Extraction of bioactive compounds from Psidium guajava leaves and its utilization in preparation of jellies.

Psidium guajava L. (guava) is predominantly grown throughout the world and known for its medicinal properties in treating various diseases and disorders. The present work focuses on aqueous extraction of bioactive compounds from the guava leaf and its utilization in the formulation of jelly to improve the public health. The guava leaf extract has been used in the preparation of jelly with pectin (1.5 g), sugar (28 g) and lemon juice (2 mL). The prepared guava leaf extract jelly (GJ) and the control jelly (CJ, without extract) were subjected to proximate, nutritional and textural analyses besides determination of antioxidant and antimicrobial activities. GJ was found to contain carbohydrate (45.78 g/100 g), protein (3.0 g/100 g), vitamin C (6.15 mg/100 g), vitamin B3 (2.90 mg/100 g) and energy (120.6 kcal). Further, the texture analysis of CJ and GJ indicated that both the jellies showed similar properties emphasizing that the addition of guava leaf extract does not bring any change in the texture properties of jelly. GJ exhibited antimicrobial activity against various bacteria ranging from 11.4 to 13.6 mm. Similarly, GJ showed antioxidant activity of 42.38% against DPPH radical and 33.45% against hydroxyl radical. Mass spectroscopic analysis of aqueous extract confirmed the presence of esculin, quercetin, gallocatechin, 3-sinapoylquinic acid, gallic acid, citric acid and ellagic acid which are responsible for antioxidant and antimicrobial properties.

Nanotechnology: an emerging approach to combat COVID-19.

The recent outbreak of coronavirus disease (COVID-19) has challenged the survival of human existence in the last 1 year. Frontline healthcare professionals were struggling in combating the pandemic situation and were continuously supported with literature, skill set, research activities, and technologies developed by various scientists/researchers all over the world. To handle the continuously mutating severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) requires amalgamation of conventional technology with emerging approaches. Nanotechnology is science, engineering, and technology dealing at the nanoscale level. It has made possible the development of nanomaterials, nano-biosensors, nanodrugs, and vaccines for diagnosis, therapy, and prevention of COVID-19. This review has elaborately highlighted the role of nanotechnology in developing various detection kits such as nanoparticle-assisted diagnostics, antibody assay, lateral flow immunoassay, nanomaterial biosensors, etc., in detection of SARS-CoV-2. Similarly, various advancements supervene through nanoparticle-based therapeutic drugs for inhibiting viral infection by blocking virus attachment/cell entry, multiplication/replication, and direct inactivation of the virus. Furthermore, information on vaccine development and the role of nanocarriers/nanoparticles were highlighted with a brief outlining of nanomaterial usage in sterilization and preventive mechanisms engineered to combat COVID-19 pandemic.

Immunotherapeutics for Covid-19 and post vaccination surveillance.

Severe acute respiratory syndrome coronavirus (SARS-CoV-2) has emerged as a pandemic and named as novel coronavirus disease (nCOVID-19). SARS-CoV-2 is different from other known viruses due to multiple mutations on the sites of nonstructural proteins (NSP) 2 and 3, and the varying nature of virulence between different persons. Immunotherapies such as vaccines and monoclonal antibodies have a protective effect on the patients bringing them to the front of the line of potential treatments. The present review intends to cover the development of 20 different vaccine candidates categorized under live attenuated vaccines, inactivated vaccines, subunit vaccines, viral vector-based vaccines, and nucleic acid vaccines. Formulation of these vaccine candidates by various companies in collaboration with global organizations and their status of clinical trials were addressed. On the other hand, various approaches for post-vaccination surveillance using nucleic acid and protein biomarkers imbued on suitable platforms were also highlighted to sum up the immune therapeutics for Covid-19.

