Introduction

Our laboratory focus on developing and evaluating the effect of stem cells (especially human adult stem cells) transplantation on the outcome of various brain injuries. We had finished a Phase I/II trial evaluating intracranial transplantation of autologous CD34+ bone marrow stem cells in patients suffering from a chronic ischemic cerebral stroke. Six transplantations were performed, and the result had so far proven the safety and feasibility of intracranial delivery of stem cells, prepared in our GTP-compatible facility.

Besides translational stem cell research, we are also interested in studying the protection mechanisms of growth factors (e.g., G-CSF), neuropeptides (e.g., urocortin), and various herbal extracts on cellular damage caused by oxidative stress, neuro-degeneration diseases, and neurotrauma. Among these candidates, we have confirmed that urocortin (UCN, a 40-amino-acids anorexigenic neuropeptide) can promote the survival of dopaminergic neurons by inhibiting the activity of HDAC in vitro, decrease the inflammatory reaction in LPS-induced microglial activation in vitro, promote the differentiation of neural stem cells both in vitro and in vivo in rats, and (4) protect rats’ brain from intra-cerebral hemorrhagic (ICH) stroke. Most importantly, UCN can still exert its therapeutic effect, even when it is administered 6 hours after the experimental ICH in rats. Our UCN research has so far produced a US patent (US 8802630), and the other patent regarding how UCN can promote neural regeneration and proliferation is currently under review.

As above-mentioned, we have shown that UCN might possess therapeutic potential in ameliorating ICH, as well as a modulator in helping the brain recovery after the injury. ICH is so far still a serious clinical problem lacking effective treatment. We are seeking collaborators who can help us to (1) develop UCN into a neuroprotective agent, and (2) delineate the exact mechanism of UCN in ameliorating ICH, as well as in modulating brain recovery/ neural regeneration.
