Physiological and biochemical characterization of Kalongi (Nigella sativa) against arsenic stress: Implications for human health risk assessment.

Arsenic (As) is a toxic metalloid that exhibits a varying degree of toxicity in plants depending upon the redox status of its species. Elemental arsenic [As(0)] is the least toxic of all the As species, however, under conducive environmental conditions, it can be readily oxidized into toxic forms. The present experiment was designed to evaluate the deleterious effects of As when applied in As(0) form on the morpho-physiological attributes of Kalongi (Nigella sativa). Seeds of N. sativa were sown in soil contaminated with various levels of As (0, 1.875, 3.75, 7.5, 15.0, and 30.0 mg nA(0) kg soil). The results indicated that plant biomass and grain yield of N. sativa were not much affected by various levels of As except at 30 mg nA(0) kg soil. Activities of antioxidant enzymes (SOD, APX, POX, and CAT), phenolic contents, and carotenoids were enhanced in response to the overproduction of HO, subsequently inhibiting lipid peroxidation. Arsenic accumulation in different plant organs increased with increasing soil As levels in the given trend root > shoot > leaf > seedpod > seed. Arsenic uptake affected the uptake of other elements (P, Fe, Zn, K, Na, Ca). Adaptive changes in total chlorophyll contents, MDA contents, and antioxidant enzymatic defense mechanism in response to As stress suggest that the N. sativa is tolerant to moderate As stress. Therefore, this crop can be cultivated on moderately As-contaminated soils without any significant risks of economic losses and food chain contamination.

Development of Liquid Diene Rubber Based Highly Deformable Interactive Fiber-Elastomer Composites.

The preparation of intelligent structures for multiple smart applications such as soft-robotics, artificial limbs, etc., is a rapidly evolving research topic. In the present work, the preparation of a functional fabric, and its integration into a soft elastomeric matrix to develop an adaptive fiber-elastomer composite structure, is presented. Functional fabric, with the implementation of the shape memory effect, was combined with liquid polybutadiene rubber by means of a low-temperature vulcanization process. A detailed investigation on the crosslinking behavior of liquid polybutadiene rubber was performed to develop a rubber formulation that is capable of crosslinking liquid rubber at 75 °C, a temperature that is much lower than the phase transformation temperature of SMA wires (90-110 °C). By utilizing the unique low-temperature crosslinking protocol for liquid polybutadiene rubber, soft intelligent structures containing functional fabric were developed. The adaptive structures were successfully activated by Joule heating. The deformation behavior of the smart structures was experimentally demonstrated by reaching a 120 mm bending distance at an activation voltage of 8 V without an additional load, whereas 90 mm, 70 mm, 65 mm, 57 mm bending distances were achieved with attached weights of 5 g, 10 g, 20 g, 30 g, respectively.

Acute spinal cord ischemia in a patient with Situs Inversus Totalis.

Situs inversus totalis (SIT) is a rare developmental abnormality where the organs throughout both the thoracic cavity and abdomen are a mirror image of normal anatomy, often occurring concomitantly with other genetic and developmental defects. Acute spinal cord ischemia is diagnosed based on the clinical presentation along with consistent imaging, but since clinical manifestations of acute spinal cord ischemia- rapidly progressive motor, sensory, and autonomic dysfunction-overlap with a wide spectrum of myelopathies, a thorough diagnostic workup with consideration of inflammatory, infectious, compressive and nutritional etiologies is required to establish the diagnosis. In this report, we present the case of an 18-year-old female patient who was admitted with acute onset of severe lower back pain, progressive weakness, paralysis, loss of sensation in both lower limbs and voiding difficulties. The diagnosis of acute spinal cord ischemia in a patient with situs inversus totalis was made. Our case highlights the spectrum of the pathological entities that can be associated with situs inversus totalis. Due to the lack of the classic signs and symptoms of sinus inversus, a diagnosis of situs inversus totalis with concomitant pathological conditions may require a more in-depth evaluation by complex imaging modalities to ensure a comprehensive assessment of the condition and its associated complications.

National Trends of Gender Disparity in Canadian Cardiovascular Society Guideline Authors, 2001-2020.

Background: The level of representation of women in cardiology remains low compared to that of men, particularly in leadership positions. We evaluated gender disparity in the authorship of Canadian Cardiovascular Society (CCS) guidelines.

Methods: All CCS guidelines from 2001-2020 were identified. Gender was assessed based on pronoun use in the biographies and social media of the authors. Only primary panel authors were included in our analysis. Stratified analyses were performed based on subspecialties.

Results: A total of 76 guidelines were identified, with 1172 authors (26% women, 74% men, < 0.0001), with no significant change in percentage of women authors over 2 decades (37.1% in 2001, 36.3% in 2020,  = 0.34). Inclusion of women as authors occurred less frequently than inclusion of men in general cardiology guidelines (20.1% vs 79.9%, < 0.0001) and all subspecialties-heart failure (36.4% vs 63.6%, < 0.0001), interventional cardiology (12.6% vs 87.4%, < 0.0001), electrophysiology (20.2% vs 79.8%, < 0.0001), and pediatric cardiology (41.7% vs 58.3%,  = 0.02). It was less likely for women to be a chair or cochair of a guideline writing committee, compared with men (20.1% vs 79.8%, < 0.0001). There were 609 unique authors (25.6% women, 74.4% men, < 0.0001), 542 unique medical doctorate (MD) authors (20.7% women, 79.3% men, < 0.0001), and 67 unique non-MD authors (65.7% women, 34.3% men,  = 0.0003).

