Introduction

During my thesis in Bordeaux (France), I studied the role of apoptosis in the immune system. More specifically, I worked on a TNF receptor member designated CD95 (also known as Fas) and its role as a potent modulator of the immune surveillance and immune tolerance 1-4. Then I joined Marcus Peter’s laboratory at University of Chicago (2002-2005, USA) where I showed that in cancer cells, CD95 was able to implement non-apoptotic signalling pathways promoting carcinogenesis5-7. Recruited as a INSERM researcher (full-time academic position) in 2005; I ran my own team and as Principal Investigator in University of Bordeaux (2005-2010, France), I demonstrated that anti-tumour agents induced rapid redistribution of CD95 into lipid rafts to facilitate the elimination of malignant cells 8,9.
Since 2010, (University of Rennes, Rennes, France) I joined the Cancer Center Eugène Marquis in Rennes (France) and established that the metalloprotease-driven cleavage of CD95L (also designated FasL) released a soluble CD95L (s-CD95L), which participated in i) the severity of a chronic inflammatory disorder designated systemic lupus erythematosus (SLE) by promoting Th17 trafficking in the inflamed organs 10,11 and ii) the metastatic dissemination of triple negative breast cancer cells 12. In addition, my Team demonstrated that in the presence of s-CD95L, CD95 elicits the formation of a molecular complex that he termed MISC, for Motility-Inducing Signalling Complex, thereby inducing a non-orthodox c-yes/Orai1/Ca2+/PI3K signalling pathway 10,12,13. Currently, my group is interested to understand how the “apoptotic receptor” CD95 participates in the aggravation of the clinical outcomes in chronic inflammatory disorders (lupus) and breast cancers.
