Melanoma-Differentiation-Associated gene 5 protects against nonalcoholic steatohepatitis in mice.

Background And Aims: Nonalcoholic steatohepatitis (NASH) is a complicated disease characterized by hepatocyte steatosis, inflammation infiltration, and liver fibrosis. Accumulating evidence suggests that the innate immunity plays a key role in NASH progression. Here, we aimed to reveal the role of melanoma-differentiation-associated gene 5 (MDA5, also known as Ifih1), a conventional innate immune regulator upon viral infection, in the progression of NASH and investigate its underlying mechanism.

Approach And Results: We first examined the expression of MDA5 and found that MDA5 was markedly downregulated in the livers with NASH in human individuals and mice models. MDA5 overexpression significantly inhibited the free fatty acid (FFA) induced lipid accumulation and inflammation in hepatocyte in vitro, while MDA5 knockdown promotes hepatocyte lipotoxicity. Using hepatocyte-specific Mda5 gene knockout and transgenic mice, we found that diet induced hepatic steatosis, inflammation and liver fibrosis were markedly exacerbated by Mda5 deficiency but suppressed by Mda5 overexpression. Mechanistically, we found that the activation of ASK1-MAPK pathway was significantly inhibited by MDA5 but enhanced by MDA5 deletion. We further validated that MDA5 directly interacted with ASK1 and suppressed its N-terminal dimerization. Importantly, blockage of ASK1 with adenovirus expressing dominant negative ASK1 (dnASK1) obviously reversed the lipid accumulation and ASK1 pathway activation when Mda5 knockout.

Conclusions: These data indicate that MDA5 is an essential suppressor in NASH, the findings support MDA5 as a novel regulator of ASK1 and a promising therapeutic target for NASH.

SIMPLE is an endosomal regulator that protects against non-alcoholic fatty liver disease by targeting the lysosomal degradation of EGFR.

Background & Aims: Nonalcoholic fatty liver disease (NAFLD) has become a tremendous burden for public health, however, there is no drug for NAFLD therapy at present. Impaired endo-lysosome-mediated protein degradation is observed in a variety of metabolic disorders, such as atherosclerosis, type 2 diabetes mellitus and NAFLD. Small integral membrane protein of lysosome/late endosome (SIMPLE) is a regulator of endosome-to-lysosome trafficking and cell signaling. But the role that SIMPLE plays in NAFLD progression remains unknown. Here we investigated SIMPLE function in NAFLD development and sophisticated mechanism therein.

Approach & Results: This study found in vitro knockdown of SIMPLE significantly aggravated lipid accumulation, inflammation in hepatocytes treated with metabolic stimulation. Consistently, in vivo experiments showed that liver-specific Simple-knockout (Simple-HKO) mice exhibited more severe high-fat diet (HFD)-, high-fat-high-cholesterol diet (HFHC)-, and methionine-choline-deficient diet (MCD)- induced steatosis, glucose intolerance, inflammation, and fibrosis than those fed with normal-chow diet. Meanwhile, RNA-sequencing demonstrated the up-regulated signaling pathways and signature genes involved in lipid metabolism, inflammation and fibrosis in Simple-HKO mice compared to control mice under metabolic stress. Mechanically, we found SIMPLE directly interact with epidermal growth factor receptor (EGFR). SIMPLE deficiency results in dysregulated degradation of EGFR, subsequently hyperactivated EGFR phosphorylation, exaggerating NAFLD development. Moreover, we further demonstrated that using EGFR inhibitor or silencing EGFR expression could ameliorate lipid accumulation induced by the knockdown of SIMPLE.

Conclusions: SIMPLE ameliorated NASH by prompting EGFR degradation and can also be a potential therapeutic candidate for NASH.

Gastrodin improves nonalcoholic fatty liver disease via activation of the AMPK signaling pathway.

Background & Aims: Nonalcoholic steatohepatitis (NASH) is currently one of the most common causes of liver transplantation and hepatocellular carcinoma. Thus far, there is still no effective pharmacological therapy for this disease. Recently, Gastrodin has demonstrated hepatoprotective effects in a variety of liver diseases. The aim of this study is to investigate the function of Gastrodin in NASH.

