NMDA receptor membrane dynamics tunes the firing pattern of midbrain dopaminergic neurons.

Authors:

J Physiol 2021 Mar 2. Epub 2021 Mar 2.

Univ. de Bordeaux, Interdisciplinary Institute for Neuroscience, UMR 5297, Bordeaux, F-33000, France.

Key Points: NMDA receptors expressed by dopamine neurons of the ventral tegmental area (VTA) play a central role in glutamate synapse plasticity, neuronal firing and adaptative behaviours. The NMDAR surface dynamics shape synaptic adaptation in hippocampal networks, as well as associative memory. We investigated the basic properties and role of the NMDAR surface dynamics onto cultured mesencephalic and ventral tegmental area dopamine neurons in rodents. Read More

Inhibition of miR-200b Promotes Angiogenesis in Endothelial Cells by Activating The Notch Pathway.

Authors:
Tie-Ying Qiu Jin Huang Li-Ping Wang Bi-Song Zhu

Cell J 2021 Apr 1;23(1):51-60. Epub 2021 Mar 1.

Organ Transplant Center, Xiangya Hospital, Central South University, Changsha 410008, P.R. China.

Objective: Patients with diabetes mellitus frequently have chronic wounds or diabetic ulcers as a result of impaired wound healing, which may lead to limb amputation. Human umbilical vein endothelial cell (HUVEC) dysfunction also delays wound healing. Here, we investigated the mechanism of miR-200b in HUVECs under high glucose conditions and the potential of miR-200b as a therapeutic target. Read More

Objective: Sexual dimorphism in mammals can be described as subsequent transcriptional differences from their distinct sex chromosome complements. Following X inactivation in females, the Y chromosome is the major genetic difference between sexes. In this study, we used a male embryonic stem cell line (Royan H6) to identify the potential role of the male-specific region of the Y chromosome (MSY) during spontaneous differentiation into embryoid bodies (EBs) as a model of early embryonic development. FoxA2 and RNA Pol II Mediate Human Islet Amyloid Polypeptide Turnover in ER-stressed Pancreatic β-cells.

Authors:
Diti Chatterjee Bhowmick Lydia Burnett Zhanar Kudaibergenova Aleksandar Jeremic

Biochem J 2021 Mar 2. Epub 2021 Mar 2.

The George Washington University, Washington, District of Columbia, United States.

Here, we investigated transcriptional and trafficking mechanisms of human islet amyloid polypeptide (hIAPP) in normal and stressed β-cells. In high glucose-challenged human islets and rat insulinoma cells overexpressing hIAPP, cell fractionation studies revealed increased accumulation of hIAPP. Unexpectedly, a significant fraction (up to 22%) of hIAPP was found in the nuclear soluble and chromatin-enriched fractions of cultured human islet and rat insulinoma cells. Read More

Functional interaction of ubiquitin ligase RNF167 with UBE2D1 and UBE2N promotes ubiquitination of AMPA receptor.

Authors:
Kim Ghilarducci Valérie C Cabana Camille Desroches Kahina Chabi Steve Bourgault Laurent Cappadocia Marc P Lussier

FEBS J 2021 Mar 2. Epub 2021 Mar 2.

Department of chemistry, Université du Québec à Montréal, Montreal, QC, Canada.

Protein ubiquitination has been historically associated with protein degradation, but recent studies have demonstrated other cellular functions associated with substrate ubiquitination. Amongst the RING type ubiquitin E3 ligase enzymes present in the human genome, RNF167 is a transmembrane protein located in endosomes and lysosomes and is implicated in controlling the endolysosomal pathway. Substrates of RNF167 have been identified, but the ubiquitin conjugating E2 enzymes involved in the mechanism remain unknown. Read More