Conclusions: There is a persistent shortfall in the inclusion of women authors for CCS guidelines, which has not changed over time. Further efforts are required to promote women's inclusion in leadership roles, which may lead to authorship of the guidelines.

A case with primary hyperaldosteronism associated with chronic kidney disease.

Primary hyperaldosteronism (PA) is one of the most common causes of secondary hypertension. PA may be associated with a decline in renal function. About 20% of cases with resistant HTN eventually cause PA, so all these patients should be evaluated for PA. Herein, we present a case with drug-resistant hypertension and chronic kidney disease (CKD), the cause of which was PA. Despite his low-salt diet modifications and treatment with several classes of antihypertensive medication, he had poorly controlled blood pressure (BP). Measurements of aldosterone and renin raised the concern of PA. Imaging confirmed bilateral adrenal hyperplasia. Due to the persistently high BP, despite the modification of the antihypertensive treatment, the patient underwent unilateral adrenalectomy, as the only feasible possibility of lowering aldosterone levels. After surgery, the patient had an improvement in both BP values and renal function. PA is difficult to diagnose in patients with CKD and Arterial Hypertension because hypertension is often associated with CKD, but PA accounts for a significant percentage of drug-resistant hypertension, so these patients should be screened for secondary arterial hypertension.

Structural characterization and antileishmanial activity of newly synthesized organo-bismuth(V) carboxylates: experimental and molecular docking studies.

In a quest to discover new formulations for the treatment of various parasitic diseases, a series of heteroleptic triorganobismuth(V) biscarboxylates of type [BiR(OCR')], where R=CH for 1-4 and p-CHCH for 5-8, were synthesized, characterized and evaluated for their biological potential against L. tropica. All the synthesized complexes were fully characterized by elemental analysis, FT-IR, multinuclear (H and C) NMR spectroscopy and X-ray crystallography. The crystal structures for [BiPh(OCCH(o-Br))] (1), [BiPh(OCCHCH)] (2), [BiPh(OCCH(m-NO))] (3) and [BiPh(OCCH(2-OH,3-CH))] (4) were determined and found to have a distorted pentagonal bipyramidal molecular geometry with seven coordinated bismuth center for 1-3 and for 4 distorted octahedral geometry, respectively. All the synthesized complexes demonstrated a moderate to significant activity against leishmania parasites. A broad analytical approach was followed to testify the stability for (1-8) in solid state as well as in solution and in leishmanial culture M199, ensuring them to be stable enough to exert a significant antileishmanial effect with promising results. Cytotoxicity profile suggests that tris(tolyl) derivatives show lower toxicity against isolated lymphocytes with higher antileishmanial potential. Molecular docking studies were carried out to reveal the binding modes for (1-8) targeting the active site of trypanothione reductase (TR) (PDB ID: 4APN) and Trypanothione Synthetase-Amidase structure (PDB ID 2vob).

Selected Thiadiazine-Thione Derivatives Attenuate Neuroinflammation in Chronic Constriction Injury Induced Neuropathy.

Neuropathic pain refers to a lesion or disease of peripheral and/or central somatosensory neurons and is an important body response to actual or potential nerve damage. We investigated the therapeutic potential of two thiadiazine-thione [TDT] derivatives, 2-(5-propyl-6-thioxo-1, 3, 5-thiadiazinan-3-yl) acetic acid [TDT1] and 2-(5-propyl-2-thioxo-1, 3, 5-thiadiazinan-3-yl) acetic acid [TDT2] against CCI (chronic constriction injury)-induced neuroinflammation and neuropathic pain. Mice were used for assessment of acute toxicity of TDT derivatives and no major toxic/bizarre responses were observed. Anti-inflammatory activity was assessed using the carrageenan test, and both TDT1 and TDT2 significantly reduced carrageenan-induced inflammation. We also used rats for the induction of CCI and performed allodynia and hyperalgesia-related behavioral tests followed by biochemical and morphological analysis using RT-qPCR, immunoblotting, immunohistochemistry and immunofluorescence. Our findings revealed that CCI induced clear-cut allodynia and hyperalgesia which was reversed by TDT1 and TDT2. To determine the function of TDT1 and TDT2 in glia-mediated neuroinflammation, Iba1 mRNA and protein levels were measured in spinal cord tissue sections from various experimental groups. Interestingly, TDT1 and TDT2 substantially reduced the mRNA expression and protein level of Iba1, implying that TDT1 and TDT2 may mitigate CCI-induced astrogliosis. molecular docking studies predicted that both compounds had an effective binding affinity for TNF-α and COX-2. The compounds interactions with the proteins were dominated by both hydrogen bonding and van der Waals interactions. Overall, these results suggest that TDT1 and TDT2 exert their neuroprotective and analgesic potentials by ameliorating CCI-induced allodynia, hyperalgesia, neuroinflammation and neuronal degeneration in a dose-dependent manner.