Approach And Result: In our study, Gastrodin showed potent therapeutic effects on NASH both in vivo and in vitro. In high-fat diet (HFD)- or high-fat and high-cholesterol (HFHC) diet-fed mice, the liver weight, hepatic and serum triglyceride and cholesterol contents, the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity levels were markedly reduced by Gastrodin treatment as compared to the corresponding vehicle groups. Notably, Gastrodin showed minimal effects on the function and histological characteristics of other major organs in mice. We further examined the effects of Gastrodin on lipid accumulation in primary mouse hepatocytes and human hepatocyte cell line, and observed that Gastrodin showed a significant decrease in lipid accumulation and inflammatory response in hepatocytes under metabolic stress. Furthermore, RNA-Seq analysis systemically indicated that Gastrodin suppressed the pathway and key regulators related to lipid accumulation, inflammation and fibrosis in the pathogenesis of NASH. Mechanistically, we found that Gastrodin protected against NASH by activating the AMPK pathway, which was supported by the result that the AMPK inhibitor compound C or AMPK knockdown blocked the Gastrodin-mediated hepatoprotective effect.

Conclusion: Gastrodin attenuates steatohepatitis by activating the AMPK pathway and represents a novel therapeutic for the treatment of NASH.

The E3 Ubiquitin Ligase Ring Finger Protein 5 Ameliorates NASH Through Ubiquitin-Mediated Degradation of 3-Hydroxy-3-Methylglutaryl CoA Reductase Degradation Protein 1.

Background And Aims: NAFLD is the most prevalent chronic liver disease worldwide, but no effective pharmacological therapeutics are available for clinical use. NASH is the more severe stage of NAFLD. During this progress, dysregulation of endoplasmic reticulum (ER)-related pathways and proteins is one of the predominant hallmarks. We aimed to reveal the role of ring finger protein 5 (RNF5), an ER-localized E3 ubiquitin-protein ligase, in NASH and to explore its underlying mechanism.

Approach And Results: We first inspected the expression level of RNF5 and found that it was markedly decreased in livers with NASH in multiple species including humans. We then introduced adenoviruses for Rnf5 overexpression or knockdown into primary mouse hepatocytes and found that palmitic acid/oleic acid (PAOA)-induced lipid accumulation and inflammation in hepatocytes were markedly attenuated by Rnf5 overexpression but exacerbated by Rnf5 gene silencing. Hepatocyte-specific Rnf5 knockout significantly exacerbated hepatic steatosis, inflammatory response, and fibrosis in mice challenged with diet-induced NASH. Mechanistically, we identified 3-hydroxy-3-methylglutaryl CoA reductase degradation protein 1 (HRD1) as a binding partner of RNF5 by systematic interactomics analysis. RNF5 directly bound to HRD1 and promoted its lysine 48 (K48)-linked and K33-linked ubiquitination and subsequent proteasomal degradation. Furthermore, Hrd1 overexpression significantly exacerbated PAOA-induced lipid accumulation and inflammation, and short hairpin RNA-mediated Hrd1 knockdown exerted the opposite effects. Notably, Hrd1 knockdown significantly diminished PAOA-induced lipid deposition, and up-regulation of related genes resulted from Rnf5 ablation in hepatocytes.

Conclusions: These data indicate that RNF5 inhibits NASH progression by targeting HRD1 in the ubiquitin-mediated proteasomal pathway. Targeting the RNF5-HRD1 axis may provide insights into the pathogenesis of NASH and pave the way for developing strategies for NASH prevention and treatment.

TfOH-Catalyzed [4 + 1] Annulation of -Quinone Methides with α-Aryl Diazoacetates: Straightforward Access to Highly Functionalized 2,3-Dihydrobenzofurans.

We have developed a methodology for the greatly efficient construction of significant 2,3-dihydrobenzofuran scaffolds bearing a quaternary carbon center at the C2 position by means of [4 + 1] annulation reactions between -quinone methides and α-aryl diazoacetates as C1 synthons through organocatalysis by readily accessible TfOH catalyst under mild and transition metal-free conditions. This metal-free protocol furnishes an operationally simple and swift process for the free assembly of diverse highly functionalized 2,3-dihydrobenzofurans and also features broad substrate scope, excellent functional group compatibility, and environmental friendliness. Mechanistic investigation suggested that the reaction undergoes a rapid cascade protonation/intermolecular Michael addition/intramolecular substitution process.

We suggests that the disease name and diagnostic criteria should refer to the authoritative standards; the clinical study design should based on the realistic clinical problems; the efficacy evaluation index should based on the clinical needs and be analyzed targetedly; further research on common acupuncture or therapy should be carried out.

Cordycepin Ameliorates Nonalcoholic Steatohepatitis by Activation of the AMP-Activated Protein Kinase Signaling Pathway.

