An evaluation of machine learning classifiers for next-generation, continuous-ethogram smart trackers.

Background: Our understanding of movement patterns and behaviours of wildlife has advanced greatly through the use of improved tracking technologies, including application of accelerometry (ACC) across a wide range of taxa. However, most ACC studies either use intermittent sampling that hinders continuity or continuous data logging relying on tracker retrieval for data downloading which is not applicable for long term study. To allow long-term, fine-scale behavioural research, we evaluated a range of machine learning methods for their suitability for continuous on-board classification of ACC data into behaviour categories prior to data transmission.

Methods: We tested six supervised machine learning methods, including linear discriminant analysis (LDA), decision tree (DT), support vector machine (SVM), artificial neural network (ANN), random forest (RF) and extreme gradient boosting (XGBoost) to classify behaviour using ACC data from three bird species (white stork Ciconia ciconia, griffon vulture Gyps fulvus and common crane Grus grus) and two mammals (dairy cow Bos taurus and roe deer Capreolus capreolus).

Results: Using a range of quality criteria, SVM, ANN, RF and XGBoost performed well in determining behaviour from ACC data and their good performance appeared little affected when greatly reducing the number of input features for model training. On-board runtime and storage-requirement tests showed that notably ANN, RF and XGBoost would make suitable on-board classifiers.

Conclusions: Our identification of using feature reduction in combination with ANN, RF and XGBoost as suitable methods for on-board behavioural classification of continuous ACC data has considerable potential to benefit movement ecology and behavioural research, wildlife conservation and livestock husbandry.

Hypoxia-Mediated Complement 1q Binding Protein Regulates Metastasis and Chemoresistance in Triple-Negative Breast Cancer and Modulates the PKC-NF-κB-VCAM-1 Signaling Pathway.

Objectives: Complement 1q binding protein (C1QBP/HABP1/p32/gC1qR) has been found to be overexpressed in triple-negative breast cancer (TNBC). However, the underlying mechanisms of high C1QBP expression and its role in TNBC remain largely unclear. Hypoxia is a tumor-associated microenvironment that promotes metastasis and paclitaxel (PTX) chemoresistance in tumor cells. In this study, we aimed to assess C1QBP expression and explore its role in hypoxia-related metastasis and chemoresistance in TNBC.

Materials And Methods: RNA-sequencing of TNBC cells under hypoxia was performed to identify C1QBP. The effect of hypoxia inducible factor 1 subunit alpha (HIF-1α) on C1QBP expression was investigated using chromatin immunoprecipitation (ChIP) assay. The role of C1QBP in mediating metastasis, chemoresistance to PTX, and regulation of metastasis-linked vascular cell adhesion molecule 1 (VCAM-1) expression were studied using and experiments. Clinical tissue microarrays were used to verify the correlation of C1QBP with the expression of HIF-1α, VCAM-1, and RELA proto-oncogene nuclear factor-kappa B subunit (P65).

Results: We found that hypoxia-induced HIF-1α upregulated C1QBP. The inhibition of C1QBP notably blocked metastasis of TNBC cells and increased their sensitivity to PTX under hypoxic conditions. Depletion of C1QBP decreased VCAM-1 expression by reducing the amount of P65 in the nucleus and suppressed the activation of hypoxia-induced protein kinase C-nuclear factor-kappa B (PKC-NF-κB) signaling.immunohistochemistry (IHC) staining of the tissue microarray showed positive correlations between the C1QBP level and those of HIF-1α, P65, and VCAM-1.

Conclusion: Targeting C1QBP along with PTX treatment might be a potential treatment for TNBC patients.

Expression and prognostic values of ARID family members in breast cancer.

The ARID family is a superfamily of 15 members containing a domain that interacts with AT-rich DNA elements. However, the expression and prognostic roles of each ARID in breast cancer are still elusive. We used the TCGA and Kaplan-Meier plotter databases to assess the expression and prognostic values of ARID mRNA levels in breast cancer respectively. In the present study, 6 members were significantly lower in tumor tissues than those in the normal tissues, while 6 members were significantly higher. Further assessment of ARID expression in breast cancer with different molecular subtypes, 3 members were significantly higher in no-luminal molecular subtype than those in the luminal molecular subtype, and 6 members were significantly higher. In regard to prognostic values, high expression of ARID1A, ARID2, ARID3B, ARID4A, ARID5A, ARID5B, JARID1A were associated with favorable outcome, while ARID4B and JARID1B were correlated to a worse outcome. We further analyzed the prognostic value of ARID in different intrinsic subtypes and clinicopathological features of breast cancer. We found many meaningful ARID family biomarkers in breast cancer. The relevant results will expound the role of ARID in breast cancer and may further provide new insight to explore the ARID-targeting reagents for treating breast cancer patients.

