Diuretic Renal Scintigraphy in Patients with Sulfonamide Allergies: Possible Alternative Use of Ethacrynic Acid.

Authors:
Ba D Nguyen Michael C Roarke Jason Robert Young Ming Yang Howard H Osborn

J Nucl Med Technol 2015 Dec 15;43(4):239-41. Epub 2015 Oct 15.

Department of Radiology, Mayo Clinic, Scottsdale, Arizona; and.

Unlabelled: Furosemide may trigger life-threatening sulfonamide cross-hypersensitivity reactions, posing a dilemma in patients who need diuretic renal scintigraphy.

Methods: We present our experience using ethacrynic acid as a nonsulfonamide alternative diuretic in 5 patients, with a discussion of the diuretic molecular structure, potential side effects, protocol development, and imaging results.

Results: Diuretic renal scintigraphy using ethacrynic acid provided useful information about the obstruction status in all patients, with no adverse clinical impact.

Conclusion: Ethacrynic acid is a potential alternative to furosemide for patients with severe sulfonamide reactions.

The linking of ethacrynic acid with ethylenediamine and 1,4-butanediamine gave EDEA and BDEA, respectively, as membrane-permeable divalent pro-inhibitors of glutathione -transferase (GST). Their divalent glutathione conjugates showed subnanomolar inhibition and divalence-binding to GSTmu (GSTM) (PDB: 5HWL) at ∼0.35 min. Read More

Novel insights into conjugation of antitumor-active unsymmetrical bisacridine C-2028 with glutathione: Characteristics of non-enzymatic and glutathione S-transferase-mediated reactions.

Authors:
Agnieszka Potęga Michał Kosno Zofia Mazerska

J Pharm Anal 2021 Dec 5;11(6):791-798. Epub 2021 Apr 5.

Department of Pharmaceutical Technology and Biochemistry, Faculty of Chemistry, Gdańsk University of Technology, Gdańsk, 80-233, Poland.

Unsymmetrical bisacridines (UAs) are a novel potent class of antitumor-active therapeutics. A significant route of phase II drug metabolism is conjugation with glutathione (GSH), which can be non-enzymatic and/or catalyzed by GSH-dependent enzymes. The aim of this work was to investigate the GSH-mediated metabolic pathway of a representative UA, C-2028. Read More

Loop Diuretics Inhibit Ischemia-Induced Intracellular Ca Overload in Neurons the Inhibition of Voltage-Gated Ca and Na Channels.

Authors:
Christopher Katnik Javier Cuevas

Front Pharmacol 2021 15;12:732922. Epub 2021 Sep 15.

Department of Molecular Pharmacology and Physiology, Morsani College of Medicine, University of South Florida, Tampa, FL, United States.

One consequence of ischemic stroke is disruption of intracellular ionic homeostasis. Intracellular overload of both Na and Ca has been linked to neuronal death in this pathophysiological state. The etiology of ionic imbalances resulting from stroke-induced ischemia and acidosis includes the dysregulation of multiple plasma membrane transport proteins, such as increased activity of sodium-potassium-chloride cotransporter-1 (NKCC-1).
