Using farm management practices to predict Campylobacter prevalence in pastured poultry farms.

Contamination of poultry products by Campylobacter is often associated with farm management practices and processing plant practices. A longitudinal study was conducted on 11 pastured poultry farms in southeastern United States from 2014 to 2017. In this study, farm practices and processing variables were used as predictors for a random forest (RF) model to predict Campylobacter prevalence in pastured poultry farms and processing environments. Individual RF models were constructed for fecal, soil and whole carcass rinse after processing (WCR-P) samples. The performance of models was evaluated by the area under curve (AUC) from the receiver operating characteristics curve. The AUC values were 0.902, 0.894, and 0.864 for fecal, soil, and WCR-P models, respectively. Relative importance plots were generated to predict the most important variable in each RF model. Animal source of feces was identified as the most important variable in fecal model and the soy content of the brood feed was the most important variable for soil model. For WCR-P model, the average flock age showed the strongest impact on RF model. These RF models can help pastured poultry growers with food safety control strategies to reduce Campylobacter prevalence in pastured poultry farms.

Synergistic Engineering of Substituents and Backbones on Donor Polymers: Toward Terpolymer Design of High-Performance Polymer Solar Cells.

Design of terpolymers via copolymerization has emerged as a potential strategy for expanding the family of high-performing donor polymers and boosting the photovoltaic performance of non-fullerene polymer solar cells (PSCs). Herein, double-ester-substituted thiophenes and thienothiophenes are incorporated as third building blocks into the donor polymer PBDB-TF, developing two groups of terpolymers with donor-acceptor 1-donor-acceptor 2 (D-A1-D-A2)-type backbones. An optimum 10% concentration of double-ester-substituted thiophene units in PBDB-TF-T10 downshifts the molecular energy and increases the dielectric constant, and delivers proper miscibility and nanostructure in blends with the high-performing acceptor Y6. These characteristics are designed to synergistically enhance the photovoltage, photocurrent, and efficiency of PSCs. The resulting power conversion efficiency (PCE) of 16.4% surpasses the conventional PBDB-TF/Y6 PSCs, and it is among the best-performing PSCs based on PBDB-TF-derived terpolymers. Gratifyingly, PBDB-TF-T10 does not show significant batch-to-batch variation and it retains high PCEs above 16% in a broad range of molecular weights. This work introduces a facile strategy to easily synthesize terpolymers in combination with Y6 for the attainment of high-performing and reproducible non-fullerene PSCs.

[Analysis of genetic variation for a child affected with congenital insensitivity to pain with anhidrosis and albinism by whole genome sequencing].

Objective: To explore the genetic variation of a Chinese family affected with congenital insensitivity to pain with anhidrosis and albinism.

Methods: Whole exome sequencing (WES) was carried out to screen potential variants within genomic DNA extracted from the proband and his parents. Whole genome sequencing (WGS) was applied when variants were not found completely. Suspected variants were validated by Sanger sequencing.

Results: WES has identified a heterozygous c.1729G>C (p.G577R) variant of NTRK1 gene and two heterozygous variants of OCA2 gene, namely c.1363A>G (p.R455G) and c.1182+1G>A. WGS has identified two additional heterozygous variants c.(851-798C>T; 851-794C>G) in deep intronic regions of the NTRK1 gene.

Conclusion: The compound heterozygous variants of the NTRK1 gene probably underlay the congenital insensitivity to pain with anhidrosis. And the compound heterozygous variants of the OCA2 gene probably underlay the albinism in the proband. In the case where no variant is detected by WES in the coding region, WGS should be considered to screen potential variants in the whole genome.

Outcome of atypia of undetermined significance/follicular lesion of undetermined significance in thyroid fine-needle aspirations: A six-year institutional experience.

Background: Atypia of undetermined significance/follicular lesion of undetermined significance (AUS/FLUS) is one of six diagnostic categories of The Bethesda System for Reporting Thyroid Cytopathology (BSRTC). The goal of our study is to assess the outcome of cases classified as AUS/FLUS at our institution.

Methods: AUS/FLUS cases were identified by computer searching of the thyroid fine-needle aspiration (FNA) cases performed between 2010 and 2016. Outcomes were categorized as: follow-up surgery, repeat FNA or no follow-up available. Demographics, ultrasound findings and FNA diagnostic criteria were reviewed for AUS/FLUS cases with follow-up surgical pathology diagnosis.

Results: Our AUS/FLUS thyroid FNA rate was 6% (117 out of 1984 FNAs). Only 15% of the AUS/FLUS cases had repeat FNA, while 41% underwent surgery. The risk of malignancy (ROM) for cases with follow-up surgery was 17%. When considering all AUS/FLUS cases, the ROM was 7%. Statistically, benign neoplasms were more likely to be single lesions on ultrasound comparing to malignant neoplasms, and to exhibit architectural atypia as opposed to non-neoplastic lesions on FNA. The malignancy rates among patients that directly went to surgical resection (17%) and patients having repeat FNA after the first AUS/FLUS diagnosis followed by surgery (29%) was not significantly different. However, repeat FNA was able to reclassify the majority of cases into more definitive categories.

Conclusion: The outcome of the thyroid FNAs diagnosed as AUS/FLUS in our institution meets the benchmark statistics for AUS/FLUS rate and ROM. This study constitutes a valuable quality assurance measure and serves as a baseline for subsequent quality improvement.