Scutellarin Ameliorates Renal Injury via Increasing CCN1 Expression and Suppressing NLRP3 Inflammasome Activation in Hyperuricemic Mice.

Considerable evidences have indicated that elevated uric acid (UA) was involved in renal tubular injury leading to hyperuricemic nephropathy (HN). Scutellarin is a biologically active flavonoid derived from the Chinese traditional herb Hand-Mazz, which has been widely used in the treatment of cardiovascular and cerebrovascular diseases. In the present study, we analyzed the effect of scutellarin on HN, by using C57BL/6 mice and human renal tubular epithelial cell line HK-2 which was subjected to adenine/potassium oxonate and UA to mimic a HN injury. The HN mice showed a significant decrease in renal function with the increased SCr and blood urea nitrogen (BUN) ( < 0.05). Hematoxylin-eosin staining results showed a histological injury in HN mice kidney tissues with severe tubular damage. Scutellarin dose dependently alleviated the renal injury of the HN model ( < 0.05), and a dose of 20 mg/kg/day remarkably reduced the Scr level (26.10 ± 3.23 μmol/ml vs. 48.39 ± 7.51 μmol/ml, < 0.05) and BUN (151.12 ± 30.24 mmol/L vs. 210.43 ± 45.67 mmol/L, < 0.05) compared with the HN model group. Similarly, scutellarin decreased NGAL, Kim-1, cystatin C, and IL-18 protein expression levels in HN mouse ( < 0.05). Overexpressed CCN1 could not induce NLRP3 inflammasome activation, with no change of mRNA and protein expression levels of NLRP3, ASC, and pro-caspase-1 compared with the control HK-2. However, HK-2 showed a significant NLRP3 inflammasome activation and apoptosis. Importantly, knockdown of CCN1 not only aggravated NLRP3 inflammasome activation and apoptosis but also abrogated the protective effect of scutellarin in UA-induced HK-2 injury. Thus, scutellarin might alleviate HN progression a mechanism involved in CCN1 regulation on NLRP3 inflammasome activation.

Inflammatory macrophages facilitate mechanical stress-induced osteogenesis.

Mechanical stress has been recognized as a key inducer of bone regeneration in bone damage, which is experimentally mimicked by distraction osteogenesis (DO), a bone-regenerative process induced by post-osteotomy distraction of the surrounding vascularized bone segments, and realized by new bone formation within the distraction gap. The mechanisms that underlie the DO-induced bone regeneration remain poorly understood and a role of macrophages in the process has been inadequately studied. Here, in a mouse model of DO, we showed significant increase in macrophages in the regeneration area. Moreover, in a loss-of-function approach by depleting inflammatory macrophages, the bone regeneration was compromised by assessment of histology and molecular biology. Thus, our study demonstrates the necessary participation of inflammatory macrophages in the process of DO-induced bone regeneration, and suggests that targeting inflammatory macrophages may help to improve clinical bone repair.

The Effects of Dynamic Work Environments on Entrepreneurs' Humble Leader Behaviors: Based on Uncertainty Reduction Theory.

Although it is widely acknowledged that the environments faced by entrepreneurs now are more dynamic than ever, little is known about the effect of dynamic work environments on entrepreneurs' leader behaviors. Based on the uncertainty reduction theory and the data from 197 entrepreneurs and their subordinates, this research found a positive relationship between dynamic work environments and entrepreneurs' humble leader behaviors. Moreover, this positive relationship can be mediated by entrepreneurs' feedback-seeking behavior. And the relationship between dynamic work environments and entrepreneurs' humble leader behaviors ( feedback-seeking) can be moderated by entrepreneurs' intolerance of uncertainty. The contributions and implications of this study are discussed.

BarcodeTree: Scalable Comparison of Multiple Hierarchies.