Background And Aims: Nonalcoholic fatty liver disease, especially nonalcoholic steatohepatitis (NASH), has become a major cause of liver transplantation and liver-associated death. NASH is the hepatic manifestation of metabolic syndrome and is characterized by hepatic steatosis, inflammation, hepatocellular injury, and different degrees of fibrosis. However, there is no US Food and Drug Administration-approved medication to treat this devastating disease. Therapeutic activators of the AMP-activated protein kinase (AMPK) have been proposed as a potential treatment for metabolic diseases such as NASH. Cordycepin, a natural product isolated from the traditional Chinese medicine Cordyceps militaris, has recently emerged as a promising drug candidate for metabolic diseases.

Approach And Results: We evaluated the effects of cordycepin on lipid storage in hepatocytes, inflammation, and fibrosis development in mice with NASH. Cordycepin attenuated lipid accumulation, inflammation, and lipotoxicity in hepatocytes subjected to metabolic stress. In addition, cordycepin treatment significantly and dose-dependently decreased the elevated levels of serum aminotransferases in mice with diet-induced NASH. Furthermore, cordycepin treatment significantly reduced hepatic triglyceride accumulation, inflammatory cell infiltration, and hepatic fibrosis in mice. In vitro and in vivo mechanistic studies revealed that a key mechanism linking the protective effects of cordycepin were AMPK phosphorylation-dependent, as indicated by the finding that treatment with the AMPK inhibitor Compound C abrogated cordycepin-induced hepatoprotection in hepatocytes and mice with NASH.

Conclusion: Cordycepin exerts significant protective effects against hepatic steatosis, inflammation, liver injury, and fibrosis in mice under metabolic stress through activation of the AMPK signaling pathway. Cordycepin might be an AMPK activator that can be used for the treatment of NASH.

Multidimensional Image and Beam Splitter Based on Hyperbolic Metamaterials.

Recently, multidimensional metasurfaces, which can modulate multiple optical parameters of input and output lights simultaneously, have aroused intensive interest. Here, we demonstrate multidimensional switchable images and 3D integrated beam splitters based on hyperbolic metamaterials (HMMs). On one hand, a switchable image controlled by output helicity and input wavelengths is achieved by arranging HMMs with different polarization conversion performance. On the other hand, polarization-multiplexed broadband beam splitter is generated by spatially engineering the subunit with broadband half-plate performance. By integrating the multiplexed beam splitter with a filter metamaterial, this multidimensional beam splitter can further realize the separation of output light by space and wavelength. This cascaded multilayer metamaterial achieves a brand new optical functionality and offers more inspiring possibilities for the design of multifunctional optical devices in the future.

[The Ultrasonographic Features of Female Reproductive System Extraosseous Ewing's Sarcoma].

Objective: This study investigated the clinicopathological traits and ultrasound features of female reproductive system extraosseous Ewing's sarcoma (EES) and explored the diagnostic value of ultrasonography for this condition.

Methods: Cases of female pelvic EES diagnosed and treated at our hospital between June 2009 and June 2019 were included in this study. Pathology data and ultrasound manifestations were assessed retrospectively to summarize the clinical traits and ultrasound features of female reproductive system EES. Based on the results, recommendations for the ultrasonography-based diagnosis of this disease were proposed.

Results: During the 10-year study period, 13 female patients were diagnosed with EES in the pelvic cavity based on the results of postoperative pathology tests. The age of the patients ranged from 8 mouth to 40 years, and no patients demonstrated specific clinical symptoms. However, an examination of tumor biomarkers revealed that certain patients had elevated levels of CA125. In the 13 patients, 19 lesions were identified, including 16 that involved the reproductive system. The primary ultrasound manifestation was uneven, low-echo solidity or cystic solidity, exhibiting large size, irregular shape, and unclear boundary. A few patients had concurrent ascites. Although some lesions lacked blood supply, the blood supply of most lesions was medium to abundant, and the blood flow was mostly characterized by low resistant. Almost none of the lesions were definitively diagnosed preoperatively.

Conclusions: Preoperative definitive diagnosis of EES in the female reproductive system remains a great clinical challenge. Although certain clinical traits and ultrasound features are associated with this disease, and color Doppler ultrasonography might provide vital information indicating the presence of EES, the final diagnosis still depends on the pathological test results of the patients.

This metal-free protocol provides an operationally simple and rapid method for the one-pot assembly of diverse α-aryl benzofuranones in high yields with broad substrate scope, a readily starting material, good chemo-regioselectivity, and excellent functional group compatibility.

Ultrasound diagnosis and clinicopathological traits of female genital system malignant lymphomas.

