Dr. Chia-Hua Kuo, Distinguished Professor at University of Taipei, is currently served as Dean for College of Kinesiology. Dr. Kuo was Dean for Research and Development at The University of Taipei on August 1, 2013. He specializes in Exercise Metabolism and Sports Nutrition. Professor Kuo received interdisciplinary Ph.D. degree across the areas of Kinesiology, Chemistry & Biochemistry, and Molecular Biology from University of Texas at Austin (1997). Along with his interdisciplinary approach in metabolic research, he published > 150 peer-reviewed articles internationally in areas of Sports Sciences, Biochemistry, Nutrition, Biology, and Physiology. He is Associate Editor for Frontiers in Physiology and Journal of Traditional and Complementary Medicine. Kuo’s current work focuses on examining the mechanisms regarding how exercise-induced entropic stress can transform human body into a more robust system in fitness and metabolic contexts. His team applies knowledge from novel findings of research work to formulate nutritional supplements and training intervention that improve biological complexity of multicellular organisms against physical challenges. His works are closely relevant in enhancement of athletic performance and improvement of life quality for aging humans. He received Fellowship of American College of Sports Medicine (2011). He has been invited to give keynote speeches or guest lectures to international conferences held in many countries including Japan, Korea, China, Singapore, Thailand, Indonesia, Russia, Romania, Kuwait, and Spain. Professor Kuo is currently President Elect for Asian Nutrition Society for Sports & Health (ANSSH).

Can mesenchymal stem cell lysate reverse aging?

Recent findings regarding uses of adipose-derived mesenchymal stem cell (MSC)-lysate on weight loss and improved glucose tolerance in mice on a high-fat diet suggest an encouraging possibility of using MSC lysate for an anti-aging intervention in humans. However, weight loss and lipopenia during late life can be as life-threatening as hyperglycemia during early adulthood. For this 3-year lifelong experiment, a total of 92 rats were randomized into the vehicle-injected group (F=22; M=24) and the MSC lysate injected group (F=22, M=24). We examined longevity, spontaneous locomotor activity, and body composition in rats maintained on a normal diet and received an intermittent treatment of human adipose-derived MSC lysate (3 times a week, 11 times a month given every second month), starting at 12 months of age until natural death. In substantiating previous knowledge regarding the effects of long-term MSC lysate treatments on fat loss and insulin resistance, the present findings also highlighted a shortened average lifespan, a longer inactive time, and a greater bone loss with a relative increase of lean mass in MSC lysate rats with respect to controls. Conclusion: Our data suggest that MSC lysate treatments stimulate disparity in tissue development and produce a cachexia-like effect to decrease longevity.

Doxorubicin inhibits muscle inflammation after eccentric exercise.

Background: Doxorubicin, a widely used anti-tumour drug, is known to cause muscle loss in cancer patients.

Methods: Following an acute dose of doxorubicin injection (2.5?mg/kg per body weight), we examined macrophage distribution in rat soleus muscle challenged by eccentric exercise (downhill running). Long-term doxorubicin treatment (one injection every 3?days) on muscle mass and survival were also determined.

Results: Under non-exercised condition, increased tumour necrosis factor (TNF)-alpha mRNA and decreased IL-10 mRNA were observed in soleus muscle of doxorubicin-treated rats, compared with saline-treated control rats. However, increases in inflammation score (leukocyte infiltration), nitrotyrosine level, and M1 macrophage (CD68 ) invasion in exercised soleus muscle were absent in doxorubicin-treated rats, whereas increased M2 macrophage (CD163 ) localization in exercised muscle was less affected by doxorubicin. Despites coenzyme Q (Q10) supplementation significantly elevated TNF-alpha mRNA, nitrotyrosine, and anti-oxidant gamma-glutamylcysteine synthetase (GCS) levels in non-exercised soleus muscle, these pro-inflammatory responses were also abolished in doxorubicin-treated rats. Results from long-term doxorubicin treatment show a significant muscle loss followed by an accelerated death, which cannot be reversed by Q10 supplementation.

Conclusions: (i) Doxorubicin impairs inflammation mechanism by depleting M1 macrophage in exercised skeletal muscle; (ii) Muscle loss and accelerated death during prolonged doxorubicin treatment cannot be reversed by Q10 supplementation.
