Evaluation of the acute toxic effects of crude oil on intertidal mudskipper (Boleophthalmus pectinirostris) based on antioxidant enzyme activity and the integrated biomarker response.

With the development of marine oil industry, oil spill accidents will inevitably occur, further polluting the intertidal zone and causing biological poisoning. The muddy intertidal zone and Boleophthalmus pectinirostris were selected as the research objects to conduct indoor acute exposure experiments within 48 h of crude oil pollution. Statistical analysis was used to reveal the activity changes of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST) in the gills and liver of mudskipper. Then, integrated biomarker response (IBR) indicators were established to comprehensively evaluate the biological toxicity. The results showed that the activities of SOD, CAT and GST in livers were higher than those in gills, and the maximum induction multipliers of SOD, CAT and GPx in livers appeared earlier than those in gills. Both SOD and GPx activities were induced at low pollutant concentrations and inhibited at high pollutant concentrations. For the dose-effect, the change trends of CAT and SOD were roughly inversed. There was substrate competition between GPx and CAT, with opposite trends over time. The activating mechanism of GST was similar to that of GPx, and the activation time was earlier than that of GPx. In terms of dose-effect trends, the IBR showed that the antioxidant enzymes activities in biological tissues were induced by low and inhibited by high pollutant concentrations. Overall, SOD and GPx in gills and CAT and GST in livers of the mudskippers were suitable as representative markers to comprehensively analyze and evaluate the biotoxicity effects of oil pollution in the intertidal zone. The star plots and IBR values obtained after data standardization were consistent with the enzyme activity differences, which can be used as valid supplementary indexes for biotoxicity evaluation. These research findings provide theoretical support for early indicators of biological toxicity after crude oil pollution in intertidal zones.

Difference in temporal and spatial distribution pattern of cyanobacteria between the sediment and water column in Lake Chaohu.

The evaluation about the relative distribution of cyanobacterial biomass between the sediment and water column would be indispensable to understand if benthic cyanobacteria are important to cyanobacterial biomass in the water column. A separation method for the rapid quantification of benthic cyanobacteria in Lake Chaohu was developed by density-gradient centrifugation. A 2 full factorial design and response surface methodology was employed to optimize the extraction protocol. Under the optimal operating parameters including 29% Percoll solution, 30 min centrifugation time, 7200 r/min centrifugation speed, and a 1:10 ratio between the volume of sediment and Percoll solution, the recovery rate of cyanobacteria in sediment was 96.73%. Temporal and spatial variations in cyanobacterial biomass in water and sediment were investigated monthly throughout a whole year. In general, cyanobacterial biomass per square centimeter in the water column showed high spatial-temporal changes, tending to increase in February and reaching a peak in April at some sites due to the growth of Dolichospermum. The second peak arrived in July and September and was caused by the rapid growth of Microcystis. Concurrently, cyanobacteria biomass per unit area in sediment showed a clear temporal change pattern, increasing from October and reaching a peak level in February at all the sampling sites. The average ratio of cyanobacterial biomass in water to that in sediment was lowest in January at 1.48 and increased to the highest level in July at 318.61. Although Microcystis and Dolichospermum were dominant species in the water column, only Microcystis was observed in the Percoll solution extraction from sediment. Microscopic observation revealed that a very small fraction of Microcystis cells could survive in sediment, and most of the cells decomposed when the water temperature increased after June. Therefore, the contribution of the recruitment of cyanobacteria could be negligible in Lake Chaohu.

The spatial distribution of the photosynthetic picoeukaryotes community structure in Lake Hongze.

The spatial dynamics of picophytoplankton were investigated by flow cytometry and high-throughput sequencing in Lake Hongze, a large river-connecting lake. Picophytoplankton were mainly composed of phycocyanin-rich picocyanobacteria (PCY) and photosynthetic picoeukaryotes (PPEs). Picocyanobacteria was the dominant picophytoplankton group since the contribution of PPEs to total picophytoplankton was only 11.78%. However, PPEs were highly diverse and were composed of Chlorophyta, Bacillariophyta and Chrysophyceae. Environmental factors showed spatial differences, particularly in total phosphorus (TP), suspended solids (SS) and chemical oxygen demand (CODMn), which showed relatively high concentrations around the river channel. The abundances of PPEs and PCY showed similar spatial patterns, which were relatively low in the river course since they were negatively related to SS and CODMn. The top 10 OTUs contributed 79.18% of the total PPEs reads and affiliated with main species in PPEs. CCA results showed that, PPEs community composition was significantly impacted by temperature and DO at sites away from river channel, while was only influenced by nitrite at sites around the river channel.

The daily effect is more important than the diurnal effect when shaping photosynthetic picoeukaryotes (PPEs) communities in Lake Taihu at a small temporal scale.