We propose BarcodeTree (BCT), a novel visualization technique for comparing topological structures and node attribute values of multiple trees. BCT can provide an overview of one hundred shallow and stable trees simultaneously, without aggregating individual nodes. Each BCT is shown within a single row using a style similar to a barcode, allowing trees to be stacked vertically with matching nodes aligned horizontally to ease comparison and maintain space efficiency. We design several visual cues and interactive techniques to help users understand the topological structure and compare trees. In an experiment comparing two variants of BCT with icicle plots, the results suggest that BCTs make it easier to visually compare trees by reducing the vertical distance between different trees. We also present two case studies involving a dataset of hundreds of trees to demonstrate BCT's utility.

Preparation of PolyHIPE Scaffolds for 3D Cell Culture and the Application in Cytotoxicity Evaluation of Cigarette Smoke.

Polystyrene-based polyHIPE (polymerized high internal phase emulsion) materials were prepared by the copolymerization of styrene and divinylbenzene in the continuous phase of a HIPE. The resultant polyHIPE materials were found to have an open-cellular morphology and high porosity, and the polyHIPE structure could be well adjusted by varying the water/oil (W/O) ratio and the amount of emulsifier in the HIPE. Cell culture results showed that the resultant polyHIPE materials, which exhibited larger voids and connected windows as well as high porosity, could promote cell proliferation on the 3D scaffold. A 3D cell cytotoxicity evaluation system was constructed with the polystyrene-based polyHIPE materials as scaffolds and the cigarette smoke cytotoxicity was evaluated. Results showed that the smoke cytotoxicity against A549 cells is much lower in the 3D cell platform compared to the traditional 2D system, showing the great potential of the polyHIPE scaffolds for 3D cell culture and the cytotoxic evaluation of cigarette smoke.

Massive haemoptysis from right middle lobe bronchus managed by customized silicon stents.

Massive haemoptysis is life-threatening. Management options include pharmacological treatment, bronchial artery embolization, surgical resection, and bronchoscopic interventions. As an alternative treatment method of controlling haemoptysis, endobronchial stent insertion has been performed in several pulmonary carcinoma patients. We presented an 89-year-old bronchiectasis patient who developed massive haemoptysis from the right middle lobe bronchus. Haemoptysis was not controlled by both pharmacological treatment and bronchial artery embolization. Two customized silicone stents with the one in the right middle lobe bronchus and another in the right intermediate bronchus were used to manage haemoptysis. One month after stents implantation, the stent inside the right intermediate bronchus was removed, another stent was left in the right middle lobe bronchus to continue occluding the bleeding passageway. Haemoptysis did not occur after silicon stents deployment. This may be the first case of receiving straight silicon stent implantation to manage massive haemoptysis from the right middle lobe bronchus.

Xueshuantong Injection (Lyophilized) Attenuates Cerebral Ischemia/Reperfusion Injury by the Activation of Nrf2-VEGF Pathway.

Xueshuantong injection (Lyophilized, XST), extracted from the traditional Chinese medicinal herb Panax notoginseng, has neuroprotective effect on cerebral ischemia. Revascularization of ischemic tissue is good for the therapy of cerebrovascular disease. In this study, angiogenic potentiality and possible mechanism of XST for cerebral ischemia were explored. Rats were subjected to transient middle cerebral artery occlusion (MCAO), and then intraperitoneally administered with XST daily for 3 or 7 consecutive days. The neurological function deficits, and endogenous antioxidant capacity were evaluated. Post-stroke angiogenesis and vascularization were assessed by ELISA and immunohistochemical staining. Transcription levels of Nrf2, HO-1, NQO1 in brain tissues were analyzed by real-time RT-PCR. The results showed that XST could remarkably ameliorate neuronal functional deficit, promote angiogenesis and vascularization after MCAO. The mechanism of angiogenesis might be related to endogenous antioxidant capacity and Nrf2 pathway. In conclusion, administered XST for 7 days after stroke could significantly improve functional recovery and promote angiogenesis, that might be related to Nrf2 signaling pathway. These findings could provide scientific evidence for the use of XST in cerebral ischemic diseases and provide theoretical support for further studies.

[Effects of transcutaneous electrical acupoint stimulation on treatment and prevention of postoperative sore throat after tracheal intubation under general anesthesia].

