Carcass characteristics and meat quality of lambs fed with cassava foliage hay and spineless cactus.

The objective of this study was to evaluate the influence of the total or partial replacement of Tifton 85 hay (TH) by cassava foliage hay (CH) and/or spineless cactus (SC) on the carcass characteristics and meat quality of confined lambs. Thirty-five Dorper x Santa Inês crossbred lambs were used, with an initial average weight of 16.0 ± 1.9 kg, fed with five treatment diets: 70% TH, 70% CH, 35% TH + 35% CH, 35% TH + 35% SC, and 35% CH + 35% SC. After 90 days of confinement, the animals were slaughtered and the carcass and meat data were compared using the Tukey test at 5%. The partial replacement of TH by SC increased the cold carcass weight by 3.9 kg (p < .05). The proportion of fat (%) increased (p < .05) when TH was replaced by CH + SC. The intensity of red (a*) was lower (p < .05) in the meat of animals fed with CH or CH + SC. It is recommended to partially replace TH with SC, or to replace totally with CH in association with SC.

A new robust adaptive mixing control for trajectory tracking with improved forward flight of a tilt-rotor UAV.

A new robust adaptive mixing control (RAMC) is proposed in order to accomplish trajectory tracking of a tilt-rotor unmanned aerial vehicle (UAV) configuration. This kind of system is a hybrid aerial vehicle that combines advantages of rotary-wing aircraft, like hovering flight and vertical take-off and landing (VTOL), and those of fixed-wing aircraft, as improved forward flight. Although the VTOL and cruise flight regimes present different dynamic behaviors, in this work a unified, highly coupled, nonlinear model is developed to cope with the considered tilt-rotor UAV full flight envelope, that is, the axial flight, hovering, transition/cruise and turning flight. The modeling is performed via Euler-Lagrange formulation considering the tilt-rotor UAV as a multi-body system and taking into account aerodynamic effects and the dynamics of the tilting servomotors. Accordingly, in order to comply with the trajectory tracking requirements and improve the tilt-rotor UAV forward flight, this paper presents a novel robust adaptive mixing controller which is formulated to deal with linear parameter-varying (LPV) systems dependent on not known a priori large parameters but measured or estimated online, and also to provide robustness against unknown disturbances. Additionally, a rigorous closed-loop stability analysis is performed. The controller performance is validated with numerical experiments conducted using a high fidelity simulator developed on Gazebo and Robot Operating System (ROS) platforms.

The inclusion of spineless cactus in the diet of lambs increases fattening of the carcass.

The objective of this study was to evaluate the effect of replacement maniçoba hay by spineless cactus on the performance, carcass characteristics, and meat quality of confined lambs. Thirty-two male lambs, with an average body weight of 20.8 ± 2.9 kg, were fed with different levels of spineless cactus in replacement for 0, 33, 66 or 100% of maniçoba hay (i.e. 0, 200, 400, 600 g kg of dietary DM). The replacement maniçoba hay by spineless cactus did not influence dry matter intake (g d) and cold carcass weight (P > .05), but increased (P < .05) the carcass finishing fat but did not influence (P < .05) the physical characteristics of the lambs' meat. Lower values for odor, flavor and purchase intention of meat were observed with 66% of replacement. Therefore, replacement of 33% maniçoba hay by spineless cactus can be recommended as optimal level, because it improve the fattening of the carcass, without causing negative effects on performance or meat quality.

Effect of association of fresh cassava root with corn silage in replacement for cactus cladodes on dairy cow performance.

The aim of this study was to investigate the replacement effect of cactus cladodes with cassava root and corn silage (CRCS) (0, 25, 50, 75, and 100%, dry matter basis) in diets of lactating dairy cows on nutrients intake and digestibility, microbial protein synthesis, nitrogen balance, and milk yield. Ten Holstein cows with an average milk yield of 16 kg/day, an initial body weight (BW) of 670 ± 35 kg, and an average of 17 weeks in milking were assigned to an experiment using a double 5 × 5 Latin square design. The ratio of forage/concentrate was 82:18, and the ingredients used were cactus cladodes, cassava root, corn silage, soybean meal, wheat meal, urea, and mineral premix. The intake of dry matter, organic matter, and total digestible nutrients were not influenced by the replacement levels. On the other hand, the intake of crude protein and neutral detergent fiber increased while the intake of non-fiber carbohydrates decreased with addition of CRCS. The digestibility of the nutrients was not changed as the milk yield as fat-corrected milk production and milk content of fat, protein, lactose, and total solids. The microbial protein synthesis, microbial protein efficiency, nitrogen balance, and efficiency of N in milk were not changed. Therefore, we recommend the complete replacement of cactus cladodes by cassava root and corn silage in the diets of lactating cows with an average milk yield of 16 kg/day.