As major primary producers in aquatic systems, the diversity and community composition dynamics of photosynthetic picoeukaryotes (PPEs) have been investigated in recent years. Here, we explored the 3-day diurnal succession of the PPE community in a highly eutrophic lake in early spring using a combination of flow cytometric sorting and high-throughput sequencing. Our results showed that the PPEs were the dominant group of autotrophic picophytoplankton, although they had relatively low diversity and were dominated by the Stephanodiscaceae family in early spring in Lake Taihu. Furthermore, PPE abundance was significantly higher at night than during the day, probably due to their high specific rate of carbon uptake during the day and rapid proliferation at night. Several environmental variables had a significant influence on the PPE community composition, and the daily effect was more important than the diurnal effect when shaping the PPE community in Lake Taihu at a temporal scale of 3 days. Furthermore, based on the variation partitioning analysis (VPA), the relative importance of abiotic factors (deterministic processes) to short-term succession was low, explaining only 20.44% of the PPE community variation. We therefore conclude that stochastic processes determined PPE community assembly over a short temporal scale based on a neutral community model (NCM).

Nidogen-1 expression is associated with overall survival and temozolomide sensitivity in low-grade glioma patients.

We investigated the prognostic significance of nidogen-1 (NID1) in glioma. Oncomine, GEPIA, UALCAN, CCGA database analyses showed that NID1 transcript levels were significantly upregulated in multiple cancer types, including gliomas. Quantitative RT-PCR analyses confirmed that NID1 expression was significantly upregulated in glioma tissues compared to paired adjacent normal brain tissue samples (n=9). NID1 silencing enhanced apoptosis and the temozolomide sensitivity of U251 and U87-MG glioma cells. Protein-protein interaction network analysis using the STRING and GeneMANIA databases showed that NID1 interacts with several extracellular matrix proteins. TIMER database analysis showed that NID1 expression in low-grade gliomas was associated with tumor infiltration of B cells, CD4 and CD8 T cells, macrophages, neutrophils, and dendritic cells. Kaplan-Meier survival curve analysis showed that low-grade gliomas patients with high NID1 expression were associated with shorter overall survival. However, NID1 expression was not associated with overall survival in glioblastoma multiforme patients. These findings demonstrate that NID1 expression in glioma tissues is associated with overall survival of low-grade glioma patients and temozolomide sensitivity. NID1 is thus a potential prognostic biomarker and therapeutic target in low-grade glioma patients.

Simulation of Nanopore Sequencing Signals Based on BiGRU.

Oxford Nanopore sequencing is an important sequencing technology, which reads the nucleotide sequence by detecting the electrical current signal changes when DNA molecule is forced to pass through a biological nanopore. The research on signal simulation of nanopore sequencing is highly desirable for method developments of nanopore sequencing applications. To improve the simulation accuracy, we propose a novel signal simulation method based on Bi-directional Gated Recurrent Units (BiGRU). In this method, the signal processing model based on BiGRU is built to replace the traditional low-pass filter to post-process the ground-truth signal calculated by the input nucleotide sequence and nanopore sequencing pore model. Gaussian noise is then added to the filtered signal to generate the final simulated signal. This method can accurately model the relation between ground-truth signal and real-world sequencing signal through experimental sequencing data. The simulation results reveal that the proposed method utilizing the powerful learning ability of the neural network can generate the simulated signal that is closer to the real-world sequencing signal in the time and frequency domains than the existing simulation method.

Identification of a primary antigenic target of epitope spreading in endemic pemphigus foliaceus.

Epitope spreading is an important mechanism for the development of autoantibodies (autoAbs) in autoimmune diseases. The study of epitope spreading in human autoimmune diseases is limited due to the major challenge of identifying the initial/primary target epitopes on autoantigens in autoimmune diseases. We have been studying the development of autoAbs in an endemic human autoimmune disease, Brazilian pemphigus foliaceus (or Fogo Selvagem (FS)). Our previous findings demonstrated that patients before (i.e. preclinical) and at the onset of FS have antibody (Ab) responses against other keratinocyte adhesion molecules in addition to the main target autoantigen of FS, desmoglein 1 (Dsg1), and anti-Dsg1 monoclonal Abs (mAbs) cross-reacted with an environmental antigen LJM11, a sand fly saliva protein. Since sand fly is prevalent in FS endemic regions, individuals in these regions could develop Abs against LJM11. The anti-LJM11 Abs could recognize different epitopes on LJM11, including an epitope that shares the structure similarity with an epitope on Dsg1 autoantigen. Thus, Ab response against this epitope on LJM11 could be the initial autoAb response detected in individuals in FS endemic regions, including those who eventually developed FS. Accordingly, this LJM11 and Dsg1 cross-reactive epitope on Dsg1 could be the primary target of the autoimmune response in FS. This investigation aimed to determine whether the autoAb responses against keratinocyte adhesion molecules are linked and originate from the immune response to LJM11. The anti-Dsg1 mAbs from preclinical FS and FS individuals were employed to determine their specificity or cross-reactivity to LJM11 and keratinocyte adhesion molecules. The cross-reactive epitopes on autoantigens were mapped. Our results indicate that all tested mAbs cross-reacted with LJM11 and keratinocyte adhesion molecules, and we identified an epitope on these keratinocyte adhesion molecules which is mimicked by LJM11. Thus, the cross-reactivity could be the mechanism by which the immune response against an environmental antigen triggers the initial autoAb responses. Epitope spreading leads to the pathogenic autoAb development and ensuing FS among genetically susceptible individuals.