Objective: To observe the clinical efficacy of transcutaneous electrical acupoint stimulation (TEAS) at Hegu (LI 4) and Neiguan (PC 6) on treatment and prevention of postoperative sore throat (POST) after tracheal intubation under general anesthesia.

Methods: One hundred patients who received elective thyroid gland lobectomy with gradeⅠand Ⅱ of American Society of Anesthesiologists criteria were randomly assigned into a TEAS group and an anesthesia group according to random number table method, 50 cases in each group. All the patients were treated with tracheal intubation under general anesthesia. Patients in the TEAS group were treated with TEAS (2 Hz/100 Hz, 8 to 12 mA) at Hegu (LI 4) and Neiguan (PC 6) from 30 min before anesthesia induction to the end of operation. Patients in the anesthesia group were treated with TEAS at the same acupoints but no electrical stimulation was given. The incidence rate, severity and visual analogue scale (VAS) of POST were recorded 1h, 6h, 12h and 24h after tracheal extubation, respectively.

Results: The incidence rate of POST was 12.0% (6/50), 22.0% (11/50) and 18.0% (9/50) 1h, 6h, 12h after tracheal extubation respectively in the TEAS group, which was significantly lower than 30.0% (15/50), 42.0% (21/50) and 36.0% (18/50) in the anesthesia group (all <0.05). However, the incidence rate was not significantly different between the two groups 24h after extubation[14.0% (7/50) vs 28.0% (14/50), >0.05]. Moreover, the VAS scores of the patients with POST in the TEAS group at each time point were lower than those in the anesthesia group (all <0.05).

Conclusions: TEAS at Hegu (LI 4) and Neiguan (PC 6) can effectively reduce the incidence rate and severity of POST induced by tracheal intubation under general anesthesia.

Cluster analysis for syndromes of real-world coronary heart disease with angina pectoris.

Syndromes of coronary heart disease with angina pectoris were analyzed to provide guidance for clinical practice and to improve accuracy of traditional Chinese medicine (TCM) diagnoses and efficacy of TCM treatment. A total of 860 cases with coronary heart disease with angina pectoris were selected from TCM Clinical Research Information Sharing System for TCM clinics and research. Syndromes were automatically extracted with the cluster method and were analyzed to provide objective evidence for clinical studies. Final syndrome classifications were recognized and confirmed by clinical experts. Popular syndromes included Qi and blood deficiency, blood stasis and obstruction collaterals, liver depression and spleen deficiency, and Qi stagnation and blood stasis. Syndromes Qi and blood deficiency and blood stasis and obstruction collaterals accounted for 28.61% of total syndromes, whereas liver depression and spleen deficiency and Qi stagnation and blood stasis accounted for 26.44%. The main syndrome elements comprised Qi deficiency, blood deficiency, blood stasis, and Qi stagnation.

Panax ginseng genome examination for ginsenoside biosynthesis.

Ginseng, which contains ginsenosides as bioactive compounds, has been regarded as an important traditional medicine for several millennia. However, the genetic background of ginseng remains poorly understood, partly because of the plant's large and complex genome composition. We report the entire genome sequence of Panax ginseng using next-generation sequencing. The 3.5-Gb nucleotide sequence contains more than 60% repeats and encodes 42 006 predicted genes. Twenty-two transcriptome datasets and mass spectrometry images of ginseng roots were adopted to precisely quantify the functional genes. Thirty-one genes were identified to be involved in the mevalonic acid pathway. Eight of these genes were annotated as 3-hydroxy-3-methylglutaryl-CoA reductases, which displayed diverse structures and expression characteristics. A total of 225 UDP-glycosyltransferases (UGTs) were identified, and these UGTs accounted for one of the largest gene families of ginseng. Tandem repeats contributed to the duplication and divergence of UGTs. Molecular modeling of UGTs in the 71st, 74th, and 94th families revealed a regiospecific conserved motif located at the N-terminus. Molecular docking predicted that this motif captures ginsenoside precursors. The ginseng genome represents a valuable resource for understanding and improving the breeding, cultivation, and synthesis biology of this key herb.

A novel classification method for aid decision of traditional Chinese patent medicines for stroke treatment.