This study investigated the clinicopathological traits and ultrasound features (including 2-dimensional and color Doppler ultrasound) of female genital system malignant lymphomas and explored the diagnostic performance of ultrasonography in this disease.Cases of female pelvic lymphoma diagnosed and treated at our hospital between July 2009 and July 2019 were included in this study. Pathological data and ultrasonic manifestations were assessed retrospectively to summarize the clinical traits and ultrasonic features of female genital system lymphoma (FGSL). Based on the results, recommendations for ultrasonography-based diagnosis of this disease are proposed.During the 10-year study period, 20 female patients were diagnosed with malignant lymphomas in the pelvic cavity based on postoperative pathology tests. The age of the patients ranged from 11 to 83 years, and no patients demonstrated specific clinical traits; however, examination of tumor biomarkers revealed that certain patients had elevated levels of CA125. Twenty-eight lesions were identified in the 20 patients, including 24 involving the reproductive system. The primary ultrasonic manifestations were hypoechoic or extremely hypoechoic solid lesions that were relatively large in size, had a relatively regular shape, and had clear boundaries. Certain patients had concurrent ascites, and although some lesions lacked blood supply, most lesions had medium to abundant blood flow, which was largely characterized by low resistance. Almost none of the lesions were definitively diagnosed preoperatively.Diagnosing malignant lymphomas in the female genital tract remains a considerable clinical challenge. Although certain clinical traits and ultrasonic features are associated with this disease and color Doppler ultrasonography might provide vital information indicating the presence of lymphoma, the final diagnosis depends on the clinical and pathological test results of the patients.

Independent tuning of bright and dark meta-atoms with phase change materials on EIT metasurfaces.

The realization of tunable metasurfaces is of fundamental importance for boosting the electromagnetic field control ability. Especially, it is important to put forward new modulation methods to further understand their underlying modulation mechanism and expand their application range. In this paper, tunable electromagnetically induced transparency (EIT) metasurfaces based on the phase change material Ge2Sb2Te5 (GST) are proposed and experimentally demonstrated. Different from previous modulation methods of directly introducing the GST film below the metasurfaces, here a two-step lithography method is introduced to combine independent GST strips with bright and dark meta-atoms in the EIT structures, respectively, achieving the independent modulation of the EIT-like spectra. In addition, by applying temporal coupled-mode theory (TCMT), the EIT-like spectra with different GST crystallization levels were analysed and the corresponding characteristic parameters were determined simultaneously. These fitting results reveal that GST strips can modulate the resonances of the bright and dark meta-atoms independently by shifting the resonant frequency and increasing the decay rate, which in turn result in the different modulation features of the EIT-like spectra. This method improves the degree of freedom of active modulation and provides a new route for tunable slow light devices.

Organocatalytic Asymmetric Domino Oxa-Michael-Mannich-[1,3]-Amino Rearrangement Reaction of -Tosylsalicylimines to α,β-Unsaturated Aldehydes by Diarylprolinol Silyl Ethers.

An organocatalytic asymmetric enantioselective domino oxa-Michael-Mannich-[1,3]-amino rearrangement reaction of -tosylsalicylimines with a wide range of α,β-unsaturated aldehydes utilizing diarylprolinol silyl ether catalysis is described. The catalytic reactions proceed with excellent enantioselectivity (up to 99% ee) to produce the corresponding chair -tosylimines-chromenes with a yield of up to 99%, tolerating a range of functional groups. This methodology offers a new method with great potential to further extend the synthetic power and versatility of chiral aminocatalysis.

[Acupuncture artificial menstrual cycle method for abnormal uterine bleeding-ovulatory dysfunction (spleen deficiency syndrome)].

Objective: To compare the clinical efficacy between acupuncture artificial menstrual cycle method and medication artificial menstrual cycle method for abnormal uterine bleeding-ovulatory (AUB-O) dysfunction (spleen deficiency syndrome).