The Case for Early Antibiotic Commencement and Source Control in Paediatric Subdural Empyema: A Single-Centre Retrospective Case Series.

Background: Subdural empyema is a neurosurgical emergency requiring prompt diagnosis and treatment. There is a debate between the benefits and risks of starting early antibiotics prior to surgical drainage as this is purported to reduce the rate of microbiological diagnosis. Here, we describe our experience of treating this potentially life-threatening condition, advocating for the early commencement of antibiotics and importance of source control in its treatment.

Methods: Retrospective review of a prospectively collected electronic departmental database included all patients who were admitted to our unit with a diagnosis of subdural empyema over an 11-year period (2008-2018). Basic demographic data were collected. Further data pertaining to mode of presentation, surgical approach, causative organism, post-operative antibiotic regime, anti-seizure medications, length of hospital stay, further surgery, and neurological outcomes were extracted.

Results: Thirty-six children underwent 44 operations for subdural empyema at our institution during the study period. Median age was 11.0 (range 0.2-15.8); 47.2% (17/36) were female. Over time, there was decreasing use of burr holes and increasing use of craniectomy as the index surgery. Using a combination of extended culture and polymerase chain reaction, a microbiological diagnosis was achieved in all 36 cases; the commonest causative microorganism was of the Streptococcus anginosus group of bacteria. Seven patients underwent repeat surgery, and 4 patients underwent a concurrent ENT procedure. No risk factors were significant in predicting the likelihood of re-operation (location of subdural empyema, age, index surgery type, inflammatory markers, concurrent ENT procedure, and microorganism) although it was notable that none of the patients undergoing a concurrent ENT procedure underwent repeat surgery (p = 0.29). Median length of stay was 12 days (range 3-74), and there were no inpatient or procedure-related mortalities. Clinical outcomes were good with 94.4% (34/36) categorized as modified Rankin Scale 0-3 at discharge and there were 2 cranioplasty-related complications.

Conclusions: We observed an evolution of practice from limited surgical approaches towards more extensive index surgery over the study period. Given that a microorganism was isolated in all cases using a comprehensive approach, initiation of antibiotic therapy should not be delayed on presentation. Concurrent ENT surgery may be an important factor in providing aggressive source control thereby reducing the need for repeat surgery.

Sexual behavior influence against HIV/AIDS in homosexuals at Palu City in 2020.

Objective: The purpose of this study is to examine the sexual behavior relationship with HIV/AIDS on homosexuals in Palu City and its risk factors.

Method: The study was held using a case-control with a retrospective study. The number of samples on the case-control group is 90 respondents with 1:1 comparison. Sampling techniques and data collection based on the medical record of VCT in Anutapura Hospital by using the questionnaire. The data is processed using the univariate analysis, bivariate analysis by using chi-square, and multivariate analysis by using logistic regression analysis.

Results: Based on bivariate analysis gained that there is a significant relationship of sexual behavior in homosexuals with HIV/AIDS in Palu City. Dependent variable is HIV/AIDS while the independent variable are the number of pairs p=0.004 (OR=12.6); Anal sex p=0.003 (OR=3.6); Oral ejaculation p=0.002 (OR=14.2); Oral sex-licking/rimming p=0.035 (OR=2.5); Fingering p=0.019 (OR=2.8); and interfemoral coitus p=0.011 (OR=3.0). Based on the modeling of multivariate logistic regression analysis obtained, dominant variables are the number of pairs, anal sex, and interfemoral coitus, so the most dominant variables are the number of pairs with OR=20.8. Homosexuals that have more than one couple do bareback sex, and interfemoral coitus have an estimated probability of HIV/AIDS in Palu City is 83.7%.

Conclusion: There is a significant influence between sexual behavior against HIV/AIDS in homosexuals at Palu City in 2020.

Core-Shell MOF-in-MOF Nanopore Bifunctional Host of Electrolyte for High-Performance Solid-State Lithium Batteries.

Solid-state lithium-ion batteries with high safety are the encouraging next-generation rechargeable electrochemical energy storage devices. Yet, low Li conductivity of solid electrolyte and instability of solid-solid interface are the key issues hampering the practicability of the solid electrolyte. In this research, core-shell MOF-in-MOF nanopores [email protected] are proposed as a unique bifunctional host of ionic liquid (IL) to fabricate core-shell ionic liquid-solid electrolyte (CSIL). In the current design of CSIL, the shell structure (UIO-67) has a large pore size and a high specific surface area, boosting the absorption amount of ionic liquid electrolyte, thus increasing the ionic conductivity. Nevertheless, the core structure (UIO-66) has a small pore size compared to the ionic liquid, which can confine the large ions, decreasing their mobility, and selectively boost the transport of Li . The CSIL solid electrolyte exhibits considerable enhancement in the lithium transference number (t ) and ionic conductivity compared to the homogenous porous host (pure UIO-66 and UIO-67). Additionally, the Li|CSIL|Li symmetric batteries maintain a stable polarization of less than 28 mV for more than 1000 h at 1000 µA cm . Overall, the results demonstrate the concept of core-shell MOF-in-MOF nanopores as a promising bifunctional host of electrolytes for solid-state or quasi-solid-state rechargeable batteries.

