Emerging translational science discoveries, clonal approaches and treatment trends in chronic myeloproliferative neoplasms.

The 60 American Society of Hematology (ASH) held in San Diego in December 2018 was followed by the 13 Post-ASH chronic myeloproliferative neoplasms (MPNs) workshop on the 4 -5 December 2018. This closed annual workshop, first introduced in 2006 by Goldman and Mughal, was organized in collaboration with Alpine Oncology Foundation and allowed experts in preclinical and clinical research in the chronic MPNs to discuss the current scenario, including relevant presentations at ASH, and address pivotal open questions that impact translational research and clinical management. This review is based on the presentations and deliberations at this workshop, and rather than provide a resume of the proceedings, we have selected some of the important translational science and treatment issues which require clarity. Biocatalytic Reversal of Advanced Glycation End Product Modification.

Advanced glycation end products (AGEs) are a heterogeneous group of molecules that emerge from the condensation of sugars and proteins via the Maillard reaction. Despite a significant number of studies showing strong associations between AGEs and the pathologies of aging-related illnesses, it has been a challenge to establish AGEs as causal agents primarily due to the lack of tools in reversing AGE modifications at the molecular level. Here, we show that MnmC, an enzyme involved in a bacterial tRNA-modification pathway, is capable of reversing the AGEs carboxyethyl-lysine (CEL) and carboxymethyl-lysine (CML) back to their native lysine structure. Performance of revised STO(1M)-3G basis set for prediction of 5-fluorocytosine chemical shift.

Nuclear shieldings and chemical shifts of 5-fluorocytosine (5FC) were predicted in the gas phase and DMSO solution modeled by polarizable continuum model (PCM) using B3LYP density functional and revised STO(1M)-3G basis set. For comparison, seven arbitrary selected basis sets including STO-3G, medium-size Pople- and larger dedicated Jensen-type ones were applied. The former basis sets were significantly smaller but the calculated structural parameters, harmonic vibrational frequencies were very accurate and close to those, obtained with larger, polarization-consistent ones. Habitat edge responses of generalist predators are predicted by prey and structural resources.

Generalist predators are thought to be less vulnerable to habitat fragmentation because they use diverse resources across larger spatial scales than specialist predators. Thus, it has been suggested that generalist predators may respond positively to habitat edges or demonstrate no edge response, because they can potentially use prey resources equally well on both sides of the habitat edge.
