Enzyme-responsive micellar JQ1 induces enhanced BET protein inhibition and immunotherapy of malignant tumors.

Biomater Sci 2021 Sep 15. Epub 2021 Sep 15.

Biomedical Polymers Laboratory, College of Chemistry, Chemical Engineering and Materials Science, and State Key Laboratory of Radiation Medicine and Protection, Soochow University, Suzhou 215123, China.

Bromodomain and extra-terminal (BET) proteins are attractive targets for treating various malignancies including melanoma. The inhibition of BET bromodomains, with JQ1, is found to downregulate the expression of both c-MYC oncoprotein and programmed cell death ligand 1 (PD-L1), which play a crucial role in tumor growth and the immunosuppressive tumor microenvironment, respectively. The BET bromodomain inhibitors like JQ1 though exhibiting high selectivity and affinity show usually low bioavailability and efficacy due to fast clearance and inferior uptake by tumor cells. Stepwise increase NdIII-based phosphorescence by AIE-active sensitizer: Broaden AIPE family from transition metals to discrete NIR lanthanide complexes.

Chemistry 2021 Sep 15. Epub 2021 Sep 15.

Hubei University, College of Chemistry & Chemical Engineering, CHINA.

We designed two near-infrared lanthanide complexes [( L ) 2 -Nd(NO 3 ) 3 ] ( L = TPE 2 -BPY for 1 , TPE-BPY for 2 ) by employing aggregation-induced emission (AIE)-active tetraphenylethylene (TPE) derivatives as sensitizers, which possessed matched energy to Nd III , effectively preventing competitive deactivation under aggregation, even shifting the excitation window toward 600 nm by twisted intramolecular charge transfer. Furthermore, benefiting from the 4 f electron shielding effect and antenna effect, the enhanced excitation energies of the AIE-active sensitizers by structural rigidification transferred into the inert Nd III excited state through 3 LMCT, affording the first aggregation-induced phosphorescence enhancement (AIPE)-active discrete NIR-emitting lanthanide complexes. As 1 equipped with more AIE-active TPE than 2 , L →Nd energy transfer efficiency of the former was larger than that of the latter under the same conditions. Read More

Ortho-Deuteration of Aromatic Aldehydes via a Transient Directing Group-Enabled Pd-Catalyzed Hydrogen Isotope Exchange.

J Org Chem 2021 Sep 13. Epub 2021 Sep 13.

School of Biological and Chemical Engineering, NingboTech University, Ningbo 315100, P. R. China.

A practical and scalable ortho-selective deuteration of aromatic aldehydes was accomplished by Pd-catalyzed hydrogen isotope exchange with deuterium oxide as an inexpensive deuterium source. The use of tert-leucine as a transient directing group facilitates the exchange, affording a wide range of ortho-deuterated aromatic aldehydes with deuterium incorporation up to 97%. The control experiments suggest that the addition of silver trifluoroacetate resists the unexpected reduction of Pd(II), while the theoretical study indicates a rapid reversible concerted metalation-deprotonation process. The ability to identify promising candidate switchable molecules computationally, prior to synthesis, represents a considerable advance in the development of switchable molecular materials. Even more useful would be the possibility of predicting the switching temperature. Cobalt-dioxolene complexes can exhibit thermally induced valence tautomeric switching between low-spin Co-catecholate and high-spin Co-semiquinonate forms, where the half-temperature () is the temperature at which there are equal amounts of the two tautomers. On-Surface Polymerization of In-Plane Highly Ordered Carbon Nitride Nanosheets toward Photocatalytic Mineralization of Mercaptan Gas.

Adv Mater 2021 Sep 3:e2101466. Epub 2021 Sep 3.

State Key Laboratory of Photocatalysis on Energy and Environment, College of Chemistry, Fuzhou University, Fuzhou, 350002, P. R. China.

2D carbon nitride nanosheets have attracted ever-increasing interest in photocatalysis due to their unique structural advantages. However, the nanosheets synthesized by the traditional methods, such as post oxidation and liquid exfoliation, have suffered from in-plane disorder with abundant structural defects, which seriously counteracts their structural benefits for photocatalysis. Herein, it is demonstrated that polymer carbon nitride nanosheets with in-plane highly ordered structure (PCNNs-IHO) can be successfully prepared by on-surface polymerization of melamine on NaCl crystal surface at elevated temperatures. Instituto de Síntesis Química y Catálisis Homogénea, ISQCH (CSIC-Universidad de Zaragoza), Pedro Cerbuna 12, 50009 Zaragoza, Spain.

The irradiation of ()-2-phenyl-4-aryliden-5(4)-thiazolones with blue light (465 nm) in CHCl solution promotes [2 + 2]-photocycloaddition of the exocyclic C═C bonds and the formation of the dispirocyclobutanes . This reaction takes place with high stereoselectivity, given that the ε-isomer (1,3 head-to-tail syn coupling) is formed in more than 90% yield in most of the cases. However, irradiation of 5(4)-thiazolones with blue light (456 nm) in dry MeOH in the presence of BF·OEt leads to the monospirocyclobutanes with full stereoselectivity, also affording the ε-isomer.
