The role of enteric inhibitory neurons in intestinal motility.

The enteric nervous system controls much of the mixing and propulsion of nutrients along the digestive tract. Enteric neural circuits involve intrinsic sensory neurons, interneurons and motor neurons. While the role of the excitatory motor neurons is well established, the role of the enteric inhibitory motor neurons (IMNs) is less clear. The discovery of inhibitory transmission in the intestine in the 1960 in the laboratory of Geoff Burnstock triggered the search for the unknown neurotransmitter. It has since emerged that most neurons including the IMNs contain and may utilise more than one transmitter substances; for IMNs these include ATP, the neuropeptide VIP/PACAP and nitric oxide. This review distinguishes the enteric neural pathways underlying the 'standing reflexes' from the pathways operating physiologically during propulsive and non-propulsive movements. Morphological evidence in small laboratory animals indicates that the IMNs are located in the myenteric plexus and project aborally to the circular muscle, where they act by relaxing the muscle. There is ongoing 'tonic 'activity of these IMNs to keep the intestinal muscle relaxed. Accommodatory responses to content further activate enteric pathways that involve the IMNs as final neural element. IMNs are activated by mechanical and chemical stimulation induced by luminal contents, which activate intrinsic sensory enteric neurons and the polarised interneuronal ascending excitatory and descending inhibitory reflex pathways. The latter relaxes the muscle ahead of the advancing bolus, thus facilitating propulsion.

Blooming artifact reduction using different cone-beam computed tomography software to analyze endodontically treated teeth with intracanal posts.

Background: The appearance of high-density root obturating materials, such as gutta-percha and intracanal posts, on cone-beam computed tomography (CBCT) scans is substantially different from that seen in the specimen. This study evaluated the effectiveness of different CBCT software in the reduction of blooming artifacts when examining teeth with intracanal posts.

Method: The sample included 42 human single-rooted teeth with two types of posts: G1 - low-fusion alloy posts; G2 - gold alloy posts. CBCT scans were obtained using two scanners, PreXion 3D Elite® and Carestream 9000C 3D®. First, the posts were measured using a digital micrometer (standard reference, control). Post diameters were determined using the Blooming Artifact Reduction (BAR) tool of the e-Vol DX software and of the PreXion3D Image Analysis System. Statistical data were evaluated using the van Der Waerden nonparametric analysis of variance and, after that, normalized data were analyzed using the Tukey test. The level of significance was set at α = 5 %.

Results: There were no significant differences (p > 0.05) between the diameters of intracanal posts on the CT scans when the e-Vol DX was used, or when these results were compared with those obtained using a micrometer. There were significant differences in post diameters determined using the PreXion3D Image Analysis System when compared with the e-Vol DX and the micrometer values (p < 0.05).

Conclusions: The use of the e-Vol DX BAR filter eliminated blooming artifacts. There were no dimensional changes in the CBCT images of the low-fusion and gold alloy intracanal posts using the e-Vol DX BAR filter.

Leishmanicidal and cytotoxic activities and 4D-QSAR of 2-arylidene indan-1,3-diones.

The indan-1,3-dione and its derivatives are important building blocks in organic synthesis and present important biological activities. Herein, the leishmanicidal and cytotoxicity evaluation of 16 2-arylidene indan-1,3-diones is described. The compounds were evaluated against the leukemia cell lines HL60 and Nalm6, and the most effective ones were 2-(4-nitrobenzylidene)-1H-indene-1,3(2H)-dione (4) and 4-[(1,3-dioxo-1H-inden-2(3H)-ylidene)methyl]benzonitrile (10), presenting IC values of around 30 µmol/L against Nalm6. The leishmanicidal activity was assessed on Leishmania amazonensis, with derivative 4 (IC = 16.6 µmol/L) being the most active. A four-dimensional quantitative structure-activity analysis (4D-QSAR) was applied to the indandione derivatives, through partial least-squares regression. The statistics presented by the regression models built with the selected field descriptors of Coulomb (C) and Lennard-Jones (L) nature, considering the activities against L. amazonensis, HL60, and Nalm6 leukemia cells, were, respectively, R = 0.88, 0.92, and 0.98; Q = 0.83, 0.88, and 0.97. The presence of positive Coulomb descriptors near the carbonyl groups indicates that these polar groups are related to the activities. Besides, the presence of positive Lennard-Jones descriptors close to substituents R or R indicates that bulky nonpolar substituents in these positions tend to increase the activities. This study provides useful insights into the mode of action of indandione derivatives for each biological activity involved.

The relationship between vocal fatigue and voice-related quality of life in university professors.

Purpose: To identify and correlate vocal fatigue and voice-related quality of life in university professors and verify possible differences between genders.

Methods: This is a cross-sectional, observational, and analytical study approved by the Research Ethics Committee of the institution under number 1,708,786. The Voice Fatigue Index (VFI) and the Voice Related Quality of Life (VRQOL) were applied to 126 university professors, 71 women and 55 men, with an average age of 43 years. Scores were calculated using the formula for each protocol and statistical analysis was performed using Spearman's Correlation.

Results: Regarding the VFI, the average score found for factors 1, 2 and 3 were 13.78, 4.05 and 7.93, respectively. As for VRQOL, professors had an average global score of 91.90; 88.49 for the physical domain and 97.02 for the socioemotional domain. There was a negative weak to strong correlation between the protocols. There was no statistical difference between genders for both VFI and VRQOL.

Conclusion: University professors have good levels of voice related quality of life, but self-reported vocal fatigue, with no differences between genders. The lower the vocal fatigue is, the higher is the voice related quality of life, and vice versa.

Relationship between Innate Immune Response Toll-Like Receptor 4 (TLR-4) and the Pathophysiological Process of Obesity Cardiomyopathy.

Background: Obesity is a chronic low-grade inflammation condition related to cardiac disorders. However, the mechanism responsible for obesity-related cardiac inflammation is unclear. The toll-like receptor 4 (TLR-4) belongs to a receptor of the transmembrane family responsible for the immune response whose activation stimulates the production of proinflammatory cytokines.

Objective: To test whether the activation of the TLR-4 receptor participates in the obesity cardiomyopathy process, due to cytokine production through NF-ĸB activation.

Methods: Male Wistar rats were randomized into two groups: the control group (C, n= 8 animals) that received standard diet/water and the obese group (OB, n= 8 animals) that were fed a high sugar-fat diet and water plus 25% of sucrose for 30 weeks. Nutritional analysis: body weight, adiposity index, food, water, and caloric intake. Obesity-related disorders analysis: plasma glucose, uric acid and triglycerides, HOMA-IR, systolic blood pressure, TNF-α in adipose tissue. Cardiac analysis included: TLR-4 and NF-ĸB protein expression, TNF-α and IL-6 levels. Comparison by unpaired Student's t-test or Mann- Whitney test with a p-value < 0.05 as statistically significant.

Results: The OB group showed obesity, high glucose, triglycerides, uric acid, HOMA, systolic blood pressure, and TNF-α in adipose tissue. OB group presented cardiac remodeling and diastolic dysfunction. TLR-4 and NF-ĸB expression and cytokine levels were higher in OB.

Conclusion: Our findings conclude that, in an obesogenic condition, the inflammation derived from cardiac TLR-4 activation can be a mechanism able to lead to remodeling and cardiac dysfunction.