Traditional Chinese patent medicines are widely used to treat stroke because it has good efficacy in the clinical environment. However, because of the lack of knowledge on traditional Chinese patent medicines, many Western physicians, who are accountable for the majority of clinical prescriptions for such medicine, are confused with the use of traditional Chinese patent medicines. Therefore, the aid-decision method is critical and necessary to help Western physicians rationally use traditional Chinese patent medicines. In this paper, Manifold Ranking is employed to develop the aid-decision model of traditional Chinese patent medicines for stroke treatment. First, 115 stroke patients from three hospitals are recruited in the cross-sectional survey. Simultaneously, traditional Chinese physicians determine the traditional Chinese patent medicines appropriate for each patient. Second, particular indicators are explored to characterize the population feature of traditional Chinese patent medicines for stroke treatment. Moreover, these particular indicators can be easily obtained byWestern physicians and are feasible for widespread clinical application in the future. Third, the aid-decision model of traditional Chinese patent medicines for stroke treatment is constructed based on Manifold Ranking. Experimental results reveal that traditional Chinese patent medicines can be differentiated. Moreover, the proposed model can obtain high accuracy of aid decision.

Objectives: The prevalence and dissemination of diverse NDM-producing bacteria in China was investigated.

Methods: We collected 1,162 isolates from 8 cities during December 2013∼May 2015 in China. The NDM-positive strains as well as the NDM genotypes in these sample were detected via Vitek 2 compact system (bioMérieux, France), 16S rRNA gene sequencing, PCR and an S1- pulsed-field gel electrophoresis assay and Southern blot hybridization. The horizontal-transfer capability of the bla gene was assessed by filter mating by using a standard E.coli J53 azide-resistant strain as the recipient.

Results: Three genotypes (NDM-1, NDM-3 and NDM-5) of NDM-producing bacteria were identified, among which the NDM-1-positive isolates were the most frequent one. For the first time, we found NDM-5-produing S.typhimurium and NDM-3-produing E.coli in China. We also found that the NDM-positive (especially NDM-3 and NDM-5) strains were completely resistant to nearly all of the antimicrobial drugs utilized and bla was mostly located on diverse plasmids with sizes ranging from 30 to 670kb.

Conclusion: Various species of bacteria especially the enteric pathogens with diverse NDM genotypes had spread in China. Hence, an ongoing surveillance of their dissemination is essential to prevent and control the spread of these organisms.

A Novel Classification Method for Syndrome Differentiation of Patients with AIDS.

We consider the analysis of an AIDS dataset where each patient is characterized by a list of symptoms and is labeled with one or more TCM syndromes. The task is to build a classifier that maps symptoms to TCM syndromes. We use the minimum reference set-based multiple instance learning (MRS-MIL) method. The method identifies a list of representative symptoms for each syndrome and builds a Gaussian mixture model based on them. The models for all syndromes are then used for classification via Bayes rule. By relying on a subset of key symptoms for classification, MRS-MIL can produce reliable and high quality classification rules even on datasets with small sample size. On the AIDS dataset, it achieves average precision and recall 0.7736 and 0.7111, respectively. Those are superior to results achieved by alternative methods.

Factors influencing the quality of preconception healthcare in China: applying a preconceptional instrument to assess healthcare needs.

Background: Preconception care is defined as the promotion of the health and well-being of a woman and her partner before pregnancy. Improving preconception health can result in improved reproductive health outcomes. China has issued latest version official guideline for preconception care in 2011. The objective of this cross-sectional study is to determine whether there is a variation in the quality of preconception healthcare services in distinct eastern and northern populations of China, and what factors are associated with such variation.

Methods: A cross-sectional survey using our previously developed preconception instrument was conducted. Women at reproductive age planning for pregnancy were surveyed along with their partners at hospitals during their pre-pregnancy health examination. Data collected include general health/life profiles, pregnancy history, alcohol/tobacco/drug exposures, immunizations, micronutrient supplements and the demands in preconception care. After quality assessment, statistical analysis were applied to evaluate the variations in preconception factors between people from Hebei and Jiangsu Provinces.

Results: 3202 women of reproductive age in from eastern province, Jiangsu, and in a northern province, Hebei, participated this study. 2806 of them and their partners have completed the questionnaire, at a rate of 87.6%, 1011 were from Jiangsu and 1795 were from Hebei. Statistical significance was obtained for maternal age (P < 0.001), body mass index (u =13.590, P <0.001), education (χ2 = 916.33, P < 0.001), occupation (χ2 = 901.78, P < 0.001), health status/common disease, immunization status, and need for preconception care.