Methods: Sixty patients were randomly assigned into an observation group and a control group, 30 cases in each one. 3 cases dropped out in the observation group and 27 cases were included into analysis; 2 cases dropped out in the control group and 28 cases were included into analysis. The patients in the observation group were treated with acupuncture artificial menstrual cycle method. The acupoints were selected according to different stages of menstrual cycle. The acupuncture was given for 30 min per treatment, once every other day; no treatment was given during menstrual period. Xuehai (SP 10), Sanyinjiao (SP 6), Taixi (KI 3), Guanyuan (CV 4), Qihai (CV 6), Zusanli (ST 36) and Zhangmen (LR 13) were selected after menstrual period. Taichong (LR 3), Hegu (LI 4), Yaoyangguan (GV 3), Geshu (BL 17), Ganshu (BL 18), Sanyinjiao (SP 6), Pishu (BL 20) were selected in ovulaton periods. Baihui (GV 20), Shenshu (BL 23), Yaoyangguan (GV 3), Geshu (BL 17), Sanyinjiao (SP 6), Zusanli (ST 36), Gongsun (SP 4) were selected before menstrual period. The patients in the control group were treated with medication artificial menstrual cycle method. Femoston (estradiol tablets/estradiol dydrogesterone tablets) was taken orally. The white tablets were estradiol (1 mg), while the gray tablets were estradiol (1 mg) and dydrogesterone (10 mg). The Femoston was taken once a day, white tablets for the first 14 days, then gray tablets for the last 14 days. The 28-day treatment was taken as one session, and one cycle menstrual was taken as one treatment course, and totally 3 courses were given in the two groups. The clinical symptom score was observed before and after treatment as well as in follow-up visit (3 months after treatment). The clinical efficacy was evaluated based on the total effective rate, the marked effective rate of main symptoms (including menstrual cycle, menstrual period, menstrual volume) and basal body temperature (BBT).

Results: ① Compared before treatment, the clinical symptom score in the two groups was decreased after treatment and in the follow-up visit (<0.05). Compared with the control group, the clinical symptom score after treatment as well as the difference of that before and after treatment had no significant difference in the observation group (>0.05). Compared with the control group, the clinical symptom score in the follow-up visit as well as the difference of that between pre-treatment and follow-up visit had significant difference in the observation group (<0.05, <0.01). ② The total effective rate was 81.5% (22/27) in the observation group and 85.7% (24/28) in the control group, without significant difference between the two groups (>0.05). ③ After treatment, the marked effective rate of menstrual cycle, menstrual period, menstrual volume had no significant difference between the two groups (>0.05); during the follow-up visit, the marked effective rate of menstrual cycle in the observation group was superior to that in the control group (<0.05), but that of menstrual period and menstrual volume had no significant difference between the two groups (>0.05). ④ After treatment, the diphasic curve rate of BBT had no significant difference between the two groups (>0.05); during the follow-up visit, the diphasic curve rate of BBT in the observation group was higher than that in the control group (<0.05).

Conclusion: Acupuncture artificial menstrual cycle method has superior efficacy for AUB-O dysfunction (spleen deficiency syndrome), which has similar efficacy with medication artificial menstrual cycle method. Moreover, acupuncture has advantages in regulating menstrual cycle and improving ovulation, and has a longer curative effect.

Access to a Key Building Block for the Prostaglandin Family via Stereocontrolled Organocatalytic Baeyer-Villiger Oxidation.

A new protocol for the construction of a crucial bicyclic lactone of prostaglandins using a stereocontrolled organocatalytic Baeyer-Villiger (B-V) oxidation was developed. The key B-V oxidation of a racemic cyclobutanone derivative with aqueous hydrogen peroxide has enabled an early-stage construction of a bicyclic lactone skeleton in high enantiomeric excess (up to 95 %). The generated bicyclic lactone is fully primed with two desired stereocenters and enabled the synthesis of the entire family of prostaglandins according to Corey's route. Furthermore, the reactivity and enantioselectivity of B-V oxidation of racemic bicyclic cyclobutanones were evaluated and 90-99 % ee was obtained, representing one of the most efficient routes to chiral lactones. This study further facilitates the synthesis of prostaglandins and chiral lactone-containing natural products to promote drug discovery.

Spin-Selective Transmission in Chiral Folded Metasurfaces.

Controlling the spin angular momentum of light (or circular polarization state) plays a crucial role in the modern photonic applications such as optical communication, circular dichroism spectroscopy, and quantum information processing. However, the conventional approaches to manipulate the spin of light require naturally occurring chiral or birefringent materials of bulky sizes due to the weak light-matter interactions. Here we experimentally demonstrate an approach to implement spin-selective transmission in the infrared region based on chiral folded metasurfaces that are capable of transmitting one spin state of light while largely prohibiting the other. Due to the intrinsic chirality of the folded metasurface, a remarkable circular dichroism as large as 0.7 with the maximum transmittance exceeding 92% is experimentally demonstrated. The giant circular dichroism is interpreted within the framework of charge-current multipole expansion. Moreover, the intrinsic chirality can be readily controlled by manipulating the folding angle of the metasurface with respect to the cardinal plane. Benefiting from its strong chirality and spin-dependent transmission characteristics, the proposed folded metasurface may be applied to a range of novel photon-spin selective devices for optical communication technologies and biophotonics.

Neocucurbitacin D, a novel lactone-type norcucurbitacin as xanthine oxidase inhibitor from .