The importance of imaging in tuberous sclerosis complex (tsc) in children: Two cases.

Tuberous sclerosis complex (TSC) is an inherited, multisystemic, hamartomatous neurocutaneous disorder, with an autosomal dominant inheritance pattern. It affects multiple organs, however the most susceptible ones include the brain, skin, kidneys, lungs, the retina, and the heart. TSC is characterized by considerable clinical heterogeneity. The majority of patients present with a constellation of clinical signs and symptoms, most prominently central nervous system manifestations including epilepsy, cognitive impairment and autism spectrum disorders, cutaneous, cardiac, renal and ophthalmic manifestations. Epilepsy affects 70% - 90% of patients, representing the primary neurological feature and 1 of the foremost clinical findings of the disorder. Cardiac rhabdomyomas are the most frequent cardiac manifestations, appearing as isolated or multiple lesions. Herein, we present 2 patients diagnosed with tuberous sclerosis. A 3-month-old male patient with cardiac rhabdomyomas and hypopigmented macules and a 19-month-old male patient with partial epilepsy and mild psychomotor retardation. As brain lesions represent some of the most prevalent clinical features and early onset seizures are associated with more severe cognitive, function delay, through this article we hope to emphasize the potential role MRI can play in the diagnostic workup of TSC, to ensure a more timely diagnosis, thus modifying the natural course of the disorder and its prognosis.

Surface-enhanced Raman scattering (SERS) spectroscopy for prostate cancer diagnosis: A review.

The present review focuses on the diagnosis of prostate cancer using surface enhanced Raman scattering (SERS) spectroscopy. On the basis of literature search, SERS-based analysis for prostate cancer detection of different sample types is reported in the present study. Prostate cancer is responsible for nearly one-tenth of all cell cancer deaths among men. Significant efforts have been dedicated to establish precise and sensitive monitoring techniques to detect prostate cancer biomarkers in different types of body samples. Among the various spectro-analytical techniques investigated to achieve this objective, SERS spectroscopy has been proven as a promising approach that provides noticeable enhancements of the Raman sensitivity when the target biomolecules interact with a nanostructured surface. The purpose of this review is to give a brief overview of the SERS-basedapproach and other spectro-analytical strategies being used for the detection and quantification of prostate cancer biomarkers. The revolutionary development of SERS methods for the diagnosis of prostate cancer has been discussed in more details based on the reported literature. It has been noticed that the SERS-based immunoassay presents reliable results for the prostate cancer quantification. The EC-SERS, which integrates electrochemistry with the SERS model, could also offer a potential ultrasensitive strategy, although its application in prostate cancer analysis has been still limited.

Genome-wide promoter analysis, homology modeling and protein interaction network of Dehydration Responsive Element Binding (DREB) gene family in Solanum tuberosum.

Dehydration Responsive Element Binding (DREB) regulates the expression of numerous stress-responsive genes, and hence plays a pivotal role in abiotic stress responses and tolerance in plants. The study aimed to develop a complete overview of the cis-acting regulatory elements (CAREs) present in S. tuberosum DREB gene promoters. A total of one hundred and four (104) cis-regulatory elements (CREs) were identified from 2.5kbp upstream of the start codon (ATG). The in-silico promoter analysis revealed variable sets of cis-elements and functional diversity with the predominance of light-responsive (30%), development-related (20%), abiotic stress-responsive (14%), and hormone-responsive (12%) elements in StDREBs. Among them, two light-responsive elements (Box-4 and G-box) were predicted in 64 and 61 StDREB genes, respectively. Two development-related motifs (AAGAA-motif and as-1) were abundant in StDREB gene promoters. Most of the DREB genes contained one or more Myeloblastosis (MYB) and Myelocytometosis (MYC) elements associated with abiotic stress responses. Hormone-responsive element i.e. ABRE was found in 59 out of 66 StDREB genes, which implied their role in dehydration and salinity stress. Moreover, six proteins were chosen corresponding to A1-A6 StDREB subgroups for secondary structure analysis and three-dimensional protein modeling followed by model validation through PROCHECK server by Ramachandran Plot. The predicted models demonstrated >90% of the residues in the favorable region, which further ensured their reliability. The present study also anticipated pocket binding sites and disordered regions (DRs) to gain insights into the structural flexibility and functional annotation of StDREB proteins. The protein association network determined the interaction of six selected StDREB proteins with potato proteins encoded by other gene families such as MYB and NAC, suggesting their similar functional roles in biological and molecular pathways. Overall, our results provide fundamental information for future functional analysis to understand the precise molecular mechanisms of the DREB gene family in S. tuberosum.

Thrombosis with Thrombocytopenia Syndrome After Administration of AZD1222 or Ad26.COV2.S Vaccine for COVID-19: A Systematic Review.

Background: Cases of thrombosis with thrombocytopenia syndrome (TTS) have been reported following vaccination with AZD1222 or Ad26.COV2.S. This review aimed to explore the pathophysiology, epidemiology, diagnosis, management, and prognosis of TTS.