Conclusions: For a country as large as China, the centralized guideline for standardized preconception healthcare does have a very crucial positive role in reproductive healthcare, but it may not be suited for all populations. Regional authorities should consider the demographics and healthcare needs of the local population and modify the centralized guideline accordingly, as well as provide a better education and professional services for the public, to improve the quality of preconception services at both the regional and the national level.

Scientific computation of big data in real-world clinical research.

The advent of the big data era creates both opportunities and challenges for traditional Chinese medicine (TCM). This study describes the origin, concept, connotation, and value of studies regarding the scientific computation of TCM. It also discusses the integration of science, technology, and medicine under the guidance of the paradigm of real-world, clinical scientific research. TCM clinical diagnosis, treatment, and knowledge were traditionally limited to literature and sensation levels; however, primary methods are used to convert them into statistics, such as the methods of feature subset optimizing, multi-label learning, and complex networks based on complexity, intelligence, data, and computing sciences. Furthermore, these methods are applied in the modeling and analysis of the various complex relationships in individualized clinical diagnosis and treatment, as well as in decision-making related to such diagnosis and treatment. Thus, these methods strongly support the real-world clinical research paradigm of TCM.

Spermicidal and antifertility effects of an imbibing and soluble nonoxynol-9 diaphragm (ISND) in rabbits.

Objective: To determine the minimum effective concentration (MEC) of an imbibing and soluble nonoxynol-9 (N-9) diaphragm (ISND) required for immobilisation of all spermatozoa in vitro and in vivo. The speed of semen absorbance, time of ISND to dissolution, and the antifertility effects were also investigated in rabbits.

Methods: In vitro spermicidal tests with ISND were conducted using fresh semen from humans and rabbits. Spermicidal and antifertility effects were observed in vivo after the ISND was placed directly into the vagina of rabbits.

Results: The MEC of N-9 required in the ISND to totally immobilise sperm within 20 seconds was 0.15 mg/ml for human sperm, and 0.5 mg/ml for rabbit sperm. The human semen was absorbed into the ISND in 45 minutes; the diaphragm dissolved in the vagina 3.5 hours later. In vivo, in rabbits, the MEC of N-9 required to immobilise sperm within five minutes of mating was 1 mg/kg in the ISND, and 10 mg/kg for the nonoxynol-9 film. The median effective dose of N-9 in the ISND was 1.07 mg/kg, whereas for the film it was 3.30 mg/kg.

Conclusion: The spermicidal and antifertility activities of a low dose N-9 in the ISND were high, with properties of imbibition and solubility confirmed.

Automated skin biopsy histopathological image annotation using multi-instance representation and learning.

With digitisation and the development of computer-aided diagnosis, histopathological image analysis has attracted considerable interest in recent years. In this article, we address the problem of the automated annotation of skin biopsy images, a special type of histopathological image analysis. In contrast to previous well-studied methods in histopathology, we propose a novel annotation method based on a multi-instance learning framework. The proposed framework first represents each skin biopsy image as a multi-instance sample using a graph cutting method, decomposing the image to a set of visually disjoint regions. Then, we construct two classification models using multi-instance learning algorithms, among which one provides determinate results and the other calculates a posterior probability. We evaluate the proposed annotation framework using a real dataset containing 6691 skin biopsy images, with 15 properties as target annotation terms. The results indicate that the proposed method is effective and medically acceptable.

Application of metabolomics on diagnosis and treatment of patients with psoriasis in traditional Chinese medicine.