Three-dimensional migration and resistivity characteristics of crude oil in heterogeneous soil layers.

An experimentally induced three-dimensional petroleum seepage flume was used to investigate its migration in heterogeneous soil layers and a method for monitoring resistivity was adopted, under conditions of fluctuating water levels and rainfall. The corresponding mechanisms were then analyzed based on the resistivity characteristics and combined with three-dimensional inversion images. Finally, physical and chemical property analysis was conducted to verify the results of resistivity monitoring. The results demonstrated that: (1) In the process of natural oil leakage, the variation of soil resistivity presents a concave shape in the resistivity profile. Thus, oil migration exhibited the following patterns. At first, circular migration front was dominant in a vertical direction. Subsequently, after vertical migration was impeded, lateral migration was dominant. As the crude oil gradually accumulated, the migration front broke through the limitation of lithologic interface and continued vertically. (2) By comparing the two resistivity monitoring methods, namely the Wenner and Pole-pole methods, it was demonstrated that the inversion resistivity measured by Wenner method was closer to the true resistivity, and the resistivity variations were more distinguishable. (3) The resistivity inversion profile demonstrated that the low resistivity anomaly of the crude oil leakage area was related to the low water content of the soil layer in the test area. (4) Fluctuations in water level increased the diffusion range of crude oil beyond the original pollution source area, especially horizontally. (5) Percolation of rainfall caused the water level to rise, and the crude oil was evenly distributed in the soil layers above the capillary zone. (6) Through sample analysis and verification, it was demonstrated that the resistivity method can accurately and intuitively present the characteristics of crude oil migration. These results provide theoretical support for the rapid determination of the migration range and characteristics of crude oil in heterogeneous soil layers.

Macrodiversity Reception with Distributed Hard-Decision Receivers for Maritime Wireless Sensor Networks.

Maritime wireless sensor networks are considered to be the primary means of monitoring methods in the marine environment. The transmission between sensor node and sink node in maritime wireless sensor networks is usually unreliable due to the harsh propagation environment. To extend the transmission range or to enhance the transmission reliability between sensor nodes and sink node, we propose a macrodiversity reception scheme in the sink node equipped with distributed multiple hard-decision receivers. Multiple receivers are divided into several clusters and placed at different locations to receive different signal copies suffering from different fadings. Furthermore, a cascaded combining strategy based on hard-decision information is used to reduce the overall complexity of receiving side. The experimental results in the ocean scenarios show that the macrodiversity reception scheme with two antenna clusters has a transmission gain of 3-4 dB compared with the single antenna reception when the package loss rate is 10 - 2 . The study casts a new method for reliable transmission in maritime wireless sensor networks using commercial transceivers which can only output hard-decision results.

Effects of Gua Sha therapy on weightlifting training: a randomized trial.

Objective: To evaluate the effect of Gua Sha therapy on weightlifting training.

Methods: The sample size was calculated by conducting a pilot study. A total of 44 male weightlifters were randomly assigned to either the Gua Sha group (n = 15), sham scraping group (n = 14), or control group (n = 14). The participants in the Gua Sha group and sham scraping group received 16 sessions of Gua Sha therapy during normal weightlifting training for 8 weeks. No treatment was applied to participants in the control group. The effectiveness of Gua Sha therapy was evaluated by measuring weightlifting ability, the rating of perceived exertion (RPE) of snatch and clean and jerk (85% of 1 repetition maximum), and creatinine kinase (CK), blood urea nitrogen (BUN), and immunoglobulin A levels.

Results: Gua Sha therapy treatment significantly increased weightlifting ability in participants in the Gua Sha group (P < 0.01). The RPE values of snatch and clean and jerk were significantly lower in the Gua Sha group and sham scraping groups compared with the control group (P < 0.05). CK levels were lower and immunoglobulin A levels were significantly higher in the Gua Sha group compared with sham scraping group and control group (both P < 0.05). BUN levels tended to be reduced only in the Gua Sha group. A close correlation between CK levels and the RPE was found in the Gua Sha group.

Conclusion: Gua Sha therapy can facilitate weightlifting ability, reduce the RPE, and inhibit muscle injury by promoting recovery from fatigue caused by normal weightlifting training. Gua Sha therapy could be an effective treatment to complement normal weightlifting training.

Bimorph deformable mirror-based adaptive optics scanning laser ophthalmoscope for the clinical design and performance.

We developed a clinical ophthalmic prototype by combining bimorph deformable mirror (DM)-based adaptive optics (AO) with a confocal scanning laser ophthalmoscope. A low-cost bimorph DM with a large stroke of and an aperture of 20 mm was utilized to realize a strategy for successive AO control of aberration correction, which permitted open-loop compensation for low-order aberrations and closed-loop correction of high-order aberrations to acceptable root mean square errors of in all subjects. Spherical mirrors were folded in a nonplanar configuration to minimize off-axis aberrations and provide a compact, cost-effective design, which achieved a diffraction-limited performance capable of imaging individual photoreceptor cells and blood vessels not only in healthy subjects but also in patients suffering from retinitis pigmentosa. The adaptive optics scanning laser ophthalmoscope (AOSLO) images of the diseased retina had much higher resolutions than those captured by the commercial AO fundus camera, and loss of the photoreceptor mosaic could be distinguished more accurately due to the improvement in resolution. The compact design and easy handling of the bimorph DM-based AO control may facilitate the translation of AOSLO into clinical settings, and this prototype development will continue with future device refinement and extensive clinical testing.