Methods: A systematic review was conducted to identify evidence on TTS till 4 September 2021. Case reports and series reporting patient-level data were included. Descriptive statistics were reported and compared across patients with different sexes, age groups, vaccines, types of thrombosis, and outcomes.

Findings: Sixty-two studies reporting 160 cases were included from 16 countries. Patients were predominantly females with a median age of 42.50 (22) years. AZD1222 was administered to 140 patients (87·5%). TTS onset occurred in a median of 9 (4) days after vaccination. Venous thrombosis was most common (61.0%). Most patients developed cerebral venous sinus thrombosis (CVST; 66.3%). CVST was significantly more common in female vs male patients (p = 0·001) and in patients aged <45 years vs ≥45 years (p = 0·004). The mortality rate was 36.2%, and patients with suspected TTS, venous thrombosis, CVST, pulmonary embolism, or intraneural complications, patients not managed with non-heparin anticoagulants or IVIG, patients receiving platelet transfusions, and patients requiring intensive care unit admission, mechanical ventilation, or inpatient neurosurgery were more likely to expire than recover.

Interpretation: These findings help to understand the pathophysiology of TTS while also recommending diagnostic and management approaches to improve prognosis in patients.

Pregnant during the COVID-19 pandemic: Knowledge, concerns, attitudes and practices of Pakistani women.

Introduction: Data regarding pregnancy and related outcomes with COVID-19 are inconsistent, which leads to difficulties in counselling pregnant women. This brings uncertainty to pregnant women regarding mode of birth, transmission and issues that may occur in case they contract the disease. We conducted this study to assess the knowledge about COVID-19 risk during pregnancy and childbirth and to assess the concerns, attitudes, and practices of pregnant women during the pandemic.

Methods: A cross-sectional survey was carried out among 376 consenting pregnant women attending antenatal clinics in Karachi, Pakistan, between 1 July and 16 July 2020, using pretested questionnaires.

Results: A total of 376 pregnant women participated in the survey. Participants had inadequate knowledge about COVID-19 risk during pregnancy, including potential vertical transmission (58.5%), preferred route of delivery (52%), safety of breastfeeding (50%), birth defects (44.7%), rooming in and skin-to-skin contact (58.5%). The majority (85.1%) had a high concern score, and negative attitude (62.8%). Only 43.6% said that they would not hide their symptoms, while 37.2% stated that they will deliver at hospital if they tested positive. Only 30.9% of respondents had good practices. During visits, 39.4% maintained social distancing and sanitized hands while only 37.2% said that they did not bring more than one person with them.

Conclusions: The study population had inadequate knowledge, negative attitude, and poor practices regarding pregnancy during the COVID-19 pandemic. More than half of the respondents thought that concealing symptoms and delivering at home would be better. Awareness programs are urgently needed.

Single-step fabrication of highly stable amorphous TiO nanotubes arrays (am-TNTA) for stimulating gas-phase photoreduction of CO to methane.

This study investigates the facile fabrication of interfacial defects assisted amorphous TiO nanotubes arrays (am-TNTA) for promoting gas-phase CO photoreduction to methane. The am-TNTA catalyst was fabricated via a one-step synthesis, without heat treatment, by anodization of Titanium in Ethylene glycol-based electrolyte in a shorter anodizing time. The samples presented a TiO nanostructured array with a nanotubular diameter of 100 ± 10 nm, a wall thickness of 26 ± 5 nm, and length of 3.7 ± 0.3 μm, resulting in a specific surface of 0.75 m g. The am-TNTA presented prolonged chemical stability, a high exposed surface area, and a large number of surface traps that can reduce the recombination of the charge carriers. The am-TNTA showed promising photoactivity when tested in the CO reduction reaction with water under UV irradiation with a methane production rate of 14.0 μmol g h for a pure TiO material without any modification procedure. This enhanced photocatalytic activity can be explained in terms of surface defects of the amorphous structure, mainly OH groups that can act as electron traps for increasing the electron lifetime. The CO interacts directly with those traps, forming carbonate species, which favors the catalytic conversion to methane. The am-TNTA also exhibited a high stability during six reaction cycles. The photocatalytic activity, the significantly reduced time for synthesis, and high stability for continuous CH production make this nanomaterial a potential candidate for a sustainable CO reduction process and can be employed for other energy applications.

COVID-19 Fear Among Pakistanis: Psychometric Evaluation of the Fear of COVID-19 Scale Using Item Response Theory and Confirmatory Factor Analysis.

The Fear of COVID-19 Scale (FCV-19S) assesses the fear of the novel coronavirus disease 2019 (COVID-19) and has been translated and validated into over 20 languages. The present study conducted confirmatory factor analysis (CFA) and item response theory (IRT) analyses on the FCV-19S among a sample of 937 Pakistani adults (mean [SD] age of 25.83 [11.80] years; 537 [57.3%] females). The CFA and IRT confirmed the unidimensionality of the FCV-19S. The Likert-type scale used in the FCV-19S was supported by the proper threshold orderings. Additionally, no DIF contrast had an absolute value larger than 0.5 regarding the participants' characteristics of gender, age, living status, and education in the IRT findings. The FCV-19S was found to be valid and reliable with strong psychometric properties among the Pakistani adult population.