A novel lactone-type norcucurbitacin, designated as neocucurbitacin D (), together with five known cucurbitane triterpenes were isolated from traditional Tibetan medicine "Se Ji Mei Duo", which is the seed of (Ser.) C.B. Clarke. The structure of neocucurbitacin D was elucidated by spectroscopic analysis, including 2D NMR and X-ray techniques. Compounds were screened for their xanthine oxidase (XOD) inhibitory activity. Compound and exhibited significant XOD inhibition with IC values ranging from 10.16 to 18.41 μM. The absolute stereochemistry and XOD inhibitiory activity of lactone-type norcucurbitacins was reported firstly.

Construction of Hierarchical MoSe Hollow Structures and Its Effect on Electrochemical Energy Storage and Conversion.

Metal selenides have attracted increased attention as promising electrode materials for electrochemical energy storage and conversion systems including metal-ion batteries and water splitting. However, their practical application is greatly hindered by collapse of the microstructure, thus leading to performance fading. Tuning the structure at nanoscale of these materials is an effective strategy to address the issue. Herein, we craft MoSe with hierarchical hollow structures via a facile bubble-assisted solvothermal method. The temperature-related variations of the hollow interiors are studied, which can be presented as solid, yolk-shell, and hollow spheres, respectively. Under the simultaneous action of the distinctive hollow structures and interconnections among the nanosheets, more intimate contacts between MoSe and electrolyte can be achieved, thereby leading to superior electrochemical properties. Consequently, the MoSe hollow nanospheres prepared under optimum conditions exhibit optimal electrochemical activities, which hold an initial specific capacity of 1287 mA h g and maintain great capacity even after 100 cycles as anode for Li-ion battery. Moreover, the Tafel slope of 58.9 mV dec for hydrogen evolution reaction is also attained.

[Clinical trial of gastroesophageal reflux disease with the disharmony between liver and stomach syndrome treated with acupuncture regulating based on the compatibility of the five meridians].

Objective: To observe the clinical effects of the acupuncture regulating based on the compatibility of the five meridians and common western medication for gastroesophageal reflux disease with the disharmony between liver and stomach.

Methods: Sixty patients were divided into two groups according to non-erosive reflux disease (NERD) and reflux esophagitis (RE). Those in the two groups were randomly assigned into an observation group and a control group. There were 57 cases included (28 in the observation group with 2 dropping, 29 in the control group with 1 dropping). Based on the life care, patients in the observation group were treated with 30 min acupuncture regulating based on the compatibility of the five meridians at Zusanli (ST 36), Chongyang (ST 42), Weishu (BL 21), Zhongwan (CV 12), Xingjian (LR 2), Shaofu (HT 8), Dadun (LR 1), Taichong (LR 3), Ganshu (BL 18), Qimen (LR 14), Danzhong (CV 17), Zhongting (CV 16), three times a week. Patients in the control group were treated with rabeprazole orally. All the treatment was given for consecutive 8 weeks. The clinical symptom score was observed before and after treatment and 4 weeks after treatment. The effects were evaluated from 3 aspects, including the main symptoms, esophagitis improvement under gastroscope and total clinical symptoms.

Results: After treatment and at follow-up, the symptom scores decreased in the two groups compared with those before treatment (all <0.05), with better improvement in the observation group at follow-up (<0.05). The symptom scores after treatment of the two types in the two groups decreased compared with those before treatment (all <0.05). The total effective rate of heartburn after treatment in the observation group was 82.1% (23/28), which was lower than 93.1% (27/29) of the control group (<0.05). The total effective rate of belching after treatment in the observation group was 94.1% (16/17), which was better than 75.0% (15/20) in the control group (<0.05). The total effective rate of the esophagitis improvement under gastroscope after treatment in the observation group was 75.0% (12/16), and that in the control group was 82.4% (14/17), without statistical significance (>0.05). The total effective rate of the clinical symptoms after treatment in the observation group was 82.1% (23/28), and that in the control group was 86.2% (25/29), without statistical significance (>0.05).

Conclusion: Acupuncture regulating based on compatibility of the five meridians can effectively improve clinical symptoms and inflammation under gastroscope for gastroesophageal reflux disease with the disharmony between liver and stomach. This treatment has a longer curative effect and a greater alleviation of belching.

Anti-HBV Activities of Three Compounds Extracted and Purified from Herpetospermum Seeds.