Intake, digestibility, and performance of lambs fed spineless cactus cv. Orelha de Elefante Mexicana.

Objective: To evaluate the effects of the carmine cochineal-resistant spineless cactus genotypes cv. Orelha de Elefante Mexicana (Opuntia) and Miúda (Nopalea) on the intake and digestibility of nutrients, ingestive behavior, performance, and ultrasound measurements of growing lambs.

Methods: Thirty-six male (non-castrated) Santa Inês lambs were used, with an average age of 6 months and an initial average weight of 22.0±2.9 kg. They were distributed in a completely randomized design with 3 treatments (Tifton hay, Nopalea and Opuntia) and 12 replications, using initial weight as a covariate. The experimental period was 86 days, with the first 30 days used for the adaptation of the animals to the facilities, diets and management, and the remaining 56 days used for evaluation and data collection.

Results: The intake and apparent digestibility of dry matter (DM), organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), total carbohydrates (TC), non-fibrous carbohydrates (NFC), and total digestible nutrients (TDN) showed a significant difference (p<0.05) as a function of the diets, with the Nopalea treatment (p<0.05) increasing DM intake (g/kg and % body weight [BW]), CP, TDN, and TC digestibility, whereas the Tifton hay diet led to a high (p<0.001) neutral detergent fiber corrected for ash and protein (NDFap) g/d intake, NDFap (BW %) and digestibility of said nutrient. There was no effect of treatments (p>0.05) on feeding time, however, rumination time and total chewing time were higher (p<0.05) for animals fed Tifton hay. The performance of the animals was similar (p>0.05). For the ultrasound measurements, Nopalea promoted an increase in the final loin eye area, compared to Tifton hay.

Conclusion: The use of spineless cactus variety Miúda leads to the greater intake and digestibility of nutrients. The evaluated carmine cochineal-resistant spineless cactus genotypes are alternatives for semi-arid regions as they do not negatively affect the performance of growing lambs.

Light-activated generation of nitric oxide (NO) and sulfite anion radicals (SO˙) from a ruthenium(ii) nitrosylsulphito complex.

This manuscript describes the preparation of a new Ru(ii) nitrosylsulphito complex, trans-[Ru(NH)(isn)(N(O)SO)] (complex 1), its spectroscopic and structural characterization, photochemistry, and thermal reactivity. Complex 1 was obtained by the reaction of sulfite ions (SO) with the nitrosyl complex trans-[Ru(NH)(isn)(NO)] (complex 2) in aqueous solution resulting in the formation of the N-bonded nitrosylsulphito (N(O)SO) ligand. To the best of our knowledge, only four nitrosylsulphito metal complexes have been described so far (J. Chem. Soc., Dalton Trans., 1983, 2465-2472), and there is no information about the photochemistry of such complexes. Complex 1 was characterized by spectroscopic means (UV-Vis, EPR, FT-IR, H- and N-NMR), elemental analysis and single-crystal X-ray diffraction. The X-ray structure of the precursor complex 2 is also discussed in the manuscript and is used as a reference for comparisons with the structure of 1. Complex 1 is water-soluble and kinetically stable at pH 7.4, with a first-order rate constant of 3.1 × 10 s for isn labilization at 298 K (t∼ 373 min). Under acidic conditions (1.0 M trifluoroacetic acid), 1 is stoichiometrically converted into the precursor complex 2. The reaction of hydroxide ions (OH) with 1 and with 2 yields the Ru(ii) nitro complex trans-[Ru(NH)(isn)(NO)] with second-order rate constants of 2.1 and 10.5 M s (at 288 K), respectively, showing the nucleophilic attack of OH at the nitrosyl in 2 (Ru-NO) and at the nitrosylsulphito in 1 (Ru-N(O)SO). The pK value of the -SO moiety of the N(O)SO ligand in 1 was determined to be 5.08 ± 0.06 (at 298 K). The unprecedented photochemistry of a nitrosylsulphito complex is investigated in detail with 1. The proposed mechanism is based on experimental (UV-Vis, EPR, NMR and Transient Absorption Laser Flash Photolysis) and theoretical data (DFT) and involves photorelease of the N(O)SO ligand followed by formation of nitric oxide (NO˙) and sulfite radicals (SO˙, sulfur trioxide anion radical).

Carcass characteristics of lambs fed diets with increasing levels of crude glycerin.

Objective: An experiment was conducted to the effects of increasing levels of crude glycerin (0, 6, 12 and 18%) used as a substitute for corn in lamb feed on the quantitative characteristics of the carcass.

Methods: A total of 40 crossbred Santa Inês lambs that were four months old with a mean initial weight of 21.0 ± 0.8 kg were randomly distributed in four treatments with ten replicates. The animals were slaughtered after 66 days of confinement. The effects of crude glycerin as a replacement for corn in the diet of the lambs on the carcass characteristics, commercial cut weight and yield and carcass measurements were studied.