Polynomial transformation model for frame-to-frame registration in an adaptive optics confocal scanning laser ophthalmoscope.

The adaptive optics (AO) technique has been integrated in confocal scanning laser ophthalmoscopy (SLO) to obtain near diffraction-limited high-resolution retinal images. However, the quality of AOSLO images is decreased by various sources of noise and fixational eye movements. To improve image quality and remove distortions in AOSLO images, the multi-frame averaging method is usually utilized, which relies on an accurate image registration. The goal of image registrations is finding the optimal transformation to best align the input image sequences. However, current methods for AOSLO image registration have some obvious defects due to the limitation of transformation models. In this paper, we first established the retina motion model by using the Taylor series and polynomial expansion. Then we generated the polynomial transformation model and provided its close-form solution for consecutively frame-to-frame AOSLO retina image registration, allowing one to consider more general retinal motions such as scale changes, shearing and rotation motions, and so on. The experimental results demonstrated that higher-order polynomial transformation models are helpful to achieve more accurate registration, and the fourth-order polynomial transformation model is preferred to accomplish an efficient registration with a satisfying computational complexity. In addition, the AKAZE feature detection method was adopted and improved to achieve more accurate image registrations, and a new strategy was validated to exclude those unsuccessful registered regions to promote the robustness of image registration.

Persistent Nociception Facilitates the Extinction of Morphine-Induced Conditioned Place Preference.

Background: As opioid abuse and addiction have developed into a major national health crisis, prescription of opioids for pain management has become more controversial. However, opioids do help some patients by providing pain relief and improving the quality of life. To better understand the addictive properties of opioids under chronic pain conditions, we used a conditioned place preference (CPP) paradigm to examine the rewarding properties of morphine in rats with persistent nociception.

Methods: Spared nerve injury (SNI) model was used to induce persistent nociception in rats. Nociceptive behavior was assessed by von Frey test. CPP test was used to examine the rewarding properties of morphine.

Results: Our findings are as follows: (1) SNI rats did not show a difference compared with sham rats in magnitude of morphine-induced CPP 1 day after last morphine injection (2-way analysis of variance; for SNI versus sham, F[1,42] = 0.014, P = .91; and 95% confidence intervals for difference of means, -5.9 [-58 to 46], 0.76 [-51 to 53], and 0.90 [-51 to 53] for 2.5, 5, and 10 mg/kg, respectively); (2) increasing morphine dosage (2.5, 5, and 10 mg/kg) did not further increase the magnitude of CPP in both sham and SNI rats (for dosage: F[2,42] = 0.94, P = .40); and (3) morphine-induced CPP persisted in sham rats but extinguished in SNI rats when tested at 8 days after last morphine injection (for sham versus SNI: Bonferroni correction, P < .006 for both 5 and 10 mg/kg doses; and 95% confidence intervals for difference of means, 80.3 [19.7-141] and 87.0 [26.3-148] for 5 and 10 mg/kg, respectively).

Conclusions: Our data provide new evidence supporting the notion that the brain's reward circuitry changes in the context of persistent pain. This observational study suggests that future investigation into the neurobiology of opioid reward requires consideration of the circumstances in which opioid analgesics are administered.

Methylphenidate and Morphine Combination Therapy in a Rat Model of Chronic Pain.

Background: The incremental dose of opioids used in chronic pain management often leads to a reduced opioid analgesic effect, opioid misuse, and addiction. Central dopamine (DA) dysfunction contributes to the chronicity of pain and a decreased opioid analgesic effect. Methylphenidate (MPH/Ritalin) enhances central DA function by inhibiting DA reuptake. In this study, we used a rat model of chronic pain to examine whether combination of MPH with morphine (MOR) would improve the MOR analgesic effect under a chronic pain condition.

Methods: Tibiotarsal joint Complete Freund's Adjuvant (CFA) injection in rats was utilized to induce chronic nociception. The analgesic effect of low-dose MPH (0.25 mg/kg), low-dose MOR (2.5 mg/kg), and their combination was examined in CFA rats. Nociceptive behavior was assessed by von Frey test. Conditioned place preference (CPP) and open field tests (OFTs) were used to examine the rewarding behavior and locomotor activity in rats, respectively.

Results: Our findings are as follows: (1) in CFA rats with chronic pain, 2.5 mg/kg of MOR had less analgesic effect than 10 mg/kg of MOR at 28 days after injury (95% confidence intervals [CIs] for difference of means of von Frey threshold in gram: -11.9 [-6.5 to -17.3]); (2) in the 1-hour time window of 30-90 minutes after injection, the combination of MPH (0.25 mg/kg) with MOR (2.5 mg/kg) increased synergistically and prolonged the analgesic effect in CFA rats as compared with MPH or MOR alone (P = .01 for MPH by MOR interaction, and 95% CIs for difference of means of von Frey threshold in gram: 3.3 [1.37-6.12] for the combination versus MPH and 3.2 [1.35-5.74] for the combination versus MOR); (3) at the low dose (0.25 mg/kg), MPH did not increase locomotor activity (MOR + MPH versus MOR, P = .13) nor significantly enhanced MOR reward behavior (MOR + MPH versus MOR, P = .63) in CFA rats.