The effect of GnRH administration/insemination time on follicular growth rate, ovulation intervals, and conception rate of Nili Ravi buffalo heifers in 7 -day-CIDR Co-synch.

The present study aimed to compare two different insemination times (72 vs 84 h) associated with an ovulation induction (GnRH) in a 7-day CIDR Co-synch to improve the conception rate of Nili Ravi buffalo heifers. Forty Nili Ravi buffalo heifers were randomly separated into two treatments based on artificial insemination (AI) timing (72 vs 84 h). All heifers were subjected to controlled internal drug release (CIDR), containing 1.38 g of progesterone for 7 days. On CIDR removal, both treatments received 150 µg of prostaglandin intramuscularly. In 7-day CIDR Co-synch (n = 20), animals were injected 100 µg of GnRH administration intramuscularly and inseminated concurrently at 72 h after CIDR removal. The remaining half (n = 20) were injected and inseminated concurrently at 84 h of CIDR removal. Pregnancy diagnosis was performed on day 40 of timed artificial insemination (TAI) with ultrasound. The follicular growth rate between 72 h after PGF/CIDR removal to pre-ovulatory follicle in 7-day CIDR Co-synch was more (0.102 ± 0.005 mm vs 0.079 ± 0.003 mm; P = 0.01) at 84 than 72 h. The interval from GnRH administration/TAI to ovulation was high (26.8 ± 1.64 h vs. 15.1 ± 1.25 h, P = 0.01) in 72 than 84 h. Conception rates were considerably higher in buffalo heifers inseminated at 84 h (65%) than 72 h (25%) in 7-day CIDR Co-synch protocol. In conclusion that in Nili Ravi buffalo heifers, GnRH administration/TAI after 84 h of CIDR removal allows greater follicular growth rate and shortens interval from AI to ovulation compared to the GnRH administration/TAI after 72 h of CIDR removal in 7-day CIDR-Co-synch protocol.

Label-free quantitation of the changes in salivary proteome associated with the chronic consumption of the betel nut ().

Areca nut or betel nut chewing is most frequently used in Pakistan and is associated with a high risk for oral cancer. Until now, however, there has not been any research conducted on the long-term effect(s) of betel nut chewing on the saliva proteome. In the present study, initially, the changes in the saliva proteome associated with betel nut chewing were investigated. Secondly, the analysis was focused on the changes in salivary proteome with respect to prolonged usage of betel nuts. After extraction, the saliva proteins were digested into peptides and these were subsequently analyzed using mass spectrometry. Data are available ProteomeXchange with identifier PXD029768. Label-free quantitation of saliva samples revealed a total of 12 proteins that were differentially expressed between betel nut addicts (BNAs), and the control group. The study groups were further divided into three subgroups, the BNA-1, BNA-2, and BNA-3 groups, with respect to the extent of consumption of betel nuts in terms of years. The data analysis revealed a more detailed profiling of proteins expressed after five, ten, and more than ten years of betel nut consumption. A total of 30, 17, and 22 proteins were found to be differentially expressed when divided into the BNA-1, BNA-2, and BNA-3 groups. The present study shows that the chronic usage of betel nuts leads to the expression of proteins, such as SPARC1, profilin, and SBSN, which are known to be involved in head and neck cancers.

Biallelic inheritance in a single Pakistani family with intellectual disability implicates new candidate gene RDH14.

In a multi-branch family from Pakistan, individuals presenting with palmoplantar keratoderma segregate in autosomal dominant fashion, and individuals with intellectual disability (ID) segregate in apparent autosomal recessive fashion. Initial attempts to identify the ID locus using homozygosity-by-descent (HBD) mapping were unsuccessful. However, following an assumption of locus heterogeneity, a reiterative HBD approach in concert with whole exome sequencing (WES) was employed. We identified a known disease-linked mutation in the polymicrogyria gene, ADGRG1, in two affected members. In the remaining two (living) affected members, HBD mapping cross-referenced with WES data identified a single biallelic frameshifting variant in the gene encoding retinol dehydrogenase 14 (RDH14). Transcription data indicate that RDH14 is expressed in brain, but not in retina. Magnetic resonance imaging for the individuals with this RDH14 mutation show no signs of polymicrogyria, however cerebellar atrophy was a notable feature. RDH14 in HEK293 cells localized mainly in the nucleoplasm. Co-immunoprecipitation studies confirmed binding to the proton-activated chloride channel 1 (PACC1/TMEM206), which is greatly diminished by the mutation. Our studies suggest RDH14 as a candidate for autosomal recessive ID and cerebellar atrophy, implicating either disrupted retinoic acid signaling, or, through PACC1, disrupted chloride ion homeostasis in the brain as a putative disease mechanism.

Single-Crystal Investigation, Hirshfeld Surface Analysis, and DFT Study of Third-Order NLO Properties of Unsymmetrical Acyl Thiourea Derivatives.