Results: There was an increasing linear effect for body weight at slaughter with the replacement of corn by crude glycerin. The dry matter (DM) and metabolizable energy (ME) intakes, weight of the empty body, hot carcass weight and cold carcass weight showed a quadratic effect, with maximum crude glycerin levels estimated at 10.9, 9.8, 10.83, 11.78 and 11.35%, respectively. The initial pH was not influenced by the substitution of corn for crude glycerin, while the final pH presented a quadratic effect. The other parameters of the carcass and the weights and yields of commercial cuts were not influenced. There was also no effect of the diets on carcass morphometric measurements, except for the thoracic perimeter and the carcass compactness index, which presented quadratic and linear effects, respectively.

Conclusion: Crude glycerin can replace up to 18% of corn because favour the muscle tissue deposition without promoting changes in the main carcass characteristics of lambs.

Intake, digestibility and metabolism in sheep fed with increasing levels of spineless cactus (Nopalea cochenillifera Salm-Dyck).

The objective of this study was to evaluate the effect of the substitution of Tifton 85 hay (Cynodon spp.) with spineless cactus (Nopalea cochenillifera) on the intake, digestibility of nutrients and ingestive behaviour of sheep. Eight rumen-cannulated sheep were used, with an average body weight of 58.44 ± 13.5 kg. The treatments consisted of the replacement of Tifton 85 hay with cactus at levels of 0, 333, 666 and 1000 g/kg. Samples of food and leftovers were collected at the beginning of each trial period. Digestibility was estimated using indigestible acid detergent fibre. Ingestive behaviour was evaluated via scan sampling. The replacement of Tifton 85 hay with spineless cactus had a significant effect (P < 0.05) on the intake and digestibility of neutral detergent fibre and non-fibre carbohydrates. The rumination efficiency of neutral detergent fibre was not affected (P > 0.05) by the substitution of Tifton 85 hay with spineless cactus. However, plasma glucose, phosphorus and magnesium were linearly influenced by Tifton 85 hay replacement with spineless cactus. Therefore, spineless cactus can be used to replace up to 500 g/kg of Tifton 85 hay in complete diets for sheep.

Physicochemical parameters, fatty acid profile, and sensory attributes of meat from lambs fed with cassava dregs in replacement of corn.

The objective of this study was to evaluate the effect of the replacement of corn by cassava dregs (0, 33, 66, and 100%) in the diet of lamb on physicochemical parameters, fatty acid profile, and sensory attributes of meat. Forty male non-castrated Santa Inês crossbred lambs with an initial average weight of 20 ± 1.87 kg were slaughtered after 70 days. The physicochemical parameters and chemical composition of meat were not affected by the diets. Eicosenoic (C20:1n-9) and linolenic (C18:3n-3) acids increased linearly and there was a linear decreasing effect for the flavor score with increasing cassava dregs in the diets. The replacement of corn by cassava dregs did not influence omega-6:omega-3 ration or the total desirable fatty acids, as well as the color, aroma, tenderness, juiciness, or overall evaluation scores. Cassava dregs may be added in the diets of lambs, without negative effects on physicochemical characteristics, chemical composition, and fatty acid indexes, with positive effects on unsaturated fatty acid content. However, the total replacement of corn altered the sensory attributes and decreased purchase intent of the meat.

Should splenic autotransplantation be considered after total splenectomy due to trauma?

Trauma is a public health problem and the most common cause of death in people under the age of 45. In blunt abdominal trauma, the spleen is the most commonly injured organ. Splenectomy remains the most common treatment, especially in high-grade lesions, despite increased nonoperative treatment. Removal of the spleen leads to increased susceptibility to infections due to its role in the immune function. Postsplenectomy sepsis is an important complication and presents a high mortality rate. Patients undergoing splenectomy should be immunized for encapsulated germs, as these are the agents most commonly associated with such infections. Splenic autotransplantation is a simple procedure, which can be an alternative to reduce infection rates consequent to total splenectomy, and reduce costs related to hospitalizations. This review aims to provide evidence-based information on splenic autotransplantation and its impact on the prognosis of patients undergoing total splenectomy. We searched the Cochrane Library, Medline/PubMed, SciELO and Embase, from January 2017 to January 2018 and selected articles in English and Portuguese, dated from 1919 to 2017. We found that the adjusted risk of death in splenectomized patients is greater than that of the general population, and when total splenectomy is performed, splenic autotransplantation is the only method capable of preserving splenic function, avoiding infections, especially postsplenectomy sepsis. Health professionals should be familiar with the consequences of the method chosen to manage the patient suffering from splenic trauma.

Different roughage:concentrate ratios with and without liquid residue of cassava for lambs.