Conclusions: Our data suggest that a combination therapy using low-dose MPH and MOR may produce a MOR-sparing effect in chronic pain management.

Walnut antigens can trigger autoantibody development in patients with pemphigus vulgaris through a "hit-and-run" mechanism.

Background: Environmental factors, as well as genetic predisposition, are known to be critical for the development of autoimmunity. However, the environmental agents that trigger autoimmune responses have remained elusive. One possible explanation is the "hit-and-run" mechanism in which the inciting antigens that initiate autoimmune responses are not present at the time of overt autoimmune disease.

Objective: After our previous findings that some allergens can incite autoimmune responses, we investigated the potential role of environmental allergens in triggering autoantibody development in patients with an autoimmune skin disease, pemphigus vulgaris (PV).

Methods: Revertant/germline mAbs (with mutations on variable regions of heavy and light chains reverted to germline forms) of 8 anti-desmoglein (Dsg) 3 pathogenic mAbs from patients with PV were tested for reactivity against a panel of possible allergens, including insects, pollens, epithelia, fungi, and food antigens.

Results: All the PV germline mAbs were reactive to antigens from walnut, including the well-known allergen Jug r 2 and an uncharacterized 85-kDa protein component. Sera from patients with PV contained significantly greater levels of anti-Dsg3 autoantibodies than walnut-specific antibodies, suggesting that the autoreactive B-cell response in patients with PV might be initially triggered by walnut antigens but is subsequently driven by Dsg3.

Conclusion: Our findings suggest that walnut antigens/allergens can initiate autoantibody development in patients with PV through a "hit-and-run" mechanism. The revertant/germline mAb approach might provide a paradigm for the etiological study of other allergic and autoimmune diseases.

Alleviation of trigeminal neuropathic pain by electroacupuncture: the role of hyperpolarization-activated cyclic nucleotide-gated channel protein expression in the Gasserian ganglion.

Introduction: The aim of this study was to examine the effect of electroacupuncture (EA) on trigeminal neuropathic pain in rats and explore the potential mechanism underlying the putative therapeutic effect of EA.

Methods: Trigeminal neuropathic pain behavior was induced in rats by unilateral chronic constriction injury of the distal infraorbital nerve (dIoN-CCI). EA was administered at ST2 () and . A total of 60 Sprague Dawley rats were divided into the following four groups ( = 15 per group) to examine the behavioral outcomes after surgery and/or EA treatment: sham (no ligation); dIoN-CCI (received isoflurane only, without EA treatment); dIoN-CCI+EA-7d (received EA treatment for 7 days); and dIoN-CCI+EA-14d (received EA treatment for 14 days). Both evoked and spontaneous nociceptive behaviors were measured. Of these, 12 rats ( = 4 from sham, dIoN-CCI, and dIoN-CCI+EA-14d groups, respectively) were used to analyze protein expression of hyperpolarization-activated cyclic nucleotide-gated (HCN) channel in the Gasserian ganglion (GG) by immunohistochemistry.

Results: dIoN-CCI rats exhibited mechanical allodynia and increased face-grooming activity that lasted at least 35 days. EA treatment reduced mechanical allodynia and face-grooming in dIoN-CCI rats. Overall, 14 days of EA treatment produced a prolonged anti-nociceptive effect as compared to 7-day EA treatment. The counts of HCN1 and HCN2 immunopositive puncta were increased in the ipsilateral GG in dIoN-CCI rats and were reduced by 14 days of EA treatment.

Discussion: EA treatment relieved trigeminal neuropathic pain in dIoN-CCI rats, and this effect was dependent on the duration of EA treatment. The downregulation of HCN expression may contribute to the anti-nociceptive effect of EA in this rat model of trigeminal neuropathic pain.

Effects of Guasha on histomorphology of scraped skins and on expression of calcitonin gene-related peptide and substance P in rats.

Objective: To reveal the effects of Guasha (scraping therapy) on the histomorphology of scraped skins and on the expressions of calcitonin gene-related peptide (CGRP) and substance P (SP).

Methods: 50 rats, as experimental subjects, were randomly divided into 5 groups according to different observation time points. Dorsal setae were shaved for the exposure of skin on both sides of the spine. Even reinforcing and reducing method was applied to the rats in Guasha groups on the site equivalent to bladder meridian of human body on one side of the spine, and the skin was scraped from top to bottom until rash of measles occurred. The skin tissues with rash of measles were taken down after perfusion. The corresponding sites of rats in group A were also taken down as control. The tissues were made to sections and used for immunofluorescence histochemical staining and HE staining of antibodies such as SP and CGRP.