In the current research work, unsymmetrical acyl thiourea derivatives, 4-((3-benzoylthioureido)methyl)cyclohexane-1-carboxylic acid and methyl 2-(3-benzoylthioureido)benzoate , have been synthesized efficiently. The structures of these crystalline thioureas were unambiguously confirmed by single-crystal diffractional analysis. The crystallographic investigation showed that the molecular configuration of both compounds is stabilized by intramolecular N-H···O bonding. The crystal packing of is stabilized by strong N-H···O bonding and comparatively weak O-H···S, C-H···O, C-H···π, and C-O···π interactions, whereas strong N-H···O bonding and comparatively weak C-H···O, C-H···S, and C-H···π interactions are responsible for the crystal packing of . The noncovalent interactions that are responsible for the crystal packing are explored by the Hirshfeld surface analysis for both compounds. The void analysis is performed to find the quantitative strength of crystal packing in both compounds. Additionally, state-of-the-art applied quantum chemical techniques are used to further explore the structure-property relationship in the above-entitled molecules. The optimization of molecular geometries showed a reasonably good correlation with their respective experimental structures. Third-order nonlinear optical (NLO) polarizability calculations were performed to see the advanced functional application of entitled compounds as efficient NLO materials. The average static γ amplitudes are found to be 27.30 × 10 and 102.91 × 10 esu for the compounds and , respectively. The γ amplitude of is calculated to be 3.77 times larger, which is probably due to better charge-transfer characteristics in . The quantum chemical analysis in the form of 3-D plots was also performed for their frontier molecular orbitals and molecular electrostatic potentials for understanding charge-transfer characteristics. We believe that the current investigation will not only report the new and compounds but also evoke the interest of the materials science community in their potential use in NLO applications.

Potential Use of Hyperspectral Reflectance as a High-Throughput Nondestructive Phenotyping Tool for Assessing Salt Tolerance in Advanced Spring Wheat Lines under Field Conditions.

The incorporation of stress tolerance indices (STIs) with the early estimation of grain yield (GY) in an expeditious and nondestructive manner can enable breeders for ensuring the success of genotype development for a wide range of environmental conditions. In this study, the relative performance of GY for sixty-four spring wheat germplasm under the control and 15.0 dS m NaCl were compared through different STIs, and the ability of a hyperspectral reflectance tool for the early estimation of GY and STIs was assessed using twenty spectral reflectance indices (SRIs; 10 vegetation SRIs and 10 water SRIs). The results showed that salinity treatments, genotypes, and their interactions had significant effects on the GY and nearly all SRIs. Significant genotypic variations were also observed for all STIs. Based on the GY under the control (GYc) and salinity (GYs) conditions and all STIs, the tested genotypes were classified into three salinity tolerance groups (salt-tolerant, salt-sensitive, and moderately salt-tolerant groups). Most vegetation and water SRIs showed strong relationships with the GYc, stress tolerance index (STI), and geometric mean productivity (GMP); moderate relationships with GYs and sometimes with the tolerance index (TOL); and weak relationships with the yield stability index (YSI) and stress susceptibility index (SSI). Obvious differences in the spectral reflectance curves were found among the three salinity tolerance groups under the control and salinity conditions. Stepwise multiple linear regressions identified three SRIs from each vegetation and water SRI as the most influential indices that contributed the most variation in the GY. These SRIs were much more effective in estimating the GYc (R = 0.64 - 0.79) than GYs (R = 0.38 - 0.47). They also provided a much accurate estimation of the GYc and GYs for the moderately salt-tolerant genotype group; YSI, SSI, and TOL for the salt-sensitive genotypes group; and STI and GMP for all the three salinity tolerance groups. Overall, the results of this study highlight the potential of using a hyperspectral reflectance tool in breeding programs for phenotyping a sufficient number of genotypes under a wide range of environmental conditions in a cost-effective, noninvasive, and expeditious manner. This will aid in accelerating the development of genotypes for salinity conditions in breeding programs.

Exploring the Impacts of Genotype-Management-Environment Interactions on Wheat Productivity, Water Use Efficiency, and Nitrogen Use Efficiency under Rainfed Conditions.

Wheat production under rainfed conditions is restrained by water scarcity, elevated temperatures, and lower nutrient uptake due to possible drought. The complex genotype, management, and environment (G × M × E) interactions can obstruct the selection of suitable high yielding wheat cultivars and nitrogen (N) management practices prerequisite to ensure food security and environmental sustainability in arid regions. The agronomic traits, water use efficiency (WUE), and N use efficiencies were evaluated under favorable and unfavorable weather conditions to explore the impacts of G × M × E on wheat growth and productivity. The multi-N rate (0, 70, 140, 210, and 280 kg N ha) field experiment was conducted under two weather conditions (favorable and unfavorable) using three wheat cultivars (AUR-809, CHK-50, and FSD-2008) in the Pothowar region of Pakistan. The experiments were laid out in randomized complete block design (RCBD), with split plot arrangements having cultivars in the main plot and N levels in the subplot. The results revealed a significant decrease in aboveground biomass, grain yield, crop N-uptake, WUE, and N use efficiency (NUE) by 15%, 22%, 21%, 18%, and 8%, respectively in the unfavorable growing season (2014-2015) as compared to favorable growing season (2013-2014) as a consequence of less rainfall and heat stress during the vegetative and reproductive growth phases, respectively. FSD-2008 showed a significantly higher aboveground biomass, grain yield, crop N-uptake, WUE, and NUE as compared to other wheat cultivars in both years. Besides, N appeared as the most suitable dose for wheat cultivars during the favorable growing season. However, any further increase in N application rates beyond N showed a non-significant effect on yield and yield components. Conversely, the wheat yield increased significantly up to 74% from N to N during the unfavorable growing season, and there was no substantial difference between N-N. The findings provide opportunities for maximizing yield while avoiding excessive N loss by selecting suitable cultivars and N application rates for rainfed areas of Pothowar Plateau by using meteorological forecasting, amount of summer rainfall, and initial soil moisture content.