Guava is considered as poor man's apple rich in phytochemicals with medicinal value and hence it is highly consumed. Gas chromatography-mass spectroscopy (GC-MS) analysis of guava leaf extract revealed the presence of various bioactive compounds with antimicrobial, antioxidant, anticancer, and antitumor properties. Hence, it is used in tooth paste formulations along with other ingredients such as Acacia arabica gum powder, stevia herb powder, sea salt, extra virgin coconut oil, peppermint oil in the present study. Three formulations F1, F2 and F3 have been made by varying the concentration of these ingredients and the prepared formulations were studied for their antimicrobial activity and physico-chemical parameters such as pH, abrasiveness, foaming activity, spreading and cleaning ability. Among these, F3 showed significant antioxidant and antimicrobial properties, minimal cytotoxicity, maximum spreadability and very high cleaning ability. This study surmises that the herbal toothpaste formulation is greener, rich in medicinal values and imparts oral hygiene.

Laccase mediated delignification of pineapple leaf waste: an ecofriendly sustainable attempt towards valorization.

Background: Escalating energy security, burgeoning population and rising costs of fossil fuels have focussed our attention on tapping renewable energy sources. As the utilization of food crops for biofuel production culminates into food vs. fuel dilemma, there is an intensive need for alternatives. Production of biofuels from lignocellulosic biomass owing to its profuse availability and high holocellulose content is a promising area for research.

Results: In the present study, pineapple leaf, an agro-industrial waste was pretreated with laccase to enhance the enzymatic digestibility of the substrate for improved production of reducing sugar. Variables determining enzymatic delignification of pineapple leaf waste have been optimized by response surface methodology based on central composite design. Maximum delignification of 78.57%(w/w) resulted in reducing sugar of 492.33 ± 3.1 mg/g in 5.30 h. The structural changes in pineapple leaf waste, after laccase treatment, were studied through Fourier transformed infrared spectroscopy, X-ray diffraction and Scanning electron microscopy. Specific surface area, pore volume, and pore diameter of the substrate were studied using the Brunauer-Emmett-Teller and Barrett-Joyner-Halenda methods and found a significant increase in the aforementioned parameters after delignification.

Conclusion: Laccase mediated delignification of pineapple leaf waste is a cleaner sustainable process for enhanced production of reducing sugar which can accomplish the demand for biofuels.

Integrated bioethanol and biomanure production from potato waste.

Disposal of potato processing waste and the problem of pollution associated with it is a vital issue that is being faced by the potato processing plants. The conventional peeling methods presently followed in the processing plants for removing the potato peel, also result in the loss of some portion of the mash which is rich in starch. Indiscriminate discharge of the waste causes detrimental effects in the environment, so this problem can be resolved by successful utilization of the waste for the generation of value added products. Hence, the present work focuses on integrated production of bioethanol and biomanure to utilize the waste completely leading to zero waste generation. The first part of the work describes a comparative study of ethanol production from potato peel and mash wastes by employing co-culture of Aspergillus niger and Saccharomyces cerevisiae at various incubation time (24-120 h) instead of application of enzymes. The solid state fermentation of potato peel and mash inoculated with co-culture, resulted in bioethanol production of 6.18% (v/v) and 9.30% (v/v) respectively. In the second part of the work, the residue obtained after ethanol production was inoculated with seven different microorganisms (Nostoc muscorum, Fischerella muscicola, Anabaena variabilis, Aulosira fertilissima, Cylindrospermum muscicola, Azospirillium lipoferum, Azotobacter chroococcum) and mixture of all the organisms in equal ratio for nitrogen (N), phosphorous (P) and potassium (K) enrichment. Among them, A. variabilis was found to enrich N, P and K content of the residue by nearly 7.66, 21.66 and 15 fold than that of the initial content, ultimately leading to improved N:P:K ratio of approximately 2:1:1. The application of simultaneous saccharification and fermentation (SSF) for the conversion of potato waste to ethanol and enrichment of residue obtained after ethanol production with microorganisms to be used as manure envisages environmental sustainability.