The goal of this research was to evaluate the anti-hepatitis B virus (HBV) activities of three compounds extracted and purified from seeds (HS) on HepG2.2.15 cells. Herpetin (HPT), herpetone (HPO), and herpetfluorenone (HPF) were isolated from HS and identified using HR-ESI-MS and NMR. Different concentrations of the drugs were added to the HepG2.2.15 cells. Cell toxicity was observed with an MTT assay, cell culture supernatants were collected, and HBsAg and HBeAg were detected by ELISA. The content of HBV DNA was determined via quantitative polymerase chain reaction (PCR) with fluorescent probes. The 50% toxicity concentration (TC) of HPF was 531.48 μg/mL, suggesting that this species is less toxic than HPT and HPO. HPT and HPF showed more potent antiviral activities than HPO. The 50% inhibition concentration (IC) values of HPF on HBsAg and HBeAg were 176.99 and 134.53 μg/mL, respectively, and the corresponding therapeutic index (TI) values were 2.66 and 3.49, respectively. HPT and HPF were shown to significantly reduce the level of HBV DNA in the HepG2.2.15 culture medium compared to the negative control. This initial investigation of the anti-HBV constituents of HS yielded three compounds that revealed a synergistic effect of multiple components in the ethnopharmacological use of HS.

Epidemiologic study of human parvovirus B19 infection in East China.

Human parvovirus B19 (B19V) infection causes a number of diseases in humans, and, in some circumstances, can be life threatening. To understand the epidemiology of B19V infection in the greater metropolitan area of Hangzhou, East China, we performed surveys of IgM and IgG antibodies against B19V and quantification of B19V DNA, by using enzyme-linked immunosorbent assay and quantitative PCR, respectively, in plasma samples from diverse groups. These groups included anemia patients, Mycoplasma pneumonia- and Treponema pallidum-infected patients, HIV-positive individuals, and healthy blood donor volunteers. Our results demonstrated a low level of B19V IgG antibody presence, ranging from 21.9% to 41.8% in all the groups tested, suggesting a low prevalence of B19V infection in the area. Of note, we found that two healthy blood donors and one Mycoplasma pneumonia-infected patient had B19V IgM antibody among 1,290 plasma samples tested. The Mycoplasma pneumonia-infected patient had viremia with viral genome copies of 2.86 × 10(6) per ml of plasma. We detected a high rate of B19V DNA (7.1%) in HIV-positive injection drug users. Importantly, an amino acid mutation of P558S in the large non-structural protein NS1 was identified to be conserved among 14 B19V isolates from the HIV-positive group but not in the B19V isolate of the Mycoplasma pneumonia-infected patient, representing a hallmark of B19V isolates that circulate in HIV1-positive patients in the greater metropolitan area of Hangzhou, East China.

The current research status of the genus Promalactis Meyrick, 1908 is introduced, and a list of 243 species of this genus are provided based on a worldwide study. Two new synonyms are proposed: P. albipars Kim et Park, 2012 syn. nov. (= P. longiuncata Wang, Kendrick et Sterling, 2009), and P. akaganea Fujisawa, 2002 syn. nov. (= P. albipunctata Park et Park, 1998); four new combinations are established: P. procerella (Denis et Schiffermüller, 1775) comb. nov., P. helminthias (Meyrick, 1918) comb. nov., P. pedicata (Meyrick, 1918) comb. nov., and P. pyrotechina (Meyrick, 1935) comb. nov.; three species are transferred from Epicallima back to Promalactis: P. atrofasciella Lvovsky, 1988 comb. rev., P. subsuzukiella Lvovsky, 1985 comb. rev. and P. nadezhdae Lvovsky, 1985 comb. rev. Maize (Zea mays L.) seedling leaf nuclear proteome and differentially expressed proteins between a hybrid and its parental lines.

To better understand the underlying molecular basis of leaf development in maize, a reference map of nuclear proteins in basal region of seedling leaf was established using a combination of 2DE and MALDI-TOF-MS. In total, 441 reproducible protein spots in nuclear proteome of maize leaf basal region were detected with silver staining in a pH range of 3-10, among which 203 spots corresponding to 163 different proteins were identified. As expected, proteins implicated in RNA and protein-associated functions were overrepresented in nuclear proteome. Remarkably, a high percentage (10%) of proteins was identified to be involved in cell division and growth. In addition, comparative nuclear proteomic analysis in leaf basal region of highly heterotic hybrid Mo17/B73 and its parental lines was also performed and 52 of 445 (11.69%) detected protein spots were differentially expressed between the hybrid and its parental lines, among which 16 protein spots displayed nonadditively expressed pattern. These results indicated that hybridization between two parental lines can cause changes in the expression of a variety of nuclear proteins, which may be responsible for the observed leaf size heterosis.

