Introduction

Professor Kutty Selva Nandakumar (h-index: 38) is an international scientist and has 30 years of research experience. He is having his new research team in the school of Pharmaceutical Sciences, Southern Medical University (SMU), and Guangzhou, China to work in inflammation and immunology research fields. He is also docent in Medical Inflammation Science, Karolinska Institute, Stockholm, Sweden for the period 2011-2023. He has more than 130 publications in international journals. He has research experience in the fields of autoimmune diseases, tumour immunology and infectious diseases. He was an adjunct faculty in the University of Arkansas for Medical Sciences (UAMS), USA for the period 2013-2016. His research experience includes several disease models to understand the pathogenic mechanisms and genetic basis of diseases. He also worked on characterising stimuli-responsive polymers in an international collaborative program with IIT-Kanpur, India. He is editorial board member and adhoc reviewer for several international journals. In addition, he evaluates project grants for research foundations in UK, Sweden, The Netherlands and EU, and doctoral theses for universities in India, Spain and Sweden. Previously he completed two independent doctorates in Madurai Kamaraj University, India (1996) and Lund University, Sweden (2006).

Autophagy induced by STING, an unnoticed and primordial function of cGAS.

Cell Mol Immunol.

Cyclic GMP-AMP synthase (cGAS), combined with dsDNA, produces cyclic GMP-AMP (cGAMP) to stimulate STING and activates the kinases TBK1 and IKK to induce interferon and other cytokines, which play an important role in host defense. In addition to the traditional interferon inflammatory signaling pathway of cGAS, recently, Chen et al.1discovered a new mechanism in which cGAS-cGAMP-STING activated autophagy through WIPI2 and ATG5. Autophagy is a process induced under various cellular stress conditions, such as oxygen deficit, nutrient deprivation, lack of growth factors, and infection, aiming to maintain the basic survival of cells or promote apoptosis of senescent cells.2,3The activation of the autophagy-related (ATG)1/Unc-51-like kinase (ULK) complex is vital for autophagy induction. The ATG1/ULK1 complex phosphorylates or recruits its downstream proteins, contributing to autophagosome formation.3 Under the regulation of related genes, “cargo” is transferred to the lysosome and digested by various enzymes. In addition, cyclic GMP-AMP synthase (cGAS), which detects exogenous or self-damaged dsDNA in the cytoplasm after binding to dsDNA, produces cGAMP to activate the adaptor protein stimulator of interferon genes (STING), triggering the downstream pathway and secretion of interferon or other cytokines.4