Effect of APOB polymorphism rs562338 (G/A) on serum proteome of coronary artery disease patients: a "proteogenomic" approach.

In the current study, APOB (rs1052031) genotype-guided proteomic analysis was performed in a cohort of Pakistani population. A total of 700 study subjects, including Coronary Artery Disease (CAD) patients (n = 480) and healthy individuals (n = 220) as a control group were included in the study. Genotyping was carried out by using tetra primer-amplification refractory mutation system-based polymerase chain reaction (T-ARMS-PCR) whereas mass spectrometry (Orbitrap MS) was used for label free quantification of serum samples. Genotypic frequency of GG genotype was found to be 90.1%, while 6.4% was for GA genotype and 3.5% was for AA genotypes in CAD patients. In the control group, 87.2% healthy subjects were found to have GG genotype, 11.8% had GA genotype, and 0.9% were with AA genotypes. Significant (p = 0.007) difference was observed between genotypic frequencies in the patients and the control group. The rare allele AA was found to be strongly associated with the CAD [OR: 4 (1.9-16.7)], as compared to the control group in recessive genetic model (p = 0.04). Using label free proteomics, altered expression of 60 significant proteins was observed. Enrichment analysis of these protein showed higher number of up-regulated pathways, including phosphatidylcholine-sterol O-acyltransferase activator activity, cholesterol transfer activity, and sterol transfer activity in AA genotype of rs562338 (G>A) as compared to the wild type GG genotype. This study provides a deeper insight into CAD pathobiology with reference to proteogenomics, and proving this approach as a good platform for identifying the novel proteins and signaling pathways in relation to cardiovascular diseases.

Bilateral basal ganglia ischemia associated with COVID-19: a case report and review of the literature.

Background: Coronavirus disease 2019, caused by the severe acute respiratory syndrome coronavirus 2, has a broad clinical spectrum, from asymptomatic to multi-organ dysfunction. Acute cerebrovascular events associated with coronavirus disease 2019 are mainly due to the severe acute respiratory syndrome coronavirus 2-induced prothrombotic state. Bilateral basal ganglia ischemia is rarely reported.

Case Presentation: We report the case of a 64-year-old Asian (Pakistani) gentleman who presented initially with fever, cough, and shortness of breath, likely due to respiratory involvement by severe acute respiratory syndrome coronavirus 2. Later, he developed bilateral lower limb pain, followed by confusion and decreased level of consciousness. Accentuated large hypodense opacities were seen in the left and right basal ganglia, with mass effects on the left frontal horn.

Conclusion: This case demonstrates the importance of neuroimaging in the effective management of patients with neurological signs associated with coronavirus disease 2019.

A survey was conducted at 720 points in different districts of the province. The STSS detected "active" and emerging clusters that are current at the end of our study area-particularly, 17 clusters were formed while adding the updated case data. Software ArcGIS 10.3 was used to find relative risk (RR) values; the maximum RR value was found to be 42.19 and maximum observed cases 53,265 during June 15-July 1, 2020. It was not always necessary that if the number of active cases in Punjab increased, there should be higher relative risk for more number of districts and vice versa. Due to the highest number of cases of COVID-19 and RR values during July, mostly farmers faced many difficulties during the cultivation of cotton and rice. Mostly farmers (72%) observed increase in prices of inputs (fertilizers and pesticides) during lockdown. If the supply chain of agriculture related inputs is disturbed, farmers may find it quite difficult to access markets, which could result in a decline in production and sales of crops and livestock in study area. It is suggested that to protect the food security and to decrease the effect of the lockdown, Punjab government needs to review food policy and analyse how market forces will respond to the imbalanced storage facilities and capacity, supply and demand and price control of products. The findings of this study can also help policy-makers to formulate an effective food security and agriculture adaptation strategy.

Facile One-Step Microwave-Assisted Method to Synthesize Nickel Selenide Nanosheets for High-Performance Hybrid Supercapacitor.

Nanosheets structures can be employed as the most promising electrode material to enhance electrochemical performance for supercapacitors. Nickel Selenide (NiSe) nanosheets are synthesized using a rapid microwave synthesis method in a single step. The NiSe nanosheets possess a high surface area (125 mg) with a hexagonal crystalline structure. It shows magnificent electrochemical properties, such as splendid specific capacitance (2530 Fg at 0.5 Ag). An asymmetric hybrid supercapacitor is fabricated with nickel selenide nanosheets as a positive electrode and activated carbon as a negative electrode. The assembled hybrid supercapacitor displays a high energy density of 63.5 WhKg at a power density of 404 WKg, and after 8000 cycles, only 5% capacitance is lost along with the better voltage window at 0-1.6 V.