[Effects of Fukang oral liquid on the prevention of intrauterine adhesion and expressions of TGF-beta1, PAI-1 and MMP-9 in endometrium of rats].

Objective: To investigate the preventing effect of Fukang oral liquid (fuk) in intrauterine adhesions and its effects on the expression of TGFbeta-1, PAI-1 and MMP-9 in endometrium of rats with intrauterine adhesions.

Methods: 50 female wistar rats were divided into high, medium, low dose of Fukang oral liquid group (Hfuk, Mfuk, Lfuk), blank control group (Bcon), and model control group (Mcon) (n = 10 in each group). The rats in Hfuk, Mfuk and Lfuk groups were treated with intragastric administration of 4 mL, 2 mL and 1 mL Fukang Oral Liquid per day, while the rats in Mcon group and Bcon group received 2 mL physiological saline intragastric administration per day. All of rats were executed on 10th day and the sample of endometrium was harvested for the study of histology and morphology and the expression of TGFbeta-1, PAI-1 and MMP-9.

Results: Under the light microscope, the organizational structure of the uterine cavity and uterine wall was clear in Bcon group, the uterine cavity disappeared in Mcon group, and the layers structure remained normal arrangement in three fuk treated groups. TGFbeta-1 and PAI-1 protein expressions in Hfuk, Mfuk, Lfuk groups were less than those in Mcon group (P < 0.001), but MMP-9 protein expressions were higher. (P < 0.001).

Conclusion: Fukang oral liquid show preventing effect on IUA, the mechanism may be related to its effects on the expressions of TGF-beta1, PAI-1, and MMP-9 in the endometrium.

[Peripheral blood leukocyte double strand RNA-dependent protein kinase gene expression in patients with systemic lupus erythematosus].

Objective: To investigate the expression of the double-stranded RNA-dependent protein kinase (PKR) gene in the peripheral blood leukocyte of patients with systemic lupus erythematosus (SLE), and to evaluate the relationship between the gene expression and the disease activity.

Methods: The clinical data of 100 SLE patients, 40 non-SLE patients with rheumatic diseases, and 40 normal controls were collected. Total RNA was extracted from the peripheral blood and then reverse transcribed into cDNA. Sybr green dye based real-time quantitative PCR method was used to compare the expression levels (indicated as 2(-ΔCt) value) of PKR in the three groups.

Results: (1) The 2(-ΔCt) value of PKR expression level in the SLE patients was (14.69 ± 7.62), which was significantly higher than those in the non-SLE patients (5.09 ± 4.73, P = 0.012)and normal controls(4.79 ± 3.49, P = 0.005). (2) The 2(-ΔCt) value of PKR expression level in the SLE patients with severe activity was (22.57 ± 2.61), which was significantly higher than those in the SLE patients with mild activity and no activity (12.94 ± 2.41, P = 0.000; 8.85 ± 2.17, P = 0.000). (3) The 2(-ΔCt) value of PKR expression level in the SLE patients with lupus nephritis was significantly higher than that in the SLE patients without lupus nephritis (16.85 ± 7.32 vs 8.35 ± 2.04, P = 0.034). (4) The 2(-ΔCt) value of PKR was correlated with the systemic lupus erythematosus index (SLEDAI) scores (r = 0.32, P = 0.000), WBC (r = 0.46, P = 0.000), Hb (r = -0.22, P = 0.035), the quantitation of urine protein in 24 hours (r = 0.21, P = 0.000), HDL-C (r = 0.21, P = 0.022), and anti-RNP antibody (r(s) = -0.21, P = 0.025).

Conclusions: The expression of PKR in the SLE patients is up-regulated, especially in those with severe activity. The expression level of PKR gene is associated with SLE disease activity.

A survey on gas sensing technology.

Sensing technology has been widely investigated and utilized for gas detection. Due to the different applicability and inherent limitations of different gas sensing technologies, researchers have been working on different scenarios with enhanced gas sensor calibration. This paper reviews the descriptions, evaluation, comparison and recent developments in existing gas sensing technologies. A classification of sensing technologies is given, based on the variation of electrical and other properties. Detailed introduction to sensing methods based on electrical variation is discussed through further classification according to sensing materials, including metal oxide semiconductors, polymers, carbon nanotubes, and moisture absorbing materials. Methods based on other kinds of variations such as optical, calorimetric, acoustic and gas-chromatographic, are presented in a general way. Several suggestions related to future development are also discussed. Furthermore, this paper focuses on sensitivity and selectivity for performance indicators to compare different sensing technologies, analyzes the factors that influence these two indicators, and lists several corresponding improved approaches.
