Vulnerable Plaque in a Hazy Angiographic Culprit Lesion During Acute Coronary Syndrome.

J Invasive Cardiol 2021 Jun;33(6):E487-E488

Cardiovascular Department, ASST Santi Paolo e Carlo, Milano, Italy.

In the presence of an equivocal culprit lesion in the context of an acute coronary syndrome, the optical coherence tomography (OCT) scan defines the severity and composition of the plaque, elucidating (as in this pathognomonic case) the mechanism related to plaque rupture. Furthermore, OCT analysis allowed stent implantation with adequate sizing and length, and subsequent final optimization of the result. In the presence of non-unequivocal lesions, in the context of acute coronary syndrome, the use of OCT imaging defined the etiology and mechanism of plaque instability, reducing cardiovascular events. Quantitative analysis of in-vivo microbubble distribution in the human brain.

Sci Rep 2021 Jun 3;11(1):11797. Epub 2021 Jun 3.

Biomedical Engineering, University of Virginia, Charlottesville, VA, USA.

Microbubbles (MB) are widely used as contrast agents to perform contrast-enhanced ultrasound (CEUS) imaging and as acoustic amplifiers of mechanical bioeffects incited by therapeutic-level ultrasound. The distribution of MBs in the brain is not yet fully understood, thereby limiting intra-operative CEUS guidance or MB-based FUS treatments. In this paper we describe a robust platform for quantification of MB distribution in the human brain, allowing to quantitatively discriminate between tumoral and normal brain tissues and we provide new information regarding real-time cerebral MBs distribution. Recognition and Repair of Oxidatively Generated DNA Lesions in Plasmid DNA by a Facilitated Diffusion Mechanism.

Biochem J 2021 Jun 1. Epub 2021 Jun 1.

New York University, NEW YORK, New York, United States.

The oxidatively generated genotoxic spiroiminodihydantoin (Sp) lesions are well-known substrates of base excision repair (BER) pathway initiated by the bifunctional DNA glycosylase NEIL1. In this work we reported that the excision kinetics of the single Sp lesions site-specifically embedded in the covalently closed circular DNA plasmids (contour length 2686 base pairs) by NEIL1 are biphasic under single-turnover conditions ([NEIL1]>[SpDNApl]) in contrast to monophasic excision kinetics of the same lesions embedded in147-mer Sp-modified DNA duplexes. Under conditions of a large excess of plasmid DNA base pairs over NEIL1 molecules, the kinetics of excision of Sp lesions are biphasic in nature, exhibiting an initial burst phase, followed by a slower rate of formation of excision products The burst phase is associated with  NEIL1-DNA plasmid complexes, while the slow kinetic phase is attributed to the dissociation of non-specific NEIL1-DNA complexes. Balloon Deflection Technique to Facilitate Stent Delivery in Impassable Situations During Percutaneous Coronary Intervention.

Cardiol Res 2021 Jun 14;12(3):201-207. Epub 2021 May 14.

Department of Cardiology, LPS Institute of Cardiology, G.S.V.M. Medical College, Kanpur, Uttar Pradesh 208002, India.

Background: Long and multiple lesions especially in the background of tortuous coronary artery often require multiple stents. The entry and passage of another stent through an already deployed stent becomes challenging because of poorly expanded stent struts, calcified tissue, underlying tortuosity, highly angulated takeoff of the vessel, and lack of coaxiality. It can be facilitated by balloon deflection technique (BDT) where balloon at proximal edge of main vessel stent over a buddy wire helps to deflect second stent either distally or into side branch by blocking potential dead.
