The drug of abuse, γ-hydroxybutyric acid (GHB), is commonly co-ingested with ethanol, resulting in a high incidence of toxicity and death. Our laboratory has previously reported that GHB is a substrate for the monocarboxylate transporters (MCT), necessary for its absorption, renal clearance, and tissue distribution, including across the blood-brain barrier. Our goal was to investigate the drug-drug interaction (DDI) between GHB-ethanol and evaluate MCT1 inhibition as a strategy to reverse toxicity. Read More

Effects of advanced maternal age and acute prenatal alcohol exposure on mouse offspring growth and craniofacial phenotype.

Alcohol Clin Exp Res 2021 May 7. Epub 2021 May 7.

Department of Anatomy & Cell Biology, Schulich School of Medicine and Dentistry, The University of Western Ontario.

Background: Prenatal alcohol exposure (PAE) can result in developmental defects including growth restriction, craniofacial anomalies and cognitive-behavioral deficits, though the presence and severity of these adverse outcomes varies dramatically. Preclinical animal models have demonstrated that dose and timing of PAE account for much but not all, of this phenotypic variation, suggesting that additional factors mitigate effects of PAE. Here we used a mouse model to investigate whether maternal age modulates the effects of PAE on the severity and variation of offspring growth and craniofacial outcomes. Read More

Fenofibrate (a PPAR-α Agonist) Administered During Ethanol Withdrawal Reverts Ethanol-Induced Astrogliosis and Restores the Levels of Glutamate Transporter in Ethanol-Administered Adolescent Rats.

Front Pharmacol 2021 20;12:653175. Epub 2021 Apr 20.

Institute of Biomedical Sciences, Faculty of Health Sciences, Universidad Autónoma de Chile, Santiago, Chile.

High-ethanol intake induces a neuroinflammatory response, which has been proposed as responsible for the maintenance of chronic ethanol consumption. Neuroinflammation decreases glutamate transporter (GLT-1) expression, increasing levels of glutamate that trigger dopamine release at the corticolimbic reward areas, driving long-term drinking behavior. The activation of peroxisome proliferator-activated receptor alpha (PPARα) by fibrates inhibits neuroinflammation, in models other than ethanol consumption. Read More

Biphasic Effects of Ethanol Exposure on Waste Metabolites Clearance in the CNS.

Mol Neurobiol 2021 Apr 25. Epub 2021 Apr 25.

Laboratory of Neurovascular Inflammation and Neurodegeneration, Department of Biomedical Engineering, Center for Injury Bio Mechanics, Materials and Medicine, New Jersey Institute of Technology, Newark, NJ, 07102, USA.

We have shown that the effects of low-dose ethanol promote the clearance of waste metabolites, such as amyloid-beta (Aβ) proteins, from the brain through the perivascular space (PVS). We demonstrated that dilative reactivity of arterial smooth muscle and endothelial cells regulate this clearance. These findings indicate the importance of blood-brain barrier (BBB) transvascular clearance of large size metabolites from the central nervous system (CNS), where the lymphatic clearance system is absent. Kinetically Controlled Synthesis of Rhodium Nanocrystals with Different Shapes and a Comparison Study of Their Thermal and Catalytic Properties.

J Am Chem Soc 2021 Apr 14;143(16):6293-6302. Epub 2021 Apr 14.

School of Chemistry and Biochemistry, Georgia Institute of Technology, Atlanta, Georgia 30332, United States.

We report the synthesis of Rh nanocrystals with different shapes by controlling the kinetics involved in the growth of preformed Rh cubic seeds. Specifically, Rh nanocrystals with cubic, cuboctahedral, and octahedral shapes can all be obtained from the same cubic seeds under suitable reduction kinetics for the precursor. The success of such a synthesis also relies on the use of a halide-free precursor to avoid oxidative etching, as well as the involvement of a sufficiently high temperature to remove Br ions from the seeds while ensuring adequate surface diffusion.