Results: After Guasha, there were significant differences in appearance, hair follicle and blood vessel on local skin scraped on the back of rats when compared with control; In different time points, the differences reduced. There was no significant difference in expression of CGRP and SP when compared local skin scraped on the back of rats in different time points.

Conclusion: Guasha didn't significantly change the morphology of nerve fibers inside local skin, and the histomorphology of hair follicle, blood vessel and etc. However, after Guasha they basically returned to normal level within five days.

Cognitive impairment in a rat model of neuropathic pain: role of hippocampal microtubule stability.

Clinical evidence indicates that cognitive impairment is a common comorbid condition of chronic pain. However, the cellular basis for chronic pain-mediated cognitive impairment remains unclear. We report here that rats exhibited memory deficits after spared nerve injury (SNI). We found that levels of stable microtubule (MT) were increased in the hippocampus of the rats with memory deficits. This increase in stable MT is marked by α-tubulin hyperacetylation. Paclitaxel, a pharmacological MT stabilizer, increased the level of stable MT in the hippocampus and induced learning and memory deficits in normal rats. Furthermore, paclitaxel reduced long-term potentiation in hippocampal slices and increased stable MT (evidenced by α-tubulin hyperacetylation) levels in hippocampal neuronal cells. Intracerebroventricular infusion of nocodazole, an MT destabilizer, ameliorated memory deficits in rats with SNI-induced nociceptive behavior. Expression of HDAC6, an α-tubulin deacetylase, was reduced in the hippocampus in rats with cognitive impairment. These findings indicate that peripheral nerve injury (eg, SNI) affects the MT dynamic equilibrium, which is critical to neuronal structure and synaptic plasticity.

Increased HCN Channel Activity in the Gasserian Ganglion Contributes to Trigeminal Neuropathic Pain.

Orofacial neuropathic pain caused by trigeminal nerve injury is a debilitating condition with limited therapeutic options. Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels regulate neuronal excitability and are involved in the development and maintenance of chronic pain. However, the effect of HCN channel activity in the Gasserian ganglion on trigeminal neuropathic pain has not been examined. We evaluated nociceptive behaviors after microinjection of the HCN channel blockers ZD7288 or ivabradine into the Gasserian ganglion in rats with trigeminal nerve injury. Both blockers dose-dependently ameliorated evoked and spontaneous nociceptive behavior in rats with trigeminal neuropathic pain. Moreover, the clinically available HCN channel blocker ivabradine showed a prolonged antinociceptive effect. In the Gasserian ganglion, HCN1 and HCN2 are major HCN isoforms. After trigeminal nerve injury, the counts of HCN1 as well as HCN2 immuno-positive punctae were increased in the ipsilateral Gasserian ganglions. These results indicate that the increased HCN channel activity in the Gasserian ganglion directly contributes to neuropathic pain resulting from trigeminal nerve injury.

Perspective: Trigeminal nerve damage-induced orofacial pain is severe and more resistant to standard pharmacological treatment than other types of neuropathic pain. Our study suggests that targeting HCN channel activities in the Gasserian ganglion may provide an alternative treatment of trigeminal neuropathy including trigeminal neuralgia.

Multi-Source Cooperative Data Collection with a Mobile Sink for the Wireless Sensor Network.

The multi-source cooperation integrating distributed low-density parity-check codes is investigated to jointly collect data from multiple sensor nodes to the mobile sink in the wireless sensor network. The one-round and two-round cooperative data collection schemes are proposed according to the moving trajectories of the sink node. Specifically, two sparse cooperation models are firstly formed based on geographical locations of sensor source nodes, the impairment of inter-node wireless channels and moving trajectories of the mobile sink. Then, distributed low-density parity-check codes are devised to match the directed graphs and cooperation matrices related with the cooperation models. In the proposed schemes, each source node has quite low complexity attributed to the sparse cooperation and the distributed processing. Simulation results reveal that the proposed cooperative data collection schemes obtain significant bit error rate performance and the two-round cooperation exhibits better performance compared with the one-round scheme. The performance can be further improved when more source nodes participate in the sparse cooperation. For the two-round data collection schemes, the performance is evaluated for the wireless sensor networks with different moving trajectories and the variant data sizes.

Divergent Specificity Development of IgG1 and IgG4 Autoantibodies in Endemic Pemphigus Foliaceus (Fogo Selvagem).

We have shown that although the IgG response in fogo selvagem (FS) is mainly restricted to desmoglein (Dsg) 1, other keratinocyte cadherins are also targeted by FS patients and healthy control subjects living in the endemic region of Limão Verde, Brazil (endemic controls). Evaluating nonpathogenic IgG1 and pathogenic IgG4 subclass responses to desmosomal proteins may reveal important differences between pathogenic and nonpathogenic responses, and how these differences relate to the pathogenic IgG4 response and resultant FS. In this study, we tested by ELISA >100 sera from each FS patient, endemic control, and nonendemic control for IgG1 and IgG4 autoantibodies to keratinocyte cadherins besides Dsg1. IgG1 and IgG4 subclass responses in endemic controls are highly correlated between Dsg1 and other keratinocyte cadherins. This correlation persists in the IgG1 response among FS patients, but diminishes in IgG4 response, suggesting that IgG1 binds highly conserved linear epitopes among cadherins, whereas IgG4 binds mainly specific conformational epitopes on Dsg1. A confirmatory test comparing serum samples of 11 individuals before and after their FS onset substantiated our findings that IgG1 recognizes primarily linear epitopes on Dsg1 both before and after disease onset, whereas IgG4 recognizes primarily linear epitopes before disease onset, but recognizes more conformational epitopes on Dsg1 after the onset of disease. This study may provide a mechanism by which a specificity convergence of the IgG4 response to unique Dsg1 epitopes, most likely conformational pathogenic epitopes, leads to the onset of FS disease.

