Flexible construct of N vacancies and hydrophobic sites on g-CN by F doping and their contribution to PFOA degradation in photocatalytic ozonation.

N vacancies, hydrophobic sites and electron rich zone were simply regulated by doping F into g-CN (CN) to accelerate photocatalytic ozonation of PFOA. Activity of F-CN was superior to that of CN, with 74.3% PFOA removal by F-CN/Vis/O but only 57.1% by CN/Vis/O. Experimental results and theory simulations suggested that the photogenerated hole (h) oxidation with the help of N vacancies was vital for PFOA degradation. N vacancies on both CN and F-CN would trap O atom of PFOA and seize electron from α -CF group, which made PFOA more easily to be oxidized. Doping of F narrowed band gap, lowered the valence band position and enhanced the oxidation potential of h. The hydrophobic sites would accelerate the mass transfer of O and PFOA, enhance O's single electron reduction with e to generate hydroxyl radicals (•OH) and reduce the recombination of h-e. Under the joint function of h, N vacancies and •OH, PFOA degradation in F-CN/Vis/O proceeded through the gradually shortening of perfluoroalky chain and loss of CF unit. The acute and chronic toxicity of generated short-chain perfluorocarboxylic acid toward fish, green algae daphnid were predicted by ECOSAR. And the toxicity change of solutions was examined by luminescent bacteria.

Self-assembly of core-shell structured multiwalled [email protected] organic frameworks composite for solid-phase extraction of four phytohormones from fruit juices.

Covalent organic frameworks (COFs) have attracted considerable attention in sample pretreatment because of their unique characteristics. However, the submicron or micron size of COFs has restricted their wider applications in solid-phase extraction (SPE). Herein, multiwalled nanotubes (MWNTs) were used as substrate materials to synthesize core-shell structured [email protected] composites ([email protected]) using a simple self-assembly method. The as-prepared [email protected] composite exhibited a high BET surface area, good thermal stability, and good adsorption capacity. The [email protected] composite was used as an adsorbent in cartridge-based SPE to extract four phytohormones before determining their levels by high-performance liquid chromatography. The experimental parameters affecting extraction efficiency, including the amount of adsorbents, solution pH, ionic strength, eluent type, and eluent volume, were investigated. The developed method showed a wide linear range (0.37-100 ng mL), low detection limits (0.11-0.32 ng mL), low limits of quantification (0.37-1.07 ng mL), high enrichment factors (45.9-49.3), and good reproducibility (<4.8%) for phytohormones. The developed analytical method was used to analyze trace phytohormones in fruit juices with good recoveries, highlighting the potential of the [email protected] composite as an adsorbent in sample preparation.

Dupilumab treatment reduces hospitalizations in adults with moderate-to-severe atopic dermatitis.

Background: Refractory disease, flares, or infections in atopic dermatitis (AD) can lead to hospitalizations.

Objective: To compare hospitalization rates among adults with moderate-to-severe AD treated with dupilumab versus control.

Methods: Data from 7 randomized, placebo-controlled trials of dupilumab (300 mg every 2 weeks [q2w] and/or weekly [qw]; with or without topical corticosteroids) were analyzed.

Results: Patients in the dupilumab 300 mg q2w, qw, and combined dupilumab (q2w and qw, n = 1841) groups compared with patients in the control group (n = 1091) had lower rates of all-cause hospitalizations (5.8, 2.7, and 3.8 events, respectively, vs 9.0 events per 100 patient-years [PY]; all P < .05 [49%, 71%, and 62% risk reduction, respectively]); AD-related hospitalizations (2.0, 0.4, 1.0 events vs 4.1 events per 100 PY; P < .05 for qw and dupilumab combined [91% and 79% risk reduction, respectively]); as well as reduced overall duration of AD-related hospitalization (10.9, 7.3, and 8.6 days vs 38.9 days per 100 PY).

Conclusions: Among adults with moderate-to-severe AD, treatment with dupilumab versus control was associated with significant reductions in all-cause and AD-related hospitalization rates, and shorter duration of AD-related hospitalization.

Aging is a slow and progressive natural process that compromises the normal functions of cells, tissues, organs and systems. The aging of the hypothalamic median eminence (ME), a structural gate linking neural and endocrine systems, may impair hormone release, energy homeostasis and central sensing of circulating molecules, leading to systemic and reproductive aging. However, the molecular and cellular features of ME aging remain largely unknown. Here we describe the transcriptional landscape of young and middle-aged mouse ME at single-cell resolution, revealing the common and cell-type-specific transcriptional changes with age. The transcriptional changes in cell-intrinsic programs, cell-cell crosstalk and cell-extrinsic factors highlight five molecular features of ME aging and also implicate several potentially druggable targets at cellular, signaling and molecular levels. Importantly, our results suggest that vascular and leptomeningeal cells (VLMCs) may lead the asynchronized aging process among diverse cell types and drive local inflammation and cellular senescence via a unique secretome. Together, our study uncovers how intrinsic and extrinsic features of each cell type in the hypothalamic ME are changed by the aging process, which will facilitate our understanding of brain aging and provide clues for efficient anti-aging intervention at the middle-aged stage.

Mesenchymal stem cells (MSCs) are promising in idiopathic pulmonary fibrosis (IPF) therapy. However, low survival rate and ambiguous behavior of MSCs after transplantation impede their clinical translation. To this end, we have developed a new strategy to improve the survival rate and monitor the behavior of the transplanted MSCs simultaneously. In our strategy, nintedanib, a tyrosine kinase inhibitor, is employed to protect the human MSCs (hMSCs) from excessive oxidative stress responses and inflammatory environment in the damaged lung. Moreover, by labeling of the transplanted hMSCs with a computed tomography (CT) nanotracer, Au nanoparticles functionalized with polyethylenimine (PEI) and polyethylene glycol (PEG) ([email protected]@PEG), in combination with red-emitting firefly luciferase (RfLuc), in vivo CT/bioluminescence (BL) dual-modal imaging tracking of the location, distribution, and survival of the transplanted hMSCs in presence of nintedanib were achieved, which facilitates the profound understanding of the role the stem cells play in IPF therapy.