This aim of this study was to evaluate the intake and apparent digestibility of nutrients, ingestive behavior, blood parameters, and performance of lambs fed two roughage:concentrate (R:C) ratios, with or without liquid residue of cassava (LRC). Forty lambs (19.5 ± 1.45 kg body weight) were distributed in a completely randomized 2 × 2 factorial design. The intake of dry matter (1311 g/day) and total digestible nutrients (755 g/day), and the intake (172 g/day) and digestibility (552 g/kg) of crude protein were higher for lambs fed R:C of 40:60. The R:C ratio of 80:20 promoted a higher intake (585 g/day) and digestibility (461 g/kg) of neutral detergent fiber. There was an intake of LRC of 1.37 L/day for lambs fed R:C of 80:20 and 0.42 L/day for lambs fed R:C of 40:60. The lambs fed R:C of 80:20 spent more time in feeding (326 m/day) and rumination (530 m/day). The metabolic profile and performance of lambs were not affected by LRC inclusion. The body weight gain (220 g/day) was higher in lambs fed R:C of 40:60. A roughage:concentrate ratio of 40:60 is recommended for lambs because it resulted in the best performance, regardless of supplementation with liquid residue of cassava.

Mate extract as feed additive for improvement of beef quality.

Mate (Ilex paraguariensis A.St.-Hil.) is generally recognized as safe (GRAS status) and has a high content of alkaloids, saponins, and phenolic acids. Addition of mate extract to broilers feed has been shown to increase the oxidative stability of chicken meat, however, its effect on beef quality from animals supplemented with mate extract has not been investigated so far. Addition of extract of mate to a standard maize/soy feed at a level of 0.5, 1.0 or 1.5% w/w to the diet of feedlot for cattle resulted in increased levels of inosine monophosphate, creatine and carnosine in the fresh meat. The content of total conjugated linoleic acid increased in the meat as mate extract concentration was increased in the feed. The tendency to radical formation in meat slurries as quantified by EPR spin-trapping decreased as increasing mate extract addition to feed, especially after storage of the meat, indicating higher oxidative stability. Mate supplementation in the diet did not affect animal performance and carcass characteristics, but meat from these animals was more tender and consequently more accepted by consumers. Mate extract is shown to be a promising additive to feedlot diets for cattle to improve the oxidative stability, nutritive value and sensory quality of beef.

Photoinduced Charge Shifts and Electron Transfer in Viologen-Tetraphenylborate Complexes: Push-Pull Character of the Exciplex.

Viologen-tetraarylborate ion-pair complexes were prepared and investigated by steady-state and time-resolved spectroscopic techniques such as fluorescence and femtosecond transient absorption. The results highlight a charge transfer transition that leads to changes in the viologen structure in the excited singlet state. Femtosecond transient absorption reveals the formation of excited-state absorption and stimulated emission bands assigned to the planar (k < 10 s) and twisted (k ∼ 10 s) structures between two pyridinium groups in the viologen ion. An efficient photoinduced electron transfer from the tetraphenylborate anionic moiety to the viologen dication was observed less than 1 μs after excitation. This is a consequence of the push-pull character of the electron donor twisted viologen structure, which helps formation of the borate triplet state. The borate triplet state is deactivated further via a second electron transfer process, generating viologen cation radical (V).

Outer membrane vesicles extracted from Neisseria meningitidis serogroup X for prevention of meningococcal disease in Africa.

Meningococcal disease is caused mainly by serogroups A, B, C, Y, W of N. meningitidis. However, numerous cases of meningitis caused by serogroup X N. meningitidis (MenX) have recently been reported in several African countries. Currently, there are no licensed vaccines against this pathogen and most of the MenX cases have been caused by meningococci from clonal complex (c.c) 181. Detergent extracted meningococcal outer membrane vesicle (dOMV) vaccines have previously shown to be safe and effective against epidemics of serogroup B meningococcal disease in all age groups. The aim of this work is therefore to obtain, characterize and evaluate the vaccine potential of dOMVs derived from a MenX strain (OMVx). Three experimental lots of OMVx were prepared by deoxycholate extraction from the MenX strain BF 2/97. Size and morphology of the vesicles was determined by Dynamic Light Scattering and electron microscopy, whereas the antigenic composition was characterized by gel electrophoresis and immunoblotting. OMVx were thereafter adsorbed to aluminium hydroxide (OMVx/AL) and two doses of OMVx were administered s.c. to groups of Balb/c mice three weeks apart. The immunogenicity and functional antibody activities in sera were evaluated by ELISA (anti-OMVx specific IgG responses) and serum bactericidal activity (SBA) assay. The size range of OMVx was shown to be between 90 and 120nm, whereas some of the antigens detected were the outer membrane proteins PorA, OpcA and RmpM. The OMVx/AL elicited high anti-OMVx antibody responses with bactericidal activity and no bactericidal activity was observed in the control group of no immunised mice. The results demonstrate that OMVx are immunogenic and could form part of a future vaccine to prevent the majority of meningococcal disease in the African meningitis belt.