Traditional Chinese medicine (TCM) is one of the oldest forms of medical system. With syndrome as the core of diagnosis and therapy in TCM, it has the advantage of collecting macroscopic information of patients for diagnosis. To understand the in vivo mechanism of TCM, a metabolomics approach was used to investigate the global biological characterization of the urine of psoriasis patients with Blood Stasis Syndrome and the therapeutic metabolomics mechanism of the Optimized Yinxieling formula. A total of 41 cases of psoriasis patients with Blood Stasis Syndrome and 19 healthy volunteers were enrolled in this study. Fasting urine samples from patients with consecutive Optimized Yinxieling intake after 0, 4, 8 and 12 weeks and from healthy volunteers were analyzed by Orthogonal Projection on Latent Structures Discriminant Analysis (OPLS-DA), which was utilized for High Performance Liquid Chromatography (HPLC) analysis and temporal metabolic changes identification. For psoriasis group, the scores of PASI of patients decreased after 12 weeks of Optimized Yinxieling treating. The metabolic variations visualized not only in the healthy group and psoriasis group, but also in the psoriasis group before and after Optimized Yinxieling treatment, demonstrated that the metabolic characteristics of the two groups were significantly different. The optimized complex structure of the target proteins from Protein Data Bank was analyzed by software package Discovery Studio. With docking score of original inhibitor and the receptor as the threshold values, two compounds from Chinese medicinal chemical database were predicted to have good interactions with the target proteins. The Metabolomics technique combining molecular docking analysis enhanced our current understanding of the metabolic response to Blood Stasis Syndrome of Psoriasis and the action mechanism of Optimized Yinxieling. Developing an efficient TiO2-coated biofilm carrier for intimate coupling of photocatalysis and biodegradation.

Intimate coupling of photocatalysis and biodegradation (ICPB) shows promise to remove recalcitrant organic compounds from water, because photocatalysis breaks the compounds into biodegradable intermediates that are immediately mineralized by microorganisms inside a macroporous carrier, where they are protected from UV light and free radicals. Key to successful ICPB is a carrier capable of accumulating biofilm in its interior and strongly adhering photocatalyst on its exterior. We employed a low-temperature sintering method, the O method, to adhere TiO(2) to sponge-type macroporous carriers. The O method achieved a 7-fold increase of TiO(2) density, compared to the previously used sol-gel method, and it conserved the macropores for biofilm accumulation. Although the O-method carrier met the basic requirements of ICPB when degrading 2,4,5 trichlorophenol, it had low photocatalytic activity for breaking down more complex aromatics, like reactive dyes. Therefore, we improved the sintering method in two steps called the D and DN methods: reducing the TiO(2) concentration and then removing trimesic acid in the coating solution. The photocatalytic efficiency towards reactive black 5 increased 5 fold for the D method and 10 fold for DN method, and the DN carrier had superior TiO(2) adherence during long-term operation.

Investigations of the barbell ultrasonic transducer operated in the full-wave vibrational mode.

In this paper, the resonance frequency equation and expression of displacement amplitude magnifications of a full-wave barber ultrasonic horn are obtained. By discussing the relationships between the displacement amplitude magnifications and the geometrical dimensions, the optimized design of the horn for the largest magnification is proposed, which is helpful to improve the radiation power and the transfer efficiency of the acoustic energy of the ultrasonic oscillatory system. Based on the optimized design of the horn, we introduced a barbell ultrasonic transducer operated in the longitudinal full-wave vibrational model and obtained the resonance frequency equations. For comparison, the resonance frequencies of the full-wave barbell horn and the full-wave barbell transducer are also analyzed by finite element method (FEM). It is shown that the values obtained by theoretical analysis and FEM are in good agreement with experimental observations. We hope that the research of this paper is helpful for the use of the barbell horn and transducer in the applications such as ultrasonic liquid processing.

2,4-DNT removal in intimately coupled photobiocatalysis: the roles of adsorption, photolysis, photocatalysis, and biotransformation.

The removal of 2,4-dinitrotoluene (2,4-DNT) by simultaneous UV-photo(cata)lysis and biodegradation was explored using intimately coupled photolysis/photocatalysis and biodegradation (ICPB) with two novel porous carriers. First, a porous ceramic carrier was used to attach the photocatalyst (TiO₂) on its exterior and accumulate biomass in its interior. UV irradiation alone decomposed 71% of the 2,4-DNT in 60 h, and TiO₂ catalyst improved the photolysis to 77%. Second, a macroporous sponge carrier was used to strongly adsorb 2,4-DNT and protect microorganisms from 2,4-DNT inhibition and UV irradiation. The main photolytic reactions were reduction of the nitryl to amino and hydrolysis of the amino to release NH₄⁺. The main biodegradation reactions were oxidative release of NO₃⁻ and accelerated reductive release of NH₄⁺. ICPB more thoroughly released inorganic N, with nearly equal amounts being oxidized to nitrate and reduced to ammonium. The genera Burkholderia and Bacillus were found inside the sponge carriers, and they are associated with biodegradation of DNT and its photolysis intermediates. Therefore, using an adsorbent and macroporous biofilm carrier enabled the effective removal of 2,4-DNT by ICPB.