MicroRNA-202 inhibits cell proliferation, migration and invasion of glioma by directly targeting metadherin.

Glioma is the most common and aggressive type of primary malignant brain tumour. Increasing evidence has revealed that microRNAs play important roles in multiple biological processes related to glioma occurrence, development, diagnosis, treatment and prognosis. MicroRNA-202 (miR-202) has been studied in several types of human cancer, whereas the biological roles of miR-202 in glioma remain unknown. The present study, aimed to investigate the expression, clinical significance and biological roles of miR-202 in glioma, as well as its underlying molecular mechanism. We found that miR-202 was significantly downregulated in glioma tissues and cell lines. Low miR-202 expression was associated with Karnofsky performance status (KPS) score and World Health Organization (WHO) grade of glioma patients. Functional assays revealed that ectopic expression of miR-202 inhibited cell proliferation, migration and invasion of glioma. In addition, metadherin (MTDH) was identified as a direct target gene of miR-202 in glioma through bioinformatic analysis, luciferase reporter assay, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and western blotting. Furthermore, MTDH expression was upregulated and negatively correlated with miR-202 expression in clinical glioma tissues. MTDH knockdown had similar roles to miR-202 overexpression in glioma cells. Rescue experiments revealed that upregulation of MTDH reversed the suppression of glioma cell growth and metastasis by miR-202. Moreover, miR-202 impaired the PI3K/Akt and Wnt/β-catenin pathways. These results highlight the tumour-suppressive effect of miR-202 in glioma, thereby suggesting that miR-202 may be a potential therapeutic target for the treatment of patients with this malignancy.

We describe the optical design of a confocal scanning laser ophthalmoscope with two deformable mirrors. Spherical mirrors are used for pupil relay. Defocus aberration of the human eye is corrected by a Badal focusing structure and astigmatism aberration is corrected by a deformable mirror. The main optical system achieves a diffraction-limited performance through the entire scanning field (6 mm pupil, 3 degrees on pupil plane). The performance of the optical system, with correction of defocus and astigmatism, is also evaluated.

Improving Retinal Image Quality Using Registration with an SIFT Algorithm in Quasi-Confocal Line Scanning Ophthalmoscope.

When high-magnification images are taken with a quasi-confocal line scanning ophthalmoscope (LSO), the quality of images always suffers from Gaussian noise, and the signal to noise ratio (SNR) is very low for a safer laser illumination. In addition, motions of the retina severely affect the stabilization of the real-time video resulting in significant distortions or warped images. We describe a scale-invariant feature transform (SIFT) algorithm to automatically abstract corner points with subpixel resolution and match these points in sequential images using an affine transformation. Once n images are aligned and averaged, the noise level drops by a factor of [Formula: see text] and the image quality is improved. The improvement of image quality is independent of the acquisition method as long as the image is not warped, particularly severely during confocal scanning. Consequently, even better results can be expected by implementing this image processing technique on higher resolution images.

An Improved Rodent Model of Trigeminal Neuropathic Pain by Unilateral Chronic Constriction Injury of Distal Infraorbital Nerve.

The number of studies on trigeminal nerve injury using animal models remains limited. A rodent model of trigeminal neuropathic pain was first developed in 1994, in which chronic constriction injury (CCI) is induced by ligation of the infraorbital nerve (IoN). This animal model has served as a major tool to study trigeminal neuropathic pain. Unfortunately, the surgical procedure in this model is complicated and far more difficult than ligation of peripheral nerves (eg, sciatic nerve). The aim of this study was to improve on the current surgical procedure of IoN ligation to induce trigeminal neuropathic pain in rats. We show that the IoN can be readily accessed through a small facial incision. CCI can be induced by ligation of a segment at the distal IoN (dIoN). This dIoN-CCI procedure is simple, minimally invasive, and time-saving. Our data show that the dIoN-CCI procedure consistently induced acute as well as chronic nociceptive behaviors in rats. Daily gabapentin treatment attenuated mechanical allodynia and reduced face-grooming episodes in dIoN-CCI rats.

Perspective: The orofacial pain caused by trigeminal nerve damage is severe and perhaps more debilitating than other types of neuropathic pain. However, studies on trigeminal neuropathic pain remain limited. This is largely because of the lack of proper animal models because of the complexity of the existing surgical procedures required to induce trigeminal nerve injury. Our improved dIoN-CCI model is likely to make it more accessible to study the cellular and molecular mechanisms of neuropathic pain caused by trigeminal nerve damage.