Immune responses of a meningococcal A + W outer membrane vesicle (OMV) vaccine with and without aluminium hydroxide adjuvant in two different mouse strains.

Meningococci (Neisseria meningiditis) of serogroups A and W have caused large epidemics of meningitis in sub-Saharan Africa for decades, and affordable and multivalent vaccines, effective in all age groups, are needed. A bivalent serogroup A and W (A + W) meningococcal vaccine candidate consisting of deoxycholate-extracted outer membrane vesicles (OMV) from representative African disease isolates was previously found to be highly immunogenic in outbred mice when formulated with the adjuvant aluminium hydroxide (AH). OMV has been shown to have inherent adjuvant properties. In order to study the importance of AH and genetical differences between mice strains on immune responses, we compared the immunogenicity of the A + W OMV vaccine when formulated with or without AH in inbred C57BL/6J and BALB/cJ mice (Th1 and Th2 dominant strains, respectively). The immunogenicity of the vaccine was found to be comparable in the two mice strains despite their different immune profiles. Adsorption to AH increased anti-OMV IgG levels and serum bactericidal activity (SBA). The immune responses were increased by each dose for the adsorbed vaccine, but the third dose did not significantly improve the immunogenicity further. Thus, a vaccine formulation with the A and W OMV will likely benefit from including AH as adjuvant.

New insight into the photophysics and reactivity of trigonal and tetrahedral arylboron compounds.

The photophysics and reactivity of two tetraphenylborate salts and triphenylborane have been studied using ultrafast transient absorption, steady-state fluorescence, electron paramagnetic resonance with spin trapping, and DFT calculations. The singlet excited state of tetraarylborates exhibit extended π-orbital coupling between two adjacent aryl groups. The maximum fluorescence band, as well as the transient absorption bands centered at 560 nm (τ = 1.05 ns) and 680 nm (τ = 4.35 ns) are influenced by solvent viscosity and polarity, indicative of a twisted intramolecular charge transfer (TICT) state. Orbital contour plots of the HOMO and LUMO orbitals of the tetraarylboron compounds support the existence of electron delocalization between two aryl groups in the LUMO. This TICT-state and aryl-aryl electron extension is not observed for the trigonal arylboron compound, in which excited π-orbital coupling only occurs between the boron atom and one aryl group, which restricts the twist motion of the aryl-boron bond. The excited triplet state is deactivated primarily through aryl-boron bond cleavage, yielding aryl and diphenylboryl radicals. In the presence of oxygen, this photochemistry results in phenoxyl and diphenylboroxyl radicals, as confirmed by EPR spectroscopy of spin trapped radical adducts. The TICT transition and radical generation is not expected for BoDIPY molecules where the rotational vibration of the B-aryl bond is rigid, restricting changes in the geometric structure. In this sense, this work contributes to the development of new BoDIPY derivatives where the TICT transition may be observed for aryl ligands with free rotational vibrations in the BoDIPY structure.

Limited proteolysis of myoglobin opens channel in ferrochelatase-globin complex for iron to zinc transmetallation.

Recombinant ferrochelatase (BsFECH) from Bacillus subtilis expressed in Escherichia coli BL21(DE3) was found by UV-visible spectroscopy to bind the model substrate tetraphenylporphyrin-sulfonate, TPPS, with Ka=3.8 10(5)mol/L in aqueous phosphate buffer pH 5.7 at 30°C, and to interact with metmyoglobin with Ka=1.07±0.13 10(5)mol/L at 30°C. The iron/zinc exchange in myoglobin occurring during maturation of Parma hams seems to depend on such substrate binding to BsFECH and was facilitated by limited pepsin proteolysis of myoglobin to open a reaction channel for metal exchange still with BsFECH associated to globin. BsFECH increased rate of zinc insertion in TPPS significantly and showed saturation kinetics with an apparent binding constant of Zn(II) to the [enzyme-TPPS] complex of 1.3 10(4)mol/L and a first-order rate constant of 6.6 10(-1)s(-1) for dissociation of the tertiary complex, a similar pattern was found for zinc/iron transmetallation in myoglobin.

Effect of water hyacinth (Eichhornia crassipes) hay inclusion in the diets of sheep.