Degradation of reactive dyes in a photocatalytic circulating-bed biofilm reactor.

Decolorization and mineralization of reactive dyes by intimately coupled TiO₂-photocatalysis and biodegradation (ICPB) on a novel TiO₂-coated biofilm carrier were investigated in a photocatalytic circulating-bed biofilm reactor (PCBBR). Two typical reactive dyes--Reactive Black 5 (RB5) and Reactive Yellow 86 (RY86)--showed similar first-order kinetics when being photocatalytically decolorized at low pH (~4-5) in batch experiments. Photocatalytic decolorization was inhibited at neutral pH in the presence of phosphate or carbonate buffer, presumably due to electrostatic repulsion from negatively charged surface sites on TiO₂, radical scavenging by phosphate or carbonate, or both. Therefore, continuous PCBBR experiments were carried out at a low pH (~4.5) to maintain high photocatalytic efficiency. In the PCBBR, photocatalysis alone with TiO₂-coated carriers could remove target compound RB5 and COD by 97% and 47%, respectively. Addition of biofilm inside macroporous carriers maintained a similar RB5 removal efficiency, but COD removal increased to 65%, which is evidence of ICPB despite the low pH. ICPB was further proven by finding microorganisms inside carriers at the end of the PCBBR experiments. A proposed ICPB pathway for RB5 suggests that a major intermediate, a naphthol derivative, was responsible for most of the residual COD, while most of the nitrogen in the azo-bonds (-N=N-) was oxidized to N₂.

Intimate coupling of photocatalysis and biodegradation (ICPB) offers potential for degrading biorecalcitrant and toxic organic compounds. This study reports on a novel sponge-type, TiO(2)-coated biofilm carrier that showed significant adherence of TiO(2) and ability to accumulate biomass in its interior. This carrier was tested for ICPB in a continuous-flow photocatalytic circulating-bed biofilm reactor (PCBBR) to mineralize 2,4,5-trichlorophenol (TCP), which is biorecalcitrant. Four mechanisms possibly acting in ICPB were tested separately: TCP adsorption to the carrier, UV photolysis, UV photocatalysis, and biodegradation by biofilm inside the carrier. The carrier exhibited strong TCP adsorption that followed a Freundlich isotherm with an exponent near 2. Whereas UV photolysis was negligible, photocatalysis produced TCP-degradation products that could be mineralized, and the strong adsorption of TCP to the carrier enhanced biodegradation by relieving toxicity. Validating the ICPB concept, biofilm was protected inside the carriers, although biomass originally on the outer surface of the carriers was eliminated. ICPB significantly lowered the diversity of the bacterial community, but five genera known to biodegrade chlorinated phenols (Ralstonia, Bradyrhizobium, Methylobacterium, Cupriavidus, and Pandoraea) were markedly enriched.

[Rule induction algorithm for brain glioma using support vector machine].

A new proposed data mining technique, support vector machine (SVM), is used to predict the degree of malignancy in brain glioma. Based on statistical learning theory, SVM realizes the principle of data dependent structure risk minimization, so it can depress the overfitting with better generalization performance, since the prediction in medical diagnosis often deals with a small sample. SVM based rule induction algorithm is implemented in comparison with other data mining techniques such as artificial neural networks, rule induction algorithm and fuzzy rule extraction algorithm based on fuzzy max-min neural networks (FRE-FMMNN) proposed recently. Computation results by 10 fold cross validation method show that SVM can get higher prediction accuracy than artificial neural networks and FRE-FMMNN, which implies SVM can get higher accuracy and more reliability. On the whole data sets, SVM gets one rule with the classification accuracy of 89.29%, while FRE-FMMNN gets two rules of 84. 64%, in which the rule got by SVM is of quantity relation and contains more information than the two rules by FRE-FMMNN. All the above show SVM is a potential algorithm for the medical diagnosis such as the prediction of the degree of malignancy in brain glioma.