Guasha improves the rating of perceived exertion scale score and reduces heart rate variability in male weightlifters: a randomized controlled trial.

Objective: To evaluate the effects of Guasha therapy on the rating of perceived exertion (RPE) scale score, and heart rate variability (HRV).

Methods: A randomized controlled trial of Guasha (skin scraping) was compared with a sham scraping group and control group. Sixteen sessions within an 8-week period were completed. Sixty-five male weightlifters who had undergone normal weightlifting training for a mean of 5 years before study commencement were recruited. The RPE scale score of "snatch", "clean and jerk" maneuvers (85% of one-repetition maximum), and HRV were measured before and after the intervention.

Results: The RPE scale score for snatch, clean and jerk were reduced significantly after intervention in the Guasha group and sham group. However, there was a significant difference in the low frequency (LF) domain and LF/high frequency (HF) ratio (P < 0.05): the LF domain decreased, and the LF/HF ratio decreased.

Conclusion: Guasha could be used to reduce the RPE scale score, and increase the response to HRV. Guasha could be considered as an alternative to some types of recovery from sports training.

Effect of acupuncture treatment on smoking cessation in smokers from Hong Kong.

Objective: To analyze the characteristics of smokers treated with acupuncture for smoking cessation in Hong Kong.

Methods: A total of 2051 subjects were recruited in a clinical pilot research project "acupuncture for smoking cessation", which was conducted jointly by Institute of Acupuncture and Moxibustion, China Academy of Chinese Medical Sciences and Hong Kong Pok Oi Hospital from January of 2011 to December of 2013. The characteristics of study subjects, including baseline information, smoking background, intention to quit and influencing factors were analyzed.

Results: The majority of subjects treated with acupuncture for smoking cessation in Hong Kong was male (66.7% ), but the proportion of female smokers in this study (33.3%) was higher than that of female smokers in Hong Kong population (13.8%, P < 0.05). Subjects were at the mean age of 43.83 years old, of which the percentage of females aged 31-40 years was the highest (38.8% , P < 0.05). The mean duration of smoking was 25.49 years. The number of cigarettes smoked per day was 17.57 cigarettes. Fagerstrom Test for Nicotine Dependence (FTND) was 5.29 points. Most of the subjects had attempted quitting smoking (81.42% ). The confidence index (7.44 points) and the readiness to quit smoking (8.13 points) were high. Subjects quitting smoking were mostly due to health cause (81.91%). The majority of subjects were at the level of middle school (61.63%). The higher the educational level was, the lower the tobacco dependence was and the higher the confidence in successfully quitting smoking was. 50.27% of subjects chose acupuncture for smoking cessation mainly through friends, television and network publicity. Subjects who had received acupuncture had the highest confidence index, while those who wanted to try a new method had the lower confidence index. smoking cessation mainly through friends, television and network publicity. Conclusion: Acupuncture for smoking cessation was more popular in female smokers, especially those aged 31-40 years. The effectiveness of acupuncture-smoking cessation was most significant in the smokers over 60.

Design of a Compact, Bimorph Deformable Mirror-Based Adaptive Optics Scanning Laser Ophthalmoscope.

We have designed, constructed and tested an adaptive optics scanning laser ophthalmoscope (AOSLO) using a bimorph mirror. The simulated AOSLO system achieves diffraction-limited criterion through all the raster scanning fields (6.4 mm pupil, 3° × 3° on pupil). The bimorph mirror-based AOSLO corrected ocular aberrations in model eyes to less than 0.1 μm RMS wavefront error with a closed-loop bandwidth of a few Hz. Facilitated with a bimorph mirror at a stroke of ±15 μm with 35 elements and an aperture of 20 mm, the new AOSLO system has a size only half that of the first-generation AOSLO system. The significant increase in stroke allows for large ocular aberrations such as defocus in the range of ±600° and astigmatism in the range of ±200°, thereby fully exploiting the AO correcting capabilities for diseased human eyes in the future.

Neuropeptide S modulates the amygdaloidal HCN activities (Ih) in rats: Implication in chronic pain.

Neuropeptide S (NPS), an endogenous anxiolytic, has been shown to protect against chronic pain through interacting with its cognate NPS receptor (NPSR) in the brain. However, the cellular mechanism of this NPS action remains unclear. We report that NPS inhibits hyperpolarization-activated cyclic nucleotide-gated (HCN) channel current (Ih) in the rat's amygdala through activation of NPSR. This NPS effect is mediated through ERK1/2 phosphorylation in a subset of pyramidal-like neurons located in the medial amygdala. The characters of the recorded Ih suggest a major role for HCN1 activity in this process. Inhibition of Ih by NPS stimulates the glutamatergic drive onto fast spiking intra-amygdalolidal GABAergic interneurons, which in turn facilitates GABA release onto pyramidal-like neurons. Moreover, the HCN1 expression is increased in the amygdala of rats with peripheral nerve injury and intra-amygdaloidal administration of the HCN channel inhibitor ZD7288 attenuates nociceptive behavior in these rats. These results suggest that NPS-mediated modulation of intra-amygdaloidal HCN channel activities may be an important central inhibitory mechanism for regulation of chronic pain.