The aim of this study was to evaluate the effects of replacing Tifton-85 hay (0, 20, 40, 60 and 80 % on a dry matter basis) with water hyacinth hay (Eichhornia crassipes) on intake and digestibility of nutrients, feeding behaviour, rumen and blood parameters of sheep. Five uncastrated male sheep, cannulated in the rumen, with an average body weight of 40 kg were assigned in a 5 × 5 Latin square design. The water hyacinth hay contained 870 g/kg dry matter (DM), 159 g/kg crude protein (CP), 547 g/kg neutral detergent fibre (NDF) and 461 g/kg total digestible nutrients (TDN). The DM intake and digestibility of NDF and non-fibre carbohydrates (NFC) were linearly reduced by replacing the Tifton-85 hay with water hyacinth hay. Similarly, there was a linear reduction of rumination time and efficiencies of feeding and rumination of DM and NDF. The concentrations of urea, glucose, AST and GGT in blood plasma were not changed by replacing the Tifton-85 hay with water hyacinth hay. Although water hyacinth hay reduced the intake and digestibility of some nutrients, the Tifton-85 hay replacement could be economically advantageous for sheep feeding.

Glucagonoma syndrome associated with necrolytic migratory erythema.

Introduction: glucagonoma is a pancreatic neuroendocrine tumor derived from alpha-cells of the islets of Langerhans. It is marked by tumoral autonomous production of glucagon and characterized, among other symptoms, by necrolytic migratory erythema, an erythematous circinate lesion with areas of necrosis and sloughing. This is a rare disease with worldwide incidence estimated at 1 case per 20 million people.

Case Report: we report a case of glucagonoma associated necrolytic migratory erythema in a male patient, 56 years, with signs of skin lesions mainly on his legs and groin, hyperglycemia and weight loss. Biopsies of the skin lesions were performed and imaging of the abdomen showed a mass of 10 x 9 cm, at the pancreatic region. The patient was subjected to body-caudal pancreatectomy and splenectomy with autotransplant of the spleen in the greater omentum. The histopathologic report indicated a tumor in the pancreatic alpha cells. Immunohistochemistry showed expression of glucagon and chromogranin A in most tumor cells, consistent with the diagnosis of glucagonoma. The patient presented 3 years of outpatient follow-up with no complications.

Conclusion: the necrolytic migratory erythema is important for the clinical recognition of glucagonoma, and its early diagnosis is essential for a successful curative therapy.

Liquid residue of cassava as a replacement for corn in the diets of sheep.

The effect of the replacement of corn with the liquid residue of cassava was evaluated at levels of 0, 25, 50, 75, and 100% in the diets of sheep on the intake and nutrient digestibility, weight gain, and carcass characteristics. Forty non-castrated crossbred Santa Ines sheep at 4 months of age and an initial body weight of 19.5 ± 2.5 kg were slaughtered after 90 days at the feedlot. A quadratic effect was found for intake of dry matter (DM), organic matter (OM), non-fiber carbohydrates (NFC), and digestible OM, with estimated values of 1130, 1080, 558, and 777 g/day at 11.2, 11.9, 30.8, and 2.18% of replacing corn with liquid residue of cassava, respectively. However, the intake of crude protein (CP) and neutral detergent fiber (NDF) decreased linearly around 105 to 45.3 g CP/day and 413 to 284 g NDF/day. The same effect was found for digestibility of DM (715 to 649 g/kg), OM (725 to 669 g/kg), CP (801 to 759 g/kg), and NDF (486 to 379 g/kg), which decreased linearly as the dietary liquid residue of cassava increased. The maximum average daily gain (174 g/day) was estimated with a 22.4% replacement level. Liquid residue of cassava can replace up to 25% of the corn in the diets of sheep, since it was possible to obtain a gain above 100 g/day, which is considered to be satisfactory.

Isomerization of Cholecalciferol through Energy Transfer as a Protective Mechanism Against Flavin-Sensitized Photooxidation.

Cholecalciferol, vitamin D3, was found to isomerize to 5,6-trans-vitamin-D3 with a quantum yield of 0.15 ± 0.01 in air-saturated 7/3 tert-butyl alcohol/water (v/v) at 25 °C, increasing to 0.32 ± 0.02 in the absence of oxygen, through quenching of triplet excited state flavin mononucleotide, FMN, rather than becoming oxidized. The quenching was found by laser flash photolysis to have a rate constant of 1.4 × 10(8) L mol(-1) s(-1) in 7/3 tert-butyl alcohol/water (v/v) at 25 °C, assigned to energy transfer from (3)FMN* to form a reactive vit.D3 diradical. vit.D3 forms a 1/1 precomplex with FMN by hydrophobic stacking with ΔH° = -36 ± 7 kJ mol(-1) and ΔS° = -4 ± 3 J mol(-1) K(-1), as shown by single photon counting fluorescence spectroscopy and steady-state fluorescence spectroscopy. Both ground-state precomplex formation and excited-state energy transfer seem important for vit.D3 protection against flavin-sensitized photooxidation of nutrients in food and biological systems.

Sexual satisfaction and distress in sexual functioning in a sample of the BDSM community: a comparison study between BDSM and non-BDSM contexts.

Introduction: Little attention has been paid to distress in sexual functioning or the sexual satisfaction of people who practice BDSM (Bondage and Discipline, Domination and Submission, Sadism and Masochism).

Aim: The purpose of this study was to describe sociodemographic characteristics and BDSM practices and compare BDSM practitioners' sexual outcomes (in BDSM and non-BDSM contexts).

Methods: A convenience sample of 68 respondents completed an online survey that used a participatory research framework. Cronbach's alpha and average inter-item correlations assessed scale reliability, and the Wilcoxon paired samples test compared the total scores between BDSM and non-BDSM contexts separately for men and women. Open-ended questions about BDSM sexual practices were coded using a preexisting thematic tree.

Main Outcome Measures: We used self-reported demographic factors, including age at the onset of BDSM interest, age at first BDSM experience, and favorite and most frequent BDSM practices. The Global Measure of Sexual Satisfaction measured the amount of sexual distress, including low desire, arousal, maintaining arousal, premature orgasm, and anorgasmia.

Results: The participants had an average age of 33.15 years old and were highly educated and waited 6 years after becoming interested in BDSM to act on their interests. The practices in which the participants most frequently engaged did not coincide with the practices in which they were most interested and were overwhelmingly conducted at home. Comparisons between genders in terms of distress in sexual functioning in BDSM and non-BDSM contexts demonstrate that, with the exception of maintaining arousal, we found distress in sexual functioning to be statistically the same in BDSM and non-BDSM contexts for women. For men, we found that distress in sexual functioning, with the exception of premature orgasm and anorgasmia, was statistically significantly lower in the BDSM context. There were no differences in sexual satisfaction between BDSM and non-BDSM contexts for men or women.

Conclusion: Our findings suggest that BDSM sexual activity should be addressed in clinical settings that account for BDSM identities, practices, relationships, preferences, sexual satisfaction, and distress in sexual function for men and women. Additional research needs are identified, such as the need to define distressful sexual functioning experiences and expand our understanding of the development of BDSM sexual identities.

Muscular pre-conditioning using light-emitting diode therapy (LEDT) for high-intensity exercise: a randomized double-blind placebo-controlled trial with a single elite runner.

Recently, low-level laser (light) therapy (LLLT) has been used to improve muscle performance. This study aimed to evaluate the effectiveness of near-infrared light-emitting diode therapy (LEDT) and its mechanisms of action to improve muscle performance in an elite athlete. The kinetics of oxygen uptake (VO2), blood and urine markers of muscle damage (creatine kinase--CK and alanine), and fatigue (lactate) were analyzed. Additionally, some metabolic parameters were assessed in urine using proton nuclear magnetic resonance spectroscopy ((1)H NMR). A LED cluster with 50 LEDs (λ = 850 nm; 50 mW 15 s; 37.5 J) was applied on legs, arms and trunk muscles of a single runner athlete 5 min before a high-intense constant workload running exercise on treadmill. The athlete received either Placebo-1-LEDT; Placebo-2-LEDT; or Effective-LEDT in a randomized double-blind placebo-controlled trial with washout period of 7 d between each test. LEDT improved the speed of the muscular VO2 adaptation (∼-9 s), decreased O2 deficit (∼-10 L), increased the VO2 from the slow component phase (∼+348 ml min(-1)), and increased the time limit of exercise (∼+589 s). LEDT decreased blood and urine markers of muscle damage and fatigue (CK, alanine and lactate levels). The results suggest that a muscular pre-conditioning regimen using LEDT before intense exercises could modulate metabolic and renal function to achieve better performance.

Kinetics and thermodynamics of 1-hydroxyethyl radical reaction with unsaturated lipids and prenylflavonoids.

Hydroxyalkyl radicals have been reported to induce lipid oxidation as the key aspect of the pathogenesis of alcoholic fatty liver disease and are responsible for the alkylation and cleavage of DNA during the metabolism of a wide range of genotoxic compounds. However, relevant kinetic data for the oxidation of unsaturated lipids by 1-hydroxyethyl radical (HER) has not been reported. In this study, the rate constants for the reaction of unsaturated fatty acid methyl esters and sterols with HER have been determined using a competitive kinetic approach employing the spin-trap α-(4-pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN) as the competitive substrate. Polyunsaturated fatty acid methyl ester is shown to react with HER with an apparent second-order rate constant ranging from (3.7 ± 0.1) × 10(6) L mol(-1) s(-1) for methyl linoleate to (2.7 ± 0.2) × 10(7) L mol(-1) s(-1) for methyl docosahexanoate at 25.0 ± 0.2 °C in ethanol. The apparent second-order rate constant for polyunsaturated fatty acid methyl ester oxidation by HER is dependent on the number of bisallylic hydrogen atoms rather than on the bond dissociation energy value for the weakest C-H bond as determined by ab initio density functional theory calculations. Sterols displayed higher reactivity compared to unsaturated fatty acid methyl esters with apparent second-order rate constants of (2.7 ± 0.1) × 10(6) and (5.2 ± 0.1) × 10(7) L mol(-1) s(-1) at 25.0 ± 0.2 °C in ethanol for cholesterol and ergosterol, respectively. Similar experiments with prenylflavonoids as potential herbal chemopreventive agents for preventing alcoholic liver diseases yield apparent second-order rate constants close to the diffusion control with kapp values of (1.5 ± 0.2) and (3.6 ± 0.1) × 10(9) L mol(-1)s(-1) for 6-prenylnarigerin and xanthohumol at 25.0 ± 0.2 °C in ethanol solution, respectively. Polyunsaturated lipids were revealed to be highly reactive oxidizable substrates toward HER-induced oxidation in biological systems leading to damage of membranes and sensitive structures.

A novel monoclonal antibody to Neisseria meningitidis serogroup X capsular polysaccharide and its potential use in quantitation of meningococcal vaccines.

A novel murine hybridoma monoclonal antibody (MAb) was produced against the capsular polysaccharide (CP) of Neisseria meningitidis serogroup X (MenX) in order to develop a sandwich enzyme linked immunosorbent assay (ELISA) for the quantitation of the meningococcal polysaccharide. The MAb only reacted with the CP from MenX and did not react with CPs from N. meningitidis serogroups A, C, Y and W (MenA, MenC, MenY, MenW). The affinity constant (Ka) of the MAb measured by non-competitive ELISA was 7.25 × 10(7) M(-1). The application of this MAb in a sandwich ELISA was demonstrated by its ability to properly quantitate three lots of an experimental meningococcal CP-based vaccine. The MAb obtained in this work could be a valuable reagent for the detection and quantitation of future meningococcal vaccines containing MenX CP.

Subcutaneous abscess secondary to xanthogranulomatous cholecystitis.

One of the known complications of gallstones consists in gallbladder perforation with cholecystocutaneous abscesses or external fistulas formation. Nowadays these types of complications are uncommon, having been more frequent. The authors report the case of an 86 year-old woman, bedridden and dependent, referred to our institution by presenting a tumefaction with inflammatory signs in the right upper quadrant. The radiological investigation revealed it to be a subcutaneous abscess associated with a perforated gallbladder, whose histological analysis revealed xanthogranulomatous cholecystitis.

Development of four sandwich ELISAs for quantitation of capsular polysaccharides from Neisseria meningitidis serogroups A, C, W and Y in multivalent vaccines.

Neisseria meningitidis is a Gram negative bacterium that has been classified in 13 serogroups according to the biochemical composition of the capsular polysaccharide (CP). However, invasive infections are most frequently caused by six of these serogroups: A, B, C, W, X and Y (MenA, MenB, MenC, MenW, MenX, MenY). Individual CP quantitation in multivalent meningococcal CP-based vaccines is required for quality control testing of these products. In this regard, four sandwich enzyme-linked immunosorbent assays (ELISAs) were developed for the quantitation of CP. The quantitation and detection limits of the four ELISAs were below 1ng/mL. The assays showed good reproducibility and repeatability as calculated for each point of the standard curve (CV<15%). In addition, five multivalent meningococcal CP-based vaccines were evaluated and the proposed ELISAs showed that these vaccines were found into the accepted range (±30%) of CP content. These assays are suitable for screening multiple plain or conjugated meningococcal CP-based vaccines and could be useful for monitoring lot-to-lot consistency and stability analysis.

Bacterial outer membrane vesicles and vaccine applications.

Vaccines based on outer membrane vesicles (OMV) were developed more than 20 years ago against Neisseria meningitidis serogroup B. These nano-sized structures exhibit remarkable potential for immunomodulation of immune responses and delivery of meningococcal antigens or unrelated antigens incorporated into the vesicle structure. This paper reviews different applications in OMV Research and Development (R&D) and provides examples of OMV developed and evaluated at the Finlay Institute in Cuba. A Good Manufacturing Practice (GMP) process was developed at the Finlay Institute to produce OMV from N. meningitidis serogroup B (dOMVB) using detergent extraction. Subsequently, OMV from N. meningitidis, serogroup A (dOMVA), serogroup W (dOMVW), and serogroup X (dOMVX) were obtained using this process. More recently, the extraction process has also been applied effectively for obtaining OMV on a research scale from Vibrio cholerae (dOMVC), Bordetella pertussis (dOMVBP), Mycobacterium smegmatis (dOMVSM), and BCG (dOMVBCG). The immunogenicity of the OMV has been evaluated for specific antibody induction, and together with functional bactericidal and challenge assays in mice has shown their protective potential. dOMVB has been evaluated with non-neisserial antigens, including with a herpes virus type 2 glycoprotein, ovalbumin, and allergens. In conclusion, OMV are proving to be more versatile than first conceived and remain an important technology for development of vaccine candidates.
